The female flowers of hops (Humulus lupulus L.) used to flavor beer contain the prenylated flavonoids xanthohumol (XN) and isoxanthohumol (IX). IX is moderately estrogenic in vitro and XN has pharmacological properties that might make it useful as a cancer chemopreventive agent. The metabolism of these dietary flavonoids was investigated in vitro using human liver microsomes. Hydroxylation of a prenyl methyl group was the primary route of oxidative metabolism forming either cis or trans hydroxylated metabolites of IX but only the trans isomer of XN. The double bond on the prenyl group of both compounds formed an epoxide which was opened by an intramolecular reaction with the neighboring hydroxyl group. The potent phytoestrogen 8-prenylnaringenin (8-PN) was detected as a demethylation product of IX. However, the analogous demethylation reaction was not observed for XN. Since XN can be converted to IX through acid-catalyzed cyclization in the stomach, XN might contribute to the in vivo levels of estrogenic 8-PN following consumption of hops extracts.  相似文献

Isotopes of hydrogen (3H, 2H) and oxygen (18O) are perfect candidates for groundwater tracers. A survey of isotopic composition of 34 groundwater samples and one Lake from Vojvodina region (Serbia) is presented here. Tritium activity concentration and stable isotope composition (δ2H, δ18O), as well as deuterium excess, were determined. The groundwater samples lie on the groundwater regression line. Minor deviations and a few lower deuterium excess values indicate waters recharged in a different climate regime and subjected to evaporation, respectively. According to the obtained results, most of the analyzed groundwater can be characterized as modern waters, recharged mostly from precipitation.

Peptides that adopt β‐helix structures are predominantly found in transmembrane protein domains or in the lipid bilayer of vesicles. Constructing a β‐helix structure in pure water has been considered difficult without the addition of membrane mimics. Herein, we report such an example; peptide 1 self‐assembles into a supramolecular β‐helix in pure water based on charge interactions between the individual peptides. Peptide 1 further showed intriguing transitions from small particles to helical fibers in a time‐dependent process. The fibers can be switched to vesicles by changing the pH value.  相似文献

This work extends in vitro screening of antimicrobial activity of avarol, the marine natural product firstly isolated from the Mediterranean sponge Dysidea avara. Its anticandidial activity was evaluated by microdilution method against eight Candida strains, two ATCC and six clinical ones. At a different extent this compound was proven to be active against all the strains tested (MIC 0.8–6.0 μg/mL and MFC 1.6–12.0 μg/mL, respectively). According to the best of our knowledge, this is the first report on avarol activity towards any yeast strain which may be of relevance for Alzheimer’s disease. Indeed, avarol derivatives showing moderate AChE activity should be screened for anticandidial activity both in vitro and in vivo.  相似文献

We have investigated the electrochemical oxidation of a number natural phenolics (salicylic acid, m-hydroxybenzoic acid, p-hydroxybenzoic acid, protocatechuic acid, o-coumaric acid, m-coumaric acid, p-coumaric acid, caffeic acid, quercetin and rutin) using cyclic voltammetry. The antioxidant properties of these compounds were also studied. A structural analysis of the tested phenolics suggests that multiple OH substitution and conjugation are important determinants of the free radical scavenging activity and electrochemical behavior. Compounds with low oxidation potentials (Epa lower than 0.45) showed antioxidant activity, whereas compounds with high Epa values (>0.45) act as prooxidants.  相似文献

Quantitative characterization of nucleic acids is becoming a frequently used method in routine analysis of biological samples, one use being the detection of genetically modified organisms (GMOs). Measurement uncertainty is an important factor to be considered in these analyses, especially where precise thresholds are set in regulations. Intermediate precision, defined as a measure between repeatability and reproducibility, is a parameter describing the real situation in laboratories dealing with quantitative aspects of molecular biology methods. In this paper, we describe the top-down approach to calculating measurement uncertainty, using intermediate precision, in routine GMO testing of food and feed samples. We illustrate its practicability in defining compliance of results with regulations. The method described is also applicable to other molecular methods for a variety of laboratory diagnostics where quantitative characterization of nucleic acids is needed.  相似文献

The new pincer ligand 2,6-bis[(1,3-di-tert-butylimidazolin-2-imino)methyl]pyridine (TL(tBu)) has been prepared in high yield from 2,6-bis(hydroxymethyl)pyridine (1) and 1,3-di-tert-butylimidazolin-2-imine (3). Reaction of TL(tBu) with [Cu(MeCN)4]PF6 affords the highly reactive copper(I) complex [(TL(tBu))Cu]PF6, [5]PF6, which forms the stable copper(I) isocyanide complexes [6a]PF6 (nu(CN) = 2179 cm(-1)) and [6b]PF6 (nu(CN) = 2140 cm(-1)) upon addition of tert-butyl or 2,6-dimethylphenyl isocyanide, respectively. For the cations 6a and 6b, DFT calculations reveal ground-state electronic structures of the type [(TL(tBu)-kappaN(1):kappaN(2))Cu(CNR)] with tricoordinate geometries around the copper atoms. Exposure of [5]PF6 to the air readily leads to trapping of atmospheric CO2 to form the square-planar complex [(TL(tBu))Cu(HCO3-kappaO)]PF6, [7]PF6, with the bicarbonate ligand adopting a rarely observed monodentate coordination mode. In chlorinated solvents such as dichloromethane or chloroform, [5]PF(6) rapidly abstracts chloride by reductive dechlorination of the solvent to yield [(TL(tBu))CuCl]PF6, [8]PF6 quantitatively. Reaction of TL(tBu) with copper(I) bromide or chloride affords complexes 9a and 9b, respectively, for which X-ray diffraction analysis, low-temperature NMR experiments and DFT calculations reveal the presence of a kappa(2)-coordinated ligand of the type [(TL(tBu)-kappaN(1):kappaN(2))CuX]. In solution, complex 9b undergoes slow disproportionation forming the mixed-valence copper(II)/copper(I) system [(TL(tBu))CuCl][CuCl2], [8]CuCl2 with a linear dichlorocuprate(I) counterion.  相似文献
