The role of N1-substitution in controlling the deactivation processes in photoexcited cytosine derivatives has been explored using picosecond time-resolved IR spectroscopy. The simplest N1-substituted derivative, 1-methylcytosine, exhibits relaxation dynamics similar to the cytosine nucleobase and distinct from the biologically relevant nucleotide and nucleoside analogues, which have longer-lived excited-state intermediates. It is suggested that this is the case because the sugar group either facilitates access to the long-lived (1)n(O)π* state or retards its crossover to the ground state.  相似文献

The combination of microchip electrophoresis with amperometric detection leads to a number of analytical challenges that are associated with isolating the detector from the high voltages used for the separation. While methods such as end-channel alignment and the use of decouplers have been employed, they have limitations. A less common method has been to utilize an electrically isolated potentiostat. This approach allows placement of the working electrode directly in the separation channel without using a decoupler. This paper explores the use of microchip electrophoresis and electrochemical detection with an electrically isolated potentiostat for the separation and in-channel detection of several biologically important anions. The separation employed negative polarity voltages and tetradecyltrimethylammonium bromide (as a buffer modifier) for the separation of nitrite (NO??), glutathione, ascorbic acid, and tyrosine. A half-wave potential shift of approximately negative 500 mV was observed for NO?? and H?O? standards in the in-channel configuration compared to end-channel. Higher separation efficiencies were observed for both NO?? and H?O? with the in-channel detection configuration. The limits of detection were approximately two-fold lower and the sensitivity was approximately two-fold higher for in-channel detection of nitrite when compared to end-channel. The application of this microfluidic device for the separation and detection of biomarkers related to oxidative stress is described.  相似文献

The application of enhanced fluidity liquid (EFL) mobile phases to improving isocratic chromatographic separation of nucleosides in hydrophilic interaction liquid chromatography (HILIC) mode is described. The EFL mobile phase was created by adding carbon dioxide to a methanol/buffer solution. Previous work has shown that EFL mobile phases typically increase the efficiency and the speed of the separation. Herein, an increase in resolution with the addition of carbon dioxide is also observed. This increase in resolution was achieved through increased selectivity and retention with minimal change in separation efficiency. The addition of CO2 to the mobile phase effectively decreases its polarity, thereby promoting retention in HILIC. Conventional organic solvents of similar nonpolar nature cannot be used to achieve similar results because they are not miscible with methanol and water. The separation of nucleosides with methanol/aqueous buffer/CO2 mobile phases was also compared to that using acetonitrile/buffer mobile phases. A marked decrease in the necessary separation time was noted for methanol/aqueous buffer/CO2 mobile phases compared to acetonitrile/buffer mobile phases. There was also an unusual reversal in the elution order of uridine and adenosine when CO2 was included in the mobile phase.  相似文献

Given \({\mathbb Z}\)-graded rings A and B, we ask when the graded module categories gr-A and gr-B are equivalent. Using \({\mathbb Z}\)-algebras, we relate the Morita-type results of Áhn-Márki and del Río to the twisting systems introduced by Zhang, and prove, for example: Theorem If A and B are \({\mathbb Z}\) -graded rings, then: (1) A is isomorphic to a Zhang twist of B if and only if the \({\mathbb Z}\) -algebras \(\overline{A} = \bigoplus_{i,j \in {\mathbb Z}} A_{j-i}\) and \(\overline{B} = \bigoplus_{i,j \in {\mathbb Z}} B_{j-i}\) are isomorphic. (2) If A and B are connected graded with A 1?≠?0, then gr-A???gr-?B if and only if \(\overline{A}\) and \( \overline{B}\) are isomorphic. This simplifies and extends Zhang’s results.  相似文献

Coherence masking protection (CMP) refers to the phenomenon in which a target formant is labeled at lower signal-to-noise levels when presented with a stable cosignal consisting of two other formants than when presented alone. This effect has been reported primarily for adults with first-formant (F1) targets and F2/F3 cosignals, but has also been found for children, in fact in greater magnitude. In this experiment, F2 was the target and F1/F3 was the cosignal. Results showed similar effects for each age group as had been found for F1 targets. Implications for auditory prostheses for listeners with hearing loss are discussed.  相似文献

Based on clinical trials showing the efficacy to reduce vertebral and non‐vertebral fractures, strontium ranelate (SrR) has been approved in several countries for the treatment of postmenopausal osteoporosis. Hence, it is of special clinical interest to elucidate how the Sr uptake is influenced by dietary Ca deficiency as well as by the formula of Sr administration, SrR versus strontium chloride (SrCl2). Three‐month‐old ovariectomized rats were treated for 90 days with doses of 25 mg kg?1 d?1 and 150 mg kg?1 d?1 of SrR or SrCl2 at low (0.1% Ca) or normal (1.19% Ca) Ca diet. Vertebral bone tissue was analysed by confocal synchrotron‐radiation‐induced micro X‐ray fluorescence and by backscattered electron imaging. Principal component analysis and k‐means clustering of the acquired elemental maps of Ca and Sr revealed that the newly formed bone exhibited the highest Sr fractions and that low Ca diet increased the Sr uptake by a factor of three to four. Furthermore, Sr uptake in bone of the SrCl2‐treated animals was generally lower compared with SrR. The study clearly shows that inadequate nutritional calcium intake significantly increases uptake of Sr in serum as well as in trabecular bone matrix. This indicates that nutritional calcium intake as well as serum Ca levels are important regulators of any Sr treatment.  相似文献

Cytochrome P450 enzymes are the predominant mediators of phase I metabolism of exogenous small molecules. As a result of their extensive role in metabolism of xenobiotics, drug compounds, and endogenous compounds, as well as their wide tissue distribution, significant drug discovery resources are spent to avoid interacting with this class of enzymes. Here we review historical and recent in silico modeling of 7 cytochrome P450 enzymes of particular interest, specifically CYP1A2, CYP2B6, CYP2C8, CYP2C9, CYP2C19, CYP2D6, and CYP3A4. For each we provide a brief biological background including known inhibitors, substrates, and inducers, as well as details of computational modeling efforts and advances in structural biology. We also provide similar details for 3 nuclear receptors known to regulate gene expression of these enzyme families.  相似文献
