The 2DE is a powerful proteomic technique, with excellent protein separation capabilities where intact proteins are spatially separated by pI and molecular weight. 2DE is commonly used in conjunction with MS to identify proteins of interest. Current 2DE workflow requires several manual processing steps that can lead to experimental variability and sample loss. One such step is the transition between first dimension IEF and second‐dimension SDS‐PAGE, which requires exchanging denaturants and the reduction and alkylation of proteins. This in‐solution‐based equilibration step has been shown to be rather inefficient, losing up to 30% of the original starting material through diffusion effects. We have developed a refinement of this equilibration step using agarose stacking gels poured on top of the second‐dimension SDS‐PAGE gel, referred to as in‐gel equilibration. We show that in‐gel equilibration is effective at reduction and alkylation in SDS‐PAGE gels. Quantification of whole‐cell extracts separated on 2DE gels shows that in‐gel equilibration increases protein retention, decreased intergel variability, and simplifies 2DE workflow.  相似文献

This study was carried out to determine the cytotoxic effect of seven plant extracts and the isolated compounds – syringin and 4-methoxycinnamyl alcohol – on cancerous and non-cancerous cells. The ethanol extract of Foeniculum vulgare was found to exhibit the most significant toxicity with an IC50 value of 19.97 μg/mL on HeLa cells. Bioassay-guided fractionation led to the isolation of two compounds, syringin (1) and 4-methoxycinnamyl alcohol (2). Both compounds showed toxicity against MCF-7, HeLa and DU145 cancer cell line. The results showed that compound 2 showed high toxicity against all the cancer cell lines with IC50 values of 14.24, 7.82 and 22.10 μg/mL, respectively. 4-Methoxycinnamyl alcohol also showed no apoptotic effect in cell cycle analysis after 48 h at a concentration of 10 μg/mL. However, DNA fragmentation study revealed that necrosis took place at a concentration of 10 μg/mL after 48 h exposure.  相似文献

The compound Ba4Fe2I5S4 has been prepared at 1223-1123 K by the "U-assisted" reaction of FeS, BaS, S, and U with BaI2 as a flux. A more rational synthesis was also found; however, the presence of U appears to be essential for the formation of single crystals suitable for X-ray diffraction studies. Ba4Fe2I5S4 crystallizes in a new structure type with two formula units in space group I4/m of the tetragonal system. The structure consists of a Ba-I network penetrated by (1)infinity[Fe2S4] chains. Each Fe atom, which is located on a site with 4 symmetry, is tetrahedrally coordinated to four S atoms. The FeS4 tetrahedra edge-share to form linear (1)infinity[Fe2S4] chains in the [001] direction. The Fe-Fe interatomic distance in these chains is 2.5630(4) A, only about 3% longer than the shortest Fe-Fe distance in -Fe metal. Charge balance dictates that the average formal oxidation state of Fe in these chains is +2.5. The M?ssbauer spectra obtained at 85 and 270 K comprise a single quadrupole doublet that has hyperfine parameters consistent with an average Fe oxidation state of +2.5. The M?ssbauer spectrum obtained at 4.2 K consists of a single magnetic sextet with a small hyperfine field of -15.5 T. This spectrum is also consistent with rapid electron delocalization and an average Fe oxidation state of +2.5. The molar magnetic susceptibility of Ba4Fe2I5S4, obtained between 3.4 and 300 K, qualitatively indicates the presence of weak pseudo-one-dimensional ferromagnetic exchange within a linear chain above 100 K and weak three-dimensional ordering between the chains at lower temperatures.  相似文献

Multifunctional colloidal core-shell nanoparticles of magnetic nanocrystals (of iron oxide or FePt) or gold nanorods encapsulated in silica shells doped with the fluorescent dye, Tris(2,2′-bipyridyl)dichlororuthenium(II) hexahydrate (Rubpy) were synthesized. The as-prepared magnetic nanocrystals are initially hydrophobic and were coated with silica using a microemulsion approach, while the as-prepared gold nanorods are hydrophilic and were coated with silica using a Stöber type of process. Each approach yielded monodisperse nanoparticles with uniform fluorescent dye-doped silica shells. These colloidal heterostructures have the potential to be used as dual-purpose tags—exhibiting a fluorescent signal that could be combined with either dark-field optical contrast (in the case of the gold nanorods), or enhanced contrast in magnetic resonance images (in the case of magnetic nanocrystal cores). The optical and magnetic properties of the fluorescent silica-coated gold nanorods and magnetic nanocrystals are reported.  相似文献

Mimosa, an excellent energy crop candidate because of its high growth yield, also contains, on a dry basis, 0.83% hyperoside and 0.90% quercitrin. Hyperoside has been documented as having anti-inflammatory and diurectic properties, whereas quercitrin may play a role in intestinal repair following chronic mucosal injury. Thus, mimosa might first be extracted for important antioxidant compounds and then used as a feedstock for energy production. This article presents results from studies aimed at determining the effect of three extraction parameters (temperature, solvent composition, and time) on the yield of these important quercetin compounds. Conditions are sought which maximize yield and concentration, whereas complementing subsequent biomass pretreatment, hydrolysis and fermentation.  相似文献

