Accumulation of cholesterol in human blood can cause several health problems such as heart disease, coronary artery disease, arteriosclerosis, hypertension, cerebral thrombosis, etc. Therefore, simple and fast cholesterol determination in blood is clinically important. In this study, two types of amperometric cholesterol biosensors were designed by physically entrapping cholesterol oxidase in conducting polymers; thiophene capped poly(ethyleneoxide)/polypyrrole (PEO-co-PPy) and 3-methylthienyl methacrylate-co-p-vinyl benzyloxy poly(ethyleneoxide)/polypyrrole (CP-co-PPy). PEO-co-PPy and CP-co-PPy were synthesized electrochemically and cholesterol oxidase was immobilized by entrapment during electropolymerization. The amperometric responses of the enzyme electrodes were measured by monitoring oxidation current of H2O2 at +0.7 V in the absence of a mediator. Kinetic parameters, such as Km and Imax, operational and storage stabilities, effects of pH and temperature were determined for both entrapment supports. Km values were found as 1.47 and 5.16 mM for PEO-co-PPy and CP-co-PPy enzyme electrodes, respectively. By using these Km values, it can be observed that ChOx immobilized in PEO-co-PPy shows higher affinity towards the substrate.  相似文献

This article reports the distribution of trace metals and major ions in lake waters of Sultansazl L < L , a reedfield near Kayseri, Turkey. The determination of elements such as Cr, Ni, Cu, Cd and Pb in lake water samples was performed by AAS after a preconcentration step using a column packed with Amberlite XAD-16 resin. Both flame- and graphite furnace-atomic absorption spectrometry was used to determine these metals in lake water samples. The concentrations of the metals bound to humic substances, and free metal ions were determined after their sorption-elution on the resin, separately. The column method optimized with sodium tetraborate reagent was used in determining the free metal ions. In the determinations of Ca2+ and Mg2+, K+ , and SO2-4 , Cl m and total hardness, flame atomic absorption spectrometry, flame photometry, and titrimetry were used, respectively. In order to evaluate the analytical data by multivariate statistical techniques which enable feature reduction and grouping of the pollutant sources in lake waters from their chemical composition, principal component analysis (PCA), cluster analysis (CA) and correlation analysis were used. As a consequence of multivariate statistical evaluation, main anthropogenic sources like traffic, industry and agricultural processes were drawn to be responsible from the pollution in the environment investigated.  相似文献

A new dinuclear copper (II) complex of 2,5–furandicarboxyclic acid with 4(5)‐methylimidazole, [Cu (FDCA)((4(5)MeI)2]2·2H2O, was synthesized, and its structure characterized by XRD, FT–IR and UV–Vis spectroscopic techniques. The α‐glucosidase inhibition and cytotoxicity study of the synthesized Cu (II) complex were determined by IC50 values. The optimized geometry and vibrational harmonic frequencies for the Cu (II) complex were obtained by using Density Functional Theory (DFT) of HSEh1PBE/6–311++G(d,p)/LanL2DZ level. TD‐DFT/HSEh1PBE/6–311++G(d,p)/LanL2DZ level with CPCM model was applied to examine the electronic spectral properties and major contributions were determined via Swizard program. To investigate linear and nonlinear optical behavior of the synthesized Cu (II) complex, the α, Δα and χ(1)/β, γ and χ(3) parameters called linear/nonlinear optical parameters in gas phase and ethanol solvent were computed at the same level and basis set. Furthermore, molecular electrostatic potential (MEP) surface was determined by using the same level. The docking study of the Cu (II) complex to the binding site of the target protein (the template structure S. cerevisiae isomaltase) is fulfilled. Natural bond orbital (NBO) analysis was used to investigate the hyperconjugative interactions, inter‐ and intra‐molecular bonding and to determine coordination around Cu (II) ion. Finally, present work is the first remarkable scientific report of mixed‐ligand (H2FDCA and 4(5)MeI) Cu (II) complex as novel drug candidate for DM II. It is also determined that microscopic third?NLO parameters for the Cu (II) complex is remarkable.  相似文献

Chitosan membrane with glutathione reductase and sulfhydryl oxidase (SOX) was subsequently integrated onto the surface of spectrographic graphite rods for obtaining a glutathione biosensor. The working principle was based on the monitoring of O2 consumption that correlates the concentration of glutathione during the enzymatic reaction. A linear relationship between sensor response and concentration was obtained between 0.5 and 2.0 mM for oxidized glutathione (GSSG), and 0.2–1.0 mM for reduced glutathione (GSH) in the presence of 2 μM nicotinamide adenine dinucleotide phosphate (NADPH) under the optimum working conditions. Also, reduced/oxidized glutathione were separated by HPLC and utility of bienzymatic system was investigated as an electrochemical detector for the analysis of these compounds. All data were given as a comparison of two systems: biosensor and diode array detector (DAD).  相似文献

