Background  

Oral administration of BCG was the route initially used by Calmette and Guérin, but was replaced by intradermal administration in virtually all countries after the Lubeck accident. However, Brazil continued to administer oral BCG Moreau RDJ, which was maintained until the mid-1970s when it was substituted by the intradermal route. Although BCG vaccination has been used in humans since 1921, little is known of the induced immune response. The aim of this study was to analyse immunological responses after oral vaccination with M. bovis BCG Moreau RDJ.  相似文献

A rapid label-free visual assay for the detection of viral RNA using peptide nucleic acid (PNA) probes and gold nanoparticles (AuNPs) is presented in this study. Diagnosis is a crucial step for the molecular surveillance of diseases, and a rapid visual test with high specificity could play a vital role in the management of viral diseases. In this assay, the specific agglomerative behavior of PNA with gold nanoparticles was manipulated by its complementation with viral RNA. The assay was able to detect 5–10 ng of viral RNA from various biological samples, such as allantoic fluids, cell culture fluids and vaccines, in 100 μl of test solution. The developed assay was more sensitive than a hemagglutination (HA) test, a routine platform test for the detection of Newcastle disease virus (NDV), and the developed assay was able to visually detect NDV with as little as 0.25 HA units of virus. In terms of the specificity, the test could discriminate single nucleotide differences in the target RNA and hence could provide visual viral genotyping/pathotyping. This observation was confirmed by pathotyping different known isolates of NDV. Further, the PNA-induced colorimetric changes in the presence of the target RNA at different RNA to PNA ratios yielded a standard curve with a linear coefficient of R2 = 0.990, which was comparable to the value of R2 = 0.995 from real-time PCR experiments with the same viral RNA. Therefore, the viral RNA in a given samples could be quantified using a simple visual spectrophotometer available in any clinical laboratory. This assay may find application in diagnostic assays for other RNA viruses, which are well known to undergo mutations, thus presenting challenges for their molecular surveillance, genotyping and quantification.  相似文献

We have investigated the superconducting properties of the Bi1.7 Pb0.3Sr2Ca2−xCe x Cu3O10+δ system with x=0.00, 0.02, 0.04, 0.08 and 0.1 by X-ray diffraction and magnetic susceptibility. The substitution of Ce for Ca has been found to drastically change the superconducting properties of the system. X-ray diffraction studies on these compounds indicate decrease in the c-parameter with increased substitution of Ce at Ca site and volume fraction of high T c (2 : 2 : 2 : 3) phase decreases and low T c phase increases. The magnetic susceptibility of this compound shows that the diamagnetic on set superconducting transition temperature (onset) varies from 109 K to 51 K for x=0.00, 0.02, 0.04, 0.08 and 0.1. These results suggest the possible existence of Ce in a tetravalent state rather than a trivalent state in this system; that is, Ca2+ → Ce4+ replacement changes the hole carrier concentration. Hole filling is the cause of lowering T c of the system.  相似文献

Samples of explosive class materials were analyzed by 14 MeV neutron activation technique to estimate concentrations of oxygen, nitrogen, aluminium and chlorine. For aluminized explosives, test of stability against temperature was carried out by heating samples at 75 °C and loss of nitrogen was studied by measuring the ratio of aluminium to nitrogen in the sample. Loss of oxygen and nitrogen in a triple base propellant heated at 71.6 °C, was also estimated by measuring residual oxygen and nitrogen in the heated samples. This technique is convenient to adopt in the laboratory and useful to decide quickly the suitability of explosive class materials for practical applications.  相似文献

A concise and experimentally straightforward method for assembling multiple benzo(crown ether) units around 1,3,5-triaroylbenzene scaffolds has been developed. Symmetrical tris(crown ether)s possessing three benzo(15-crown-5) or three benzo(18-crown-6) peripheral substituents have been prepared in good yield via cyclotrimerization of monomeric enaminones. Efficient cross-cyclotrimerizations have also been demonstrated through construction of unsymmetrical triaroylbenzenes functionalized with only one or two benzo(15-crown-5) moieties. The alkali cation-binding abilities of these mono- and polytopic crown ethers have been probed through picrate extraction experiments and isothermal titration calorimetry. Thermodynamic binding parameters uncovered using the latter technique reveal increasing K+/Na+ selectivity in the benzo(15-crown-5) series of compounds as a function of increasing numbers of benzo(crown) units. The data also indicate that the triaroylbenzene-derived bis- and tris-crown ethers do not engage in intramolecular chelation of cations too large to be accommodated by individual crown macrorings. Instead, cation/triaroylbenzene stoichiometries and binding profiles indicate formation of alkali metal-bridged dimers.  相似文献

An efficient NaNO2/I2‐mediated one‐pot transformation of Morita–Baylis–Hillman (MBH) acetates into alkyl 3‐nitro‐5‐(aryl/alkyl)isoxazole‐4‐carboxylates is described. In a cascade event, initial Michael addition of NaNO2 to the MBH acetate furnishes the allylnitro intermediate which undergoes I2‐catalyzed oxidative α‐C H nitration of the nitromethyl subunit followed by [3+2] cycloaddition to afford the title compounds. Structural elaborations of these highly substituted isoxazoles by SNAr reactions and hydrogenolysis allows access to useful products.  相似文献

An isomeric series of homologous crownophanes (i.e., macrocycles possessing structural features of crown ethers and cyclophanes) has been prepared via a concise and modular synthetic route. Macrocyclization is achieved in reasonable yield during the course of an enaminone-triggered benzannulation with bis(aryl ethynyl ketone) reaction partners. The crownophanes examined were active alkali cation binding agents in the gas phase, but failed to exhibit ionophoric properties in solution. On the basis of X-ray crystallographic analysis, it is concluded that the cyclophane framework of these macrocycles is too large and rigid to allow efficient interaction with the cations examined.  相似文献
