The sensitive BRET system for the homogeneous immunoassay of a low‐molecular weight antigen was developed using progesterone as an example. Two thermostable mutants of the Luciola mingrelica firefly luciferase (Luc)—the “red” mutant with λmax.em = 590 nm (RedLuc) and the “green” mutant with λmax.em = 550 nm (GreenLuc)—were tested as the donors. The water‐soluble Alexa Fluor 610× (AF) dye was selected as the acceptor because its two absorption maxima, located at 550 and 610 nm, are close to the bioluminescence maxima of the GreenLuc and RedLuc, respectively. The methods for the synthesis of the luciferase–progesterone (Luc–Pg) conjugate and the conjugate of the dye and the polyclonal antiprogesterone antibody (AF–Ab) were developed. Both conjugates retained their functional properties, had high antigen–antibody binding activity, and demonstrated a high BRET signal. The homogeneous immunoassay system based on the BRET from the firefly luciferase to the synthetic dye was established to assay progesterone as a model antigen. Optimization of the assay conditions, the composition of the reaction mixture, and the concentrations of the donor and the acceptor made it possible to reach the minimum detectable progesterone concentration of 0.5 ng mL?1.  相似文献

“Chemistry‐on‐the‐complex” synthetic methods have allowed the selective addition of 1‐ethynylpyrene appendages to the 3‐, 5‐, 3,8‐ and 5,6‐positions of IrIII‐coordinated 1,10‐phenanthroline via Sonogashira cross‐coupling. The resulting suite of complexes has given rise to the first rationalization of their absorption and emission properties as a function of the number and position of the pyrene moieties. Strong absorption in the visible region (e.g. 3,8‐substituted Ir‐3 : λabs=481 nm, ?=52 400 m ?1 cm?1) and long‐lived triplet excited states (e.g. 5‐substituted Ir‐2 : τT=367.7 μs) were observed for the complexes in deaerated CH2Cl2. On testing the series as triplet sensitizers for triplet–triplet annihilation upconversion, those IrIII complexes bearing pyrenyl appendages at the 3‐ and 3,8‐positions ( Ir‐1 , Ir‐3 ) were found to give optimal upconversion quantum yields (30.2 % and 31.6 % respectively).  相似文献

An efficient general synthetic approach giving the possibility for facile, rapid and cheap access to a wide range of novel nitrogen-bisphosphonates (N-BPs) as potent drug candidates, based on the reaction of mono- and bis-propargyl-substituted bisphosphonates with a variety of azides under Cu(i) catalysis ("click" methodology), has been developed. The method allows the incorporation of two functionalities into the N-BP molecule simultaneously, as well as to ligate in situ two N-BPs to one another via the one-pot reaction of organic dibromides with propargyl-substituted bisphosphonates, generating both the diazide and Cu(I) moieties.  相似文献

We have investigated the nucleation and growth of sodium chloride in both single quiescent charged droplets and charged droplet populations that were levitated in an electrodynamic levitation trap (EDLT). In both cases, the magnitude of a droplet's net excess charge (ions(DNEC)) influenced NaCl nucleation and growth, albeit in different capacities. We have termed the phenomenon ion-induced nucleation in solution. For single quiescent levitated droplets, an increase in ions(DNEC) resulted in a significant promotion of NaCl nucleation, as determined by the number of crystals observed. For levitated droplet populations, a change in NaCl crystal habit, from regular cubic shapes to dome-shaped dendrites, was observed once a surface charge density threshold of -9 x 10(-4) e.nm(-2) was surpassed. Although promotion of NaCl nucleation was observed for droplet population experiments, this can be attributed in part to the increased rate of solvent evaporation observed for levitated droplet populations having a high net charge. Promotion of nucleation was also observed for two organic acids, 2,4,6-trihydroxyacetophenone monohydrate (THAP) and alpha-cyano-4-hydroxycinnamic acid (CHCA). These results are of direct relevance to processes that occur in both soft-ionization techniques for mass spectrometry and to a variety of industrial processes. To this end, we have demonstrated the use of ion-induced nucleation in solution to form ammonium nitrate particles from levitated droplets to be used in in vitro toxicology studies of ambient particle types.  相似文献