A mild and practically convenient one-pot procedure for the Mannich reaction via condensation of amines, aldehydes and malonates, β-ketoesters, or β-dicarbonyl compounds has been carried out without using any organic solvent, metallic catalyst, or Lewis acids or bases at room temperature. Water was a suitable medium for Paal–Knorr pyrrole cyclocondensation. Hexa-2,5-dione was reacted with several aliphatic and aromatic primary amines, affording N-substituted 2,5-dimethyl pyrrole derivatives in good to excellent yields. An on-line preconcentration procedure for the determination of bismuth by flame atomic absorption spectrometry (FAAS) has been described. Lewatit TP-207 chelating resin, including iminodiacetate group, packed in a minicolumn was used as adsorbent material. Bi(III) was sorbed on the chelating resin, from which it could be eluted with 3 mol L−1 HNO3 and then introduced directly to the nebulizer-burner system of FAAS. Best preconcentration conditions were established by testing different resin quantities, acidity of sample, types of eluent, sample and eluent solution volumes, adsorption and elution flow rates, and effect of interfering ions. The detection limit of the method was 2.75 μg L−1 while the relative standard deviation was 3.0% for 0.4 μg mL−1 Bi(III) concentration. The developed method has been applied successfully to the determination of bismuth in pharmaceutical cream, standard reference materials and various natural water samples with satisfactory results.  相似文献

Poly(acrylamidoxime-co-2-acrylamido-2-methylpropane sulfonic acid) (PAMSA) hydrogel was prepared by copolymerization of acrylonitrile and 2-acrylamido-2-methylpropane sulfonic acid as monomer, N,N′-methylenebis(acrylamide) as crosslinking agent and potassium peroxodisulfate as initiator. Amidoximated copolymer network was prepared by the reaction of copolymer network with hydroxylamine hydrochloride. A batch procedure was used for the determination of the characteristics of the U(VI) solid phase extraction from the amidoximated hydrogel. The determination of U(VI) was performed by spectrophotometric method using arsenazo-III as complexing agent. Optimal pH value for the quantitative preconcentration was 3, and full desorption was achieved with 3 mol L−1 HClO4. The adsorption process can be well described by the pseudo-second-order kinetic model, and the equilibrium adsorption isotherm was closely fitted with the Langmuir model. A preconcentration factor of 20 and the three sigma detection limit of 2.8 μg L−1 (n = 20) were achieved for uranium(VI) ions. The PAMSA hydrogel was used for separating and preconcentrating the uranyl ion existing in sea water samples, thermal spring water samples and the certified reference materials (TMDA 64; fortified lake water sample).  相似文献

A simple speciation, separation and enrichment method has been developed for the determination of Cr(III) and Cr(VI) ions in different samples by ion-pair solvent extraction with a β-diketone ligand, 2-(4-methoxybenzoyl)-N′-benzylidene-3-(4-methoxyphenyl)-3-oxo-N-phenyl-propono hydrazide (MBMP). Cr(III) was separated from Cr(VI) as Cr(III)-(MBMP)-perchlorate ternary ion-pair complex. The influences of various analytical parameters including pH, amount of reagent, shaking time, sample volume and ionic strength on the recovery of Cr(III) and/or Cr(VI) were investigated. Total chromium was obtained after reducing Cr(VI) to Cr(III) with NH2OH?·?HCl. Recoveries were found to be higher than 95% and the relative standard deviation (RSD) was less than 2%. The method detection limit based on 3σ criterion for Cr(III) was found to be 0.32?µg?L?1. The formed ternary ion-pair complex, Cr(III)?:?MBMP?:?2ClO4, has a molar ratio of 1?:?1?:?2. The developed method has been applied successfully to the speciation of chromium in various natural water, soil, sediment and hair samples with satisfactory results.  相似文献

Iron corroles modified with a xanthene scaffold are delivered from easily available starting materials in abbreviated reaction times. These new iron corroles have been spectroscopically examined with particular emphasis on defining the oxidation state of the metal center. Investigation of their electronic structure using (57)Fe Mo?ssbauer spectroscopy in conjunction with density functional theory (DFT) calculations reveals the non-innocence of the corrole ligand. Although these iron corroles contain a formal Fe(IV) center, the deprotonated corrole macrocycle ligand is one electron oxidized. The electronic ground state of these complexes is best described as an intermediate spin S = 3/2 Fe(III) site strongly antiferromagnetically coupled to the S = 1/2 of the monoradical dianion corrole [Fe(III)Cl-corrole(+?)]. We show here that iron corroles as well as xanthene-modified and hangman xanthene iron corroles are redox active and catalyze the disproportionation of hydrogen peroxide via the catalase reaction, and that this activity scales with the oxidation potential. The meso position of corrole macrocycle is susceptible toward nucleophilic attack during catalase turnover. The reactivity of peroxide within the hangman cleft reported here adds to the emerging theme that corroles are good at catalyzing two-electron activation of the oxygen-oxygen bond in a variety of substrates.  相似文献
