As benchmarks for judging the catalytic power of sulfate monoesterases, we sought to determine the rates of spontaneous hydrolysis of unactivated alkyl sulfate monoesters by S-O bond cleavage. Neopentyl sulfate proved to be unsuitable for this purpose, since it was found to undergo hydrolysis by a C-O bond cleaving mechanism with rearrangement of its carbon skeleton. Instead, we examined the temperature dependence of the spontaneous hydrolyses of aryl sulfate monoesters, which proceed by S-O cleavage. Extrapolation of a Bronsted plot [log(k(25)(N)) = (-1.81 ± 0.09) pK(a)(LG) + (3.6 ± 0.7)] based on the rate constants at 25 °C for hydrolysis of a series of sulfate monoesters to a pK(a)(LG) value of 16.1, typical of an aliphatic alcohol, yields k(25)(N) = 3 × 10(-26) s(-1). Comparison of that value with established k(cat) values of bacterial sulfatases indicates that these enzymes produce rate enhancements (k(cat)/k(uncat)) of up to 2 × 10(26)-fold for the hydrolysis of sulfate monoesters. These rate enhancements surpass by several orders of magnitude the ~10(21)-fold rate enhancements that are generated by phosphohydrolases, the most powerful biological catalysts previously known. The hydrolytic rates of phosphate and sulfate monoesters are compared directly, and the misleading impression that the two classes of ester are of similar reactivity is dispelled.  相似文献

The final step in the degradation of heparin sulfate involves the enzymatic hydrolysis of its 2-sulfamido groups. To evaluate the power of the corresponding sulfamidases as catalysts, we examined the reaction of N-neopentyl sulfamate at elevated temperatures and found it to undergo specific acid catalyzed hydrolysis even at alkaline pH. A rate constant of 10(-16) s(-1) was calculated using the Eyring equation for water attack on the N-protonated species at pH 7, 25 °C. As a model for the pH neutral reaction, a rate constant for hydroxide attack on (CH(3))(3)CCH(2)N(+)H(2)SO(3)(-) at pH 7, 25 °C was calculated to be 10(-19) s(-1). The corresponding rate enhancement (k(cat)/k(non)) produced by the N-sulfamidase of F. heparinum is approximately 10(16)-fold, which is somewhat larger than those generated by most hydrolytic enzymes but considerably smaller than those generated by S-O cleaving sulfatases.  相似文献

There is interest in monitoring plasma concentrations of N‐desalkylquetiapine in relation to antidepressant effect. A simple LC‐MS/MS method for quetiapine and four metabolites in human plasma (50 μL) has been developed to measure concentrations of these compounds attained during therapy. Analytes and internal standard (quetiapine‐d8) were extracted into butyl acetate–butanol (10:1, v/v) and a portion of the extract analysed by LC‐MS/MS (100 × 2.1 mm i.d. Waters Spherisorb S5SCX; eluent: 50 mmol/L methanolic ammonium acetate, pH* 6.0; flow‐rate 0.5 mL/min; positive ion APCI‐SRM, two transitions per analyte). Assay calibration (human plasma calibrators) was linear across the ranges studied (quetiapine and N‐desalkylquetiapine 5–800, quetiapine sulfoxide 100–15,000, others 2–100 µg/L). Assay validation was as per FDA guidelines. Quetiapine sulfone was found to be unstable and to degrade to quetiapine sulfoxide. In 47 plasma samples from patients prescribed quetiapine (prescribed dose 200–950 mg/day), the (median, range) concentrations found (µg/L) were: quetiapine 83 (7–748), N‐desalkylquetiapine, 127 (7–329), O‐desalkylquetiapine 12 (2–37), 7‐hydroxyquetiapine 3 (<1–48), and quetiapine sulfoxide 3,379 (343–21,704). The analyte concentrations found were comparable to those reported by others except that the concentrations of the sulfoxide were markedly higher. The reason for this discrepancy in unclear. Monitoring of cell cultures in microbioreactors is a crucial task in cell bioassays and toxicological tests. In this work a novel tool based on a miniaturized sensor array fabricated using low-temperature cofired ceramics (LTCC) technology is presented. The developed device is applied to the monitoring of cell-culture media change, detection of the growth of various species, and in toxicological studies performed with the use of cells. The impressive potential of the metallosupramolecular approach in designing new functional magnetic materials constitutes a great scientific challenge for the chemical research community that requires an interdisciplinary collaboration. New fundamental concepts and future applications in nanoscience and nanotechnology will emerge from the study of magnetism as a supramolecular function in metallosupramolecular chemistry. Our recent work on the rich supramolecular coordination chemistry of a novel family of aromatic polyoxalamide (APOXA) ligands with first-row transition metal ions has allowed us to move one step further in the rational design of metallosupramolecular assemblies of increasing structural and magnetic complexity. Thus, we have taken advantage of the new developments of metallosupramolecular chemistry and, in particular, the molecular-programmed self-assembly methods that exploit the coordination preferences of paramagnetic metal ions and suitable designed polytopic ligands. The resulting self-assembled di- and trinuclear metallacyclic complexes with APOXA ligands, either metallacyclophanes or metallacryptands, are indeed ideal model systems for the study of the electron exchange mechanism between paramagnetic metal centers through extended π-conjugated aromatic bridges. So, the influence of different factors such as the topology and conformation of the bridging ligand or the electronic configuration and magnetic anisotropy of the metal ion have been investigated in a systematic way. These oligonuclear metallacyclic complexes can be important in the development of a new class of molecular magnetic devices, such as molecular magnetic wires (MMWs) and switches (MMSs), which are major goals in the field of molecular electronics and spintronics. On the other hand, because of their metal binding capacity through the outer carbonyl-oxygen atoms of the oxamato groups, they can further be used as ligands, referred to as metal–organic ligands (MOLs), toward either coordinatively unsaturated metal complexes or fully solvated metal ions. This well-known “complex-as-ligand” approach affords a wide variety of high-nuclearity metal–organic clusters (MOCs) and high-dimensionality metal–organic polymers (MOPs). The judicious choice of the oligonuclear MOL, ranging from mono- to di- and trinuclear species, has allowed us to control the overall structure and magnetic properties of the final oxamato-bridged multidimensional (nD, n = 0–3) MOCs and MOPs. The intercrossing between short- (nanoscopic) and long-range (macroscopic) magnetic behavior has been investigated in this unique family of oxamato-bridged metallosupramolecular magnetic materials expanding the examples of low-dimensional, single-molecule (SMMs) and single-chain (SCMs) magnets and high-dimensional, open-framework magnets (OFMs), which are brand-new targets in the field of molecular magnetism and materials science.  相似文献

