[reaction: see text] Monoenolates of C(2)-symmetric, proline-derived piperazine-2,5-diones were generated and trapped with a variety of electrophiles to produce, in a highly diastereoselective fashion, functionalized diketopiperazines (DKPs). These reactions provide the basis for an asymmetric, desymmetrization strategy toward the marine alkaloids phakellstatin and phakellin. The relative stereochemistry of the functionalized DKPs was confirmed by single-crystal X-ray analysis and/or NOE experiments. Bis-functionalization of the DKPs was also found to proceed with high levels of diastereoselectivity.  相似文献

The tetraphenylimidodiphosphinate [N-(P,P-diphenylphosphinoyl)-P,P-diphenylphosphinimidate] ion forms stable tris-chelates with the Bi(III), In(III), and Ga(III) cations. The crystal and molecular structures of [M{(OPPh(2))(2)N}(3)] (M = Ga, In, Bi) were determined by X-ray diffractometry. The geometry around the bismuth atom in compound 3 displays an approximately C(3)(v)() symmetry. This arrangement suggests the presence of a stereoactive lone pair of electrons, which is located in one of the triangular octahedral faces. Derivative 3 crystallizes in the triclinic space group P&onemacr; with Z = 2, a = 14.006(6) ?, b = 14.185(4) ?, c = 17.609(8) ?, alpha = 88.45(2) degrees, beta = 79.34(2) degrees, and gamma = 78.23(2) degrees. The structures of the gallium(III) and indium(III) tris-chelate oxygen-based complexes (1 and 2, respectively) were compared with the bismuth analogue in order to determine the ligand steric bulk influence on the coordination sphere in the absence of the electron lone pair. Complex 1 crystallizes as the [Ga{(OPPh(2))(2)N}(3)].CH(2)Cl(2) solvate in the triclinic space group P&onemacr;; Z = 2, a = 13.534(4) ?, b = 13.855(4) ?, c = 18.732(7) ?, alpha = 95.48(2) degrees, beta = 98.26(2) degrees, and gamma = 97.84(2) degrees. Crystal data for the benzene solvate of 2, [In{(OPPh(2))(2)N}(3)].C(6)H(6): triclinic space group P&onemacr;, Z = 2, a = 13.542(9) ?, b = 15.622(3) ?, c = 18.063(5) ?, alpha = 98.21(1) degrees, beta = 104.77(0) degrees, and gamma = 92.260(0) degrees.  相似文献

The synthesis of α-substituted N-[((2S)-2-hydroxy-2-phenyl)-ethyl]-2-phenylglycine derivatives is reported. The key step of the sequence is the highly diastereoselective alkylation of (6R)-2,3,5,6-tetrahydro-3,6-diaryl-N-[(2′R)-(2′-methyl)phenylmethyl]-4H-1,4-oxazin-2-ones after deprotonation with t-BuOK. Opening of the resulting oxazinone with ethanolic KOH, followed by hydrogenolysis of the corresponding N-[(2R)-(2-methyl)phenylmethyl] compound to furnish the expected 2-phenylglycine derivative, is also described.  相似文献

The hypothesis that the degree of hydration of poly(oxyethylene) (POE) in aqueous solution depends on the mole ratio of water molecules to ether oxygen atoms in the molecule has been verified by studying the isotropic Raman spectra in the O−H stretching region for four short-chain POEs (C 1E n C 1 withn=1−4). Excellent coincidence of the O−H stretching Raman band for all four POEs studied in the range of mole ratio H2O/O ether from 25 to 0.6 was observed, thus confirming the assumption stated above. A conclusion that all ether oxygen atoms in the POE molecule participate in hydrogen bonding with water molecules has been made.  相似文献

Endoplasmic reticulum (ER) has been prepared and analysed from germinating and developing castor bean endosperm. A combination of one- and two-dimensional (1-D and 2-D) gel electrophoresis was used to study the complexity of sample and protein differences between the two stages. The ER of the developing oilseed is central to the synthesis, sorting and storage of protein and lipid reserves while the germinating seed is concerned with their degradation. Sample complexity has been reduced by separation of ER proteins into lumenal, peripheral membrane and integral membrane subfractions. Membrane proteins pose specific problems in aggregation and binding to passive surfaces. We have overcome this by collection of membranes at density gradient interfaces and by silanization of plastic ware. Several major components have been identified from 1-D gels by N-terminal sequencing and matrix-assisted laser desorption/ionization (MALDI) peptide mass fingerprints. These include protein disulphide isomerase (PDI), calreticulin and developing-ER-specific oleate-12-hydroxylase involved in the biosynthesis of ricinoleic acid. In excess of 300 spots are detectable in each developmental fraction by high sensitivity 2-D gels. This is the first 2-D electrophoretic analysis of plant ER. These gels reveal significant differences between germinating and developing ER. Preparative loading 2-D gels of germinating ER have been run and 14 selected spots characterized by quadrupole time of flight tandem mass spectrometry (Q-TOF MS/MS). Ten of these proteins were assigned function on the basis of identity with existing castor database entries, or by homology with other species. Two proteins, aspartate proteinase precursor and N-carbamyl-L-aminohydrolase-like protein, appear to be absent from developing profiles. Most of the proteins identified are concerned with roles in protein processing and storage, and lipid metabolism which occur in the ER. Data from three of the assigned spots included unidentified peptides indicating the presence of more than one protein in these spots following 2-D electrophoresis. More extensive analysis will have to await developments in genomics but the basic separation technologies to simplify sample identity for a plant ER preparation have been established.  相似文献