This work involved the development and application of a new analytical procedure for in-situ characterization of the lability of metal species in aquatic systems by using a system equipped with a diffusion membrane and cellulose organomodified with p-aminobenzoic acid groups (DM-Cell-PAB). To this end, the DM-Cell-PAB system was prepared by adding cellulose organomodified with p-aminobenzoic acid groups (Cell-PAB) to pre-purified cellulose bags. After the DM-Cell-PAB system was sealed, it was examined in the laboratory. The in-situ application involved immersing the DM-Cell-PAB system in two different rivers, enabling us to study the relative lability of metal species (Cu, Cd, Fe, Mn, and Ni) as a function of time and quantity of exchanger. The procedure is simple and opens up a new perspective for understanding environmental phenomena relating to the complexation, transport, stability, and lability of metal species in aquatic systems rich in organic matter.  相似文献

Satellite estimates of surface ultraviolet A (UVA) (315–400 nm) from the Global Ozone Monitoring Experiment (GOME)-2 were compared to ground-based measurements at four stations in South Africa for 2015. The comparison of daily exposure and daily maximum irradiance was completed for all-sky and clear-sky conditions. There is a strong linear correlation between the satellite and ground-based data with a correlation coefficient (r) between 0.86 and 0.97 for all-sky conditions. However, at three of the stations the satellite data are underestimated compared to ground-based data with a mean bias error (MBE) between −8.7% and −20.6%. A seasonal analysis indicated that there is a link between the bias in ground-based and GOME-2 UVA and cloud fraction. Factors such as aerosols, surface albedo, altitude and data resolution may contribute to the underestimations found at the three sites. These results indicate that satellite estimates of surface UVA over South Africa do not exhibit the same behavior as other stations around the world and therefore require further validation.  相似文献

The incorporation of synthetic nucleoside analogues into DNA duplexes provides a unique opportunity to probe both structure and function of nucleic acids. We used 1H and 19F NMR and molecular dynamics calculations to determine the solution structures of two similar DNA decamer duplexes, one containing a central G-T mismatched or "wobble" base pair, and one in which the thymine in this base pair is replaced by difluorotoluene (a thymine isostere) creating a G-F pair. Here, we show that the non-hydrogen-bonding G-F pair stacks relatively well into the helix and that the distortions caused by each non-Watson-Crick G-T or G-F base pair are quite localized to a three base pair site around the mismatch. A detailed structural analysis reveals that the absence of hydrogen bonding introduces more dynamic motion into the G-F pair relative to G-T and permits the G-F pair to exhibit stacking and conformational features characteristic of both a Watson-Crick base pair (on the guanine containing strand) and a wobble base pair (on the strand containing the difluorotoluene). We used these results to posit a rationale for recognition and repair of mismatch sites in DNA.  相似文献

A new series of neutral oxamato-bridged M(II)Cu(II) chiral chains of general formula [MCuL(x)(S)(m)(H(2)O)(n)]·aS·bH(2)O [L(1)=(M)-1,1'-binaphthalene-2,2'-bis(oxamate) with M=Mn (1a) and Co (1b); L(2)=(P)-1,1'-binaphthalene-2,2'-bis(oxamate) with M=Mn (2a) and Co (2b)] and the analogous racemic chains of formula [MCuL(3)(S)(m)(H(2)O)(n)]·aS·bH(2)O [L(3)=1,1'-binaphthalene-2,2'-bis(oxamate) with M=Mn (3a) and Co (3b)] have been prepared by reaction of the corresponding dianionic oxamatocopper(II) complex [Cu(L(x))](2-) with Mn(2+) or Co(2+) cations in either dimethylformamide (DMF) or dimethyl sulfoxide (DMSO). Solid circular dichroism (CD) spectra of the bimetallic chain compounds were recorded to establish their chiral and enantiomeric nature. They exhibit maximum positive and negative Cotton effects, each pair of enantiomeric chains being non-superimposable mirror images. The crystal structures of the Mn(II)Cu(II) (1a-3a) and the Co(II)Cu(II) (1b and 2b) chain compounds were solved by single-crystal X-ray diffraction methods. Our attempts to obtain X-ray quality crystals of 3b were unsuccessful. The values of the shortest interchain Mn···Mn and Co···Co distances are indicative of a good isolation of neighbouring chains in the crystal lattice, which is caused by the bulky aromatic ligand. Although all the Mn(II)Cu(II) and Co(II)Cu(II) chains exhibit ferrimagnetic behaviour (-J(MnCu)=18.9-26.6 cm(-1) and -J(CoCu)=19.5-32.5 cm(-1)), only the enantiopure Co(II)Cu(II) chains (1b and 2b) show slow magnetic relaxation at low temperatures (T(B)=0.6-1.8 K), which is a characteristic of single-chain magnets (SCMs) and is related to the magnetic anisotropy of the high-spin Co(II) ion. Analysis of the SCM behaviour of 1b and 2b, based on Glauber's theory for an Ising one-dimensional system, shows a thermally activated mechanism for the magnetic relaxation (Arrhenius law dependence). The energy barriers (E(a)) to reverse the magnetisation direction are 8.2 (1b) and 8.1cm(-1) (2b), whereas the pre-exponential factor (τ(0)) is 1.9×10(-8) (1b) and 6.0×10(-9) s (2b). Interestingly, the racemic Co(II)Cu(II) chain analogue, 3b, showed no evidence of SCM behaviour.  相似文献