The use of carotenoids as biologically friendly labels for third harmonic generation (THG) microscopy is demonstrated. Carotenoid containing liposomes are used to label cell structures via liposome cell fusion. The THG microscopy labels, called harmonophores, were characterized by measuring the third-order nonlinear susceptibility (χ((3))) of carotenoids: violaxanthin, neoxanthin, lutein, β-carotene, zeaxanthin, canthaxanthin and astaxanthin. The THG ratio method was used, which is based on measuring the THG intensity from two interfaces using a nonlinear optical microscope. The second hyperpolarizability values of carotenoids were extracted from χ((3)) measurements taking into account the refractive index at fundamental and third harmonic wavelengths. The length dependence of the second hyperpolarizability of conjugated polyenes from 9 to 13 double bonds with varying oxygen functional groups was investigated. It appears that the presence of epoxides can have a higher influence than an additional conjugated double bond. Furthermore, labelling of both Drosophila Schneider 2 cells and Drosophila melanogaster larvae myocytes with β-carotene was achieved. This study demonstrates that THG enhancement by carotenoids can be used for nontoxic in vivo labelling of subcellular structures for third harmonic generation microscopy.  相似文献

Luehea candicans Mart. et Zucc. (Tiliaceae) is known as 'a?oita-cavalo' and is one of the most important medicinal plants found in the Brazilian cerrado. The crude methanolic extracts of the branches and leaves and their fractions were evaluated using the following cancer cell lines: MCF-7 (breast), NCI-ADR (breast expressing the multidrug resistance phenotype), NCI-460 (lung), UACC-62 (melanoma), 786-0 (kidney), OVCAR (ovarian), PCO-3 (prostate), HT-29 (colon) and K-562 (leukaemia). The crude methanolic extracts from the branches (B) and leaves (L) were able to inhibit the growth of the K-562 and 786-0 cell lines in a dose-dependent manner, with GI(50) values of 8.1 and 5.4 μg mL(-1), respectively. The hexane (L1), chloroform (L2) and methanol (L4) fractions derived from extract L showed a high selectivity and pronounced cytostatic activity against 786-0 (GI(50) ~ 40 μg mL(-1)). A significant amount of lupeol was isolated from fraction L2. The chloroform (B2) and methanol (B3) fractions derived from extract (B) exhibited less selectivity, showing the highest cytostatic activity against K-562, NCI-ADR, OVCAR, MCF-7 and NCI-460 cells, with GI(50) values between 27 and 40 μg mL(-1). Lupeol, betulin, a mixture of steroids, (-)-epicatechin, vitexin and liriodendrin were isolated from these active fractions.  相似文献

Spontaneous oscillator synchrony occurs when populations of interacting oscillators begin cycling together in the absence of environmental forcing. Synchrony has been documented in many physical and biological systems, including oestrus/menstrual cycles in rats and humans. In previous work we showed that Glaucous-winged Gulls (Larus glaucescens) can lay eggs synchronously on an every-other-day schedule, and that synchrony increases with colony density. Here we pose a discrete-time model of avian ovulation to study the dynamics of synchronization. We prove the existence and uniqueness of an equilibrium solution which bifurcates to increasingly synchronous cycles as colony density increases.  相似文献