Chiral stationary phases (CSPs) based on polymeric (R,R)- or (S,S)-1,2-diaminocyclohexane (DACH) derivatives are synthesized. When bonded to 5 microm porous spherical silica gel, the poly (trans-1,2-cyclohexanediyl-bis acrylamide) based poly-cyclic amine polymer (P-CAP) stationary phases is proved to be effective chiral stationary phases that could be used in the normal-phase mode, polar organic mode and with halogenated solvents mobile phases, if desired. Since these are entirely synthetic CSPs, the elution order of all enantiomers can be reversed between the (R,R) P-CAP and (S,S) P-CAP columns. Because of the high loading of chiral selectors, the columns exhibit very high sample capacities. Thus, P-CAP columns are useful for preparative and semi-preparative enantiomeric separations. The application of these CSPs and optimization of their separations are discussed.  相似文献

Plasmepsin II (PMII), a malarial aspartic protease involved in the catabolism of hemoglobin in parasites of the genus Plasmodium, and renin, a human aspartic protease, share 35% sequence identity in their mature chains. Structures of 4‐arylpiperidine inhibitors complexed to human renin were reported by Roche recently. The major conformational changes, compared to a structure of renin, with a peptidomimetic inhibitor were identified and subsequently modeled in a structure of PMII (Fig. 1). This distorted structure of PMII served as active‐site model for a novel class of PMII inhibitors, according to a structure‐based de novo design approach (Fig. 2). These newly designed inhibitors feature a rigid 7‐azabicyclo[2.2.1]heptane scaffold, which, in its protonated form, is assumed to undergo ionic H‐bonding with the two catalytic Asp residues at the active site of PMII. Two substituents depart from the scaffold for occupancy of either the S1/S3 or S2′‐pocket and the hydrophobic flap pocket, newly created by the conformational changes in PMII. The inhibitors synthesized starting from N‐Boc‐protected 7‐azabicyclo[2.2.1]hept‐2‐ene ( 6 ; Schemes 15) displayed up to single‐digit micromolar activity (IC50 values) toward PMII and good selectivity towards renin. The clear structure? activity relationship (SAR; Table) provides strong validation of the proposed conformational changes in PMII and the occupancy of the resulting hydrophobic flap pocket by our new inhibitors.  相似文献

Summary This work reports the room-temperature stabilization of the Bi4V2-xFexIIO11-1.5x γ ‘ phase, a promising ionic conductive material that finds application in solid oxide fuel cell and oxygen sensor devices. The Fe(II) cation proved to be a better stabilizer than Fe(III), which was previously used, since a lower substitution degree of V5+ is needed for the former. Powder X-ray diffraction, Fourier-transform infrared spectroscopy and differential scanning calorimetry were used in these experiments.  相似文献

The synthesis and oxygen atom transfer (OAT) photoreactivity of a diiron(III) mu-oxo meso-tripentafluorophenyl bisporphyrin appended to a dibenzofuran spacer are presented. Reaction of 4,6-diformyldibenzofuran under standard Lindsey conditions furnishes the parent cofacial porphyrin architecture in a single step. These cofacial porphyrins photocatalyze the oxidation of sulfides and olefins using visible light and molecular oxygen as the terminal oxidant. High turnover numbers reflect the enhanced stability of the electron-deficient diiron(III) mu-oxo bisporphyrin core appended to a dibenzofuran spacer under aerobic conditions.  相似文献

Due to the relatively large spread in the results of microbiological proficiency tests, the z-scores are often not able to detect zero or low results as being ‘bad’ results. This paper describes an adapted z-score based on the average or the standard deviation of the 50% ‘highest’ results. The combination of the adapted z-scores of four samples enables a better judgement of the performance of each laboratory.