DNA complexes with cationic polymers (polyvinylamine (PVA), polyallylamine (PAA), polydimethylaminoethylmethacrylate (PDMAEM), poly-(N,N,N-trimethylammonio)ethyl methacrylate chloride (PTMAEM), poly-l-lysine (PLL)) were investigated. It was shown that volume and persistent length of DNA do not change essentially at low cationic polymer concentration in a solution. DNA packaging in 0.005 M NaCl was observed at charge ratio N/P ≈ 1. Secondary DNA structure in complexes was not disrupted, and DNA was protected from protonation. The comparison between DNA packaging in complexes with polycations and DNA condensation induced by trivalent ions was made.  相似文献

The bacterial infection of post-operative wounds is a common health problem. Therefore, it is important to investigate fast and accurate methods of identifying bacteria in clinical samples. The aim of the study was to analyse the use of the MALDI-TOF MS technique to identify microorganism wounds that are difficult to heal. The most common bacteria are Escherichia coli, Staphylococcus spp., and Enterococcus spp. We also demonstrate the effect of culture conditions, such as the used growth medium (solid: Brain Heart Infusion Agar, Mueller Hilton Agar, Glucose Bromocresol Purple Agar, and Vancomycin Resistance Enterococci Agar Base and liquid: Tryptic Soy Broth and BACTEC Lytic/10 Anaerobic/F), the incubation time (4, 6, and 24h), and the method of the preparation of bacterial protein extracts (the standard method based on the Bruker guideline, the Sepsityper method) to identify factors and the quality of the obtained mass spectra. By comparing the protein profiles of bacteria from patients not treated with antibiotics to those treated with antibiotics based on the presence/absence of specific signals and using the UniProt platform, it was possible to predict the probable mechanism of the action of the antibiotic used and the mechanism of drug resistance.  相似文献

The successful Diels-Alder [2+4] cycloaddition of dipyrimidyl acetylene and suitably substituted 2,3,4,5-tetraarylcyclopenta-2,4-dien-1-ones (3-7) generates a series of selectively functionalized hexaarylbenzenes. Each has two pairs of peripheral functional groups (R' and R=tert-butyl 8 and R=methyl 9, methoxy 10, bromo 11, triisopropylsilylethynyl 12) and four ortho-imine nitrogen atoms. The dibromo derivative 11 is a useful precursor for the formation of a mono ethynyl 13 and diethynyl 14 substituted polyphenylene. Changing the dienophile to di(2-thienyl)acetylene gives an S-heteroatom polyphenylene 15. The compounds were fully characterized by using (1)H, (13)C and a range of 2 D NMR spectroscopic techniques, elemental analysis, and mass spectrometry. Oxidative cyclodehydrogenation of dimethoxy hexaphenylbenzene 10 by using iron(III) chloride results in the formation of a spirocyclic dienone 16, which in a separate reaction undergoes dienone/phenol rearrangement to give the first 4-fused-ring, N-heterosuperbenzene (HSB) 17. Six single crystal molecular structures reveal the commonality of unidirectional twisting of the external aromatic rings in these heteroatom polyphenylenes. The twist angles and any H-bonding or interdigitation in these structures are discussed.  相似文献

Nonlinear properties of a multi‐layer stack of graphene sheets are studied. It is predicted that such a structure may support dissipative plasmon‐solitons generated and supported by an external laser radiation. Novel nonlinear equations describing spatial dynamics of the nonlinear plasmons driven by a plane wave in the Otto configuration are derived and the existence of single and multi‐hump dissipative solitons in the graphene structure is predicted.  相似文献

Aniline joins the club: A β-diketiminato copper(I) catalyst enables C-H amination of anilines employing low catalyst loadings to preclude oxidation to the diazene ArN=NAr. Electron-poor anilines are particularly resistant towards diazene formation and participate in the amination of strong and unactivated C-H bonds.
