An ionic liquid‐based ultrasound‐assisted extraction method has been developed for the effective extraction of fangchinoline and tetrandrine from Stephaniae tetrandrae. The effects of some ultrasound‐assisted extraction parameters including the concentration of [BMIM][BF4], pH, ultrasonic power and time were investigated to optimize the ultrasound‐assisted extraction conditions. Compared to the regular ultrasound‐assisted extraction and traditional refluent extraction, the proposed [BMIM][BF4]‐based ultrasound‐assisted extraction offered shorter extraction times (from 6 h to 40 min) and remarkable higher efficiencies (approximately 30% improved), which supported the suitability of the proposed approach. In addition, the proposed approach was confirmed by the good correlation coefficient (R2), recovery and reproducibility (RSD, n = 5), which were in the range of 0.9992–0.9995, 85.5–101.1%, and 1.87–4.33%, respectively.  相似文献

Gynostemma pentaphyllum (Thunb.) Makino represents the popular health food and supplemental product with broad pharmacological activities. The highly polar glycosides, including flavonoids and saponins, are major effective active components that contain diverse sugar positions and quantities, which result in diverse chemical polarities, making it challenging to separate and isolate these components. The present work described the rapid and efficient linear gradient counter-current chromatography to preparatively separate glycosides from aboveground parts of G. pentaphyllum. Besides, the ethyl acetate and n-butanol binary mobile phases were achieved through adjusting associated proportions. Six glycosides, including quercetin-3-O-neohesperidoside ( 1 ), kaempferol-3-O-robinobioside ( 2 ), kaempferol-3-O-neohesperidoside ( 3 ), gypenoside LVI ( 4 ), ginsenoside Rb3 ( 5 ), and gypenoside XLVI ( 6 ), were isolated at the purities greater than 98%. Moreover, electrospray ionization mass spectrometry and nuclear magnetic resonance tandem mass spectrometry were conducted for structural identification. According to our findings, the established linear gradient counter-current chromatography was an efficient approach to separate the highly polar glycosides from aboveground parts of G. pentaphyllum. Our proposed strategy can be used to separate active compounds from other complex natural products.  相似文献

With the ever-increasing threat posed by the multi-drug resistance of bacteria, the development of non-antibiotic agents for the broad-spectrum eradication of clinically prevalent superbugs remains a global challenge. Here, we demonstrate the simple supramolecular self-assembly of structurally defined graphene nanoribbons (GNRs) with a cationic porphyrin (Pp4N) to afford unique one-dimensional wire-like GNR superstructures coated with Pp4N nanoparticles. This Pp4N/GNR nanocomposite displays excellent dual-modal properties with significant reactive-oxygen-species (ROS) production (in photodynamic therapy) and temperature elevation (in photothermal therapy) upon light irradiation at 660 and 808 nm, respectively. This combined approach proved synergistic, providing an impressive antimicrobial effect that led to the complete annihilation of a wide spectrum of Gram-positive, Gram-negative, and drug-resistant bacteria both in vitro and in vivo. The study also unveils the promise of GNRs as a new platform to develop dual-modal antimicrobial agents that are able to overcome antibiotic resistance.  相似文献

The study explored the isolation and characterisation of three compounds of high purity salvianolic acid B freeze-dried powder extracted from Salvia miltiorrhiza Bunge. A new salvianolic acid, salvianolic acid V (2) together with two known compounds (3–4) was identified. The antibacterial activity tests showed that compound 2 combined with clinical antibiotics such as Levofloxacin or Colistin sulphate together exhibited potent effects against MRSA or Acinetobacter baumanii. This report has considerably extended our knowledge about the diversity and bioactivity of caffeic acid derivatives from S. miltiorrhiza.  相似文献

Altertoxin I (ATX I) is one of the common mycotoxins produced by genus Alternaria which is a common food pathogen of fruits and grains. To prepare enough quantity of pure ATX I for further research of mutagenicity and toxicology tests, a novel method using preparative high-speed counter-current chromatography (HSCCC) was developed. The ethyl acetate crude extracts of the acetone washes obtained after fermentation of Alternaria sp. was separated using a two-phase solvent system composed of n-hexane–ethyl acetate–methanol–water (2:5:5:6, v/v). Collected fractions were analyzed by LC and identified by EI–MS and NMR analysis. The technique can isolate mg levels of the target compound per run.

Salvianolic acid B was separated and purified from Salvia miltiorrhiza Bunge (danshen) by microbial transformation together with chromatography of microsphere resin. The aqueous extract of danshen was transformed by Fusarium graminearum in a bioreactor containing phosphate buffer (PBS), in which rosmarinic acid was transformed into danshensu and caffeic acid and the yield of salvianolic acid B was higher than 85%. After biotransformation, salvianolic acid B was purified by microsphere resin. A parallel test for making a comparison of microsphere resin chromatography between elution by methanol water solution and water was done. The purity of salvianolic acid B was up to 95% at the yield of 62% when impurities and salvianolic acid B were eluted by 45% and 55% methanol solution respectively. The purity of salvianolic acid B was up to 99% at the yield of 90% when distilled water was used to elute the impurities and salvianolic acid B. The total yield of salvianolic acid B was up to 75% at the purity over 99% while biotransformation combined with microsphere resin chromatography by water elution. Microbial biotransformation together with water elution of microsphere resin supplied an efficient method to eliminate the micromolecular impurities and a possible method to purify water-soluble compounds in traditional Chinese medicine.  相似文献
