Protonation states of water ligands and oxo bridges are intimately involved in tuning the electronic structures and oxidation potentials of the oxygen evolving complex (OEC) in Photosystem II, steering the mechanistic pathway, which involves at least five redox state intermediates S(n) (n = 0-4) resulting in the oxidation of water to molecular oxygen. Although protons are practically invisible in protein crystallography, their effects on the electronic structure and magnetic properties of metal active sites can be probed using spectroscopy. With the twin purpose of aiding the interpretation of the complex electron paramagnetic resonance (EPR) spectroscopic data of the OEC and of improving the view of the cluster at the atomic level, a complete set of protonation configurations for the S(2) state of the OEC were investigated, and their distinctive effects on magnetic properties of the cluster were evaluated. The most recent X-ray structure of Photosystem II at 1.9 ? resolution was used and refined to obtain the optimum structure for the Mn(4)O(5)Ca core within the protein pocket. Employing this model, a set of 26 structures was constructed that tested various protonation scenarios of the water ligands and oxo bridges. Our results suggest that one of the two water molecules that are proposed to coordinate the outer Mn ion (Mn(A)) of the cluster is deprotonated in the S(2) state, as this leads to optimal experimental agreement, reproducing the correct ground state spin multiplicity (S = 1/2), spin expectation values, and EXAFS-derived metal-metal distances. Deprotonation of Ca(2+)-bound water molecules is strongly disfavored in the S(2) state, but dissociation of one of the two water ligands appears to be facile. The computed isotropic hyperfine couplings presented here allow distinctions between models to be made and call into question the assumption that the largest coupling is always attributable to Mn(III). The present results impose limits for the total charge and the proton configuration of the OEC in the S(2) state, with implications for the cascade of events in the Kok cycle and for the water splitting mechanism.  相似文献

Heteroleptic copper(I) halide complexes containing the bis[2-(diphenylphosphano)phenyl]ether (DPEphos) ligand and the heterocyclic thioamides pyridine-2(1H)-thione (py2SH), pyrimidine-2(1H)-thione (pymtH) or 4,6-dimethylpyrimidine-2(1H)-thione (dmpymtH) have been synthesized and characterized by (1)H-NMR, IR spectroscopy, elemental analyses and melting point determinations. The complexes can be readily obtained by the addition of the thione ligand to a CuX-diphosphane adduct in dichloromethane-ethanol solution. The molecular structure of [CuCl(DPEphos)(dmpymtH)] complex has been established by single-crystal X-ray diffraction. The structure features a tetrahedral copper(I) center with two phosphorus atoms from the chelating diphos ligand, one halogen atom and the exocyclic sulfur atom of the heterocyclic thioamide unit. The complexes are strongly emissive in the solid state at ambient temperature. DFT and TD-DFT calculations were employed to study the structural, electronic and photophysical properties of the novel complexes. Electronic absorption spectra show two broad bands in the regions 275-290 and 380-398 nm of mixed MLCT/IL character. Intense blue-green emission is observed in the region 500-558 nm for complexes having py2SH or dmpymtH thione ligands. The emitting first triplet excited state, T(1) is mainly localized on the thione ligand.  相似文献

The stereochemistry of istanbulin-B has been defined by X-ray crystal-structure analysis. Rings A and B are transfused, with the Me groups at C(4) and C(5) cis. The α,β-unsaturated,γ -lactone ring is essentially planar. Crystals of istanbulin-B are orthorhombic, space group P2121,21, with a = 6.729(2), b = 13.447(3), c = 14.721(3)Â,and Z = 4. The crystal structure was determined by direct phasing and the atomic parameters were subsequently adjusted by least-squares calculations that converged at R 38% over 1179 diffractometer |Fa| values  相似文献

The UV absorption spectrum of acetylperoxy radicals, produced in several photochemical systems, has been investigated using the molecular modulation technique. The transport properties of oxygen in poly(dimethyl siloxane) have been measured using the method of quenching of fluorescence. This paper discusses the uniqueness of this method and its use in measuring the diffusion coefficient of oxygen in unfilled PDMS. The results show (1) a large value for the diffusion coefficient of oxygen in pure PDMS at 25°C, D = 3.55 × 10?5 cm2/s, (2) a low value of the acitivation energy, ED = 4.77 kcal/mole, which was not temperature dependent in the ranges evaluated, and (3) a large value of the preexponential term, D0 = 0.115 cm2/s. The diffusion coefficient was found to be independent of both the oxygen concentration and fluorophor concentration in the pressure and temperature ranges used in these experiments. The import of these experiments lies in their application to a unique biomedical oxygen sensor which is fast, sensitive, and does not consume oxygen.  相似文献

A key contact in the active site of an aminoglycoside phosphotransferase enzyme (APH(3')-IIIa) is a pi-pi stacking interaction between Tyr42 and the adenine ring of bound nucleotides. We investigated the prevalence of similar Tyr-adenine contacts and found that many different protein systems employ Tyr residues in the recognition of the adenine ring. The geometry of these stacking interactions suggests that electrostatics play a role in the attraction between these aromatic systems. Kinetic and calorimetric experiments on wild-type and mutant forms of APH(3')-IIIa yielded further experimental evidence of the importance of electrostatics in the adenine binding region and suggested that the stacking interaction contributes approximately 2 kcal/mol of binding energy. This type of information concerning the forces that govern nucleotide binding in APH(3')-IIIa will facilitate inhibitor design strategies that target the nucleotide binding site of APH-type enzymes.  相似文献

Resonant inelastic x-ray scattering (RIXS) yields clear evidence of spectroscopic Kondo scales in heavy fermions. In YbInCu4 and YbAgCu4 RIXS probes the Yb2+ component of the hybrid ground state and the temperature dependence of the Yb 4f occupation. We report a sudden valence change at a phase transition in YbInCu4, but a continuous temperature dependence in YbAgCu4, consistent with the predictions of the Anderson impurity model, for a Kondo temperature T(K) = 70 K. These results solve a long-standing controversy and establish RIXS as a quantitative probe of the electronic structure of strongly correlated electron systems.  相似文献

Twelve normal-hearing subjects rated the intelligibility of 35-s, hearing-aid-processed continuous discourse (CD) passages. Three talkers (two male, one female), four hearing aids, and two signal-to-babble (S/B) ratios were used in a completely crossed design. Research questions concerned: (1) ability of listeners to rate intelligibility, (2) sensitivity of hearing aid rankings were based on intelligibility ratings for three CD passages per instrument, and (3) dependence of hearing aid rankings on (a) S/B ratio, and (b) talker characteristics. Results were: (1) listeners were able to rate intelligibility, (2) rankings based on intelligibility ratings of three CD passages per hearing aid were capable of identifying two superior instruments within a group of four hearing aids that were similar in frequency/gain function, (3) listening in a more difficult S/B ratio substantially decreased the sensitivity of the hearing aid rankings for the female talker but had only minor effects on the rankings for the male talkers, and (4) hearing aid intelligibility rankings were found to be different for different talkers. Applications to hearing aid selection are discussed.  相似文献

The 13C? 13C spin coupling constants have been determined in substituted [1-13C]tetrahydronaphthylenes, [5-13C]hexahydrobenzanthracenes and [5-13C]benzanthracene. In addition, the 13C? 13C spin coupling constants for 7-hydroxy[7-13C]benzopyrene, trans-7,8-dihydro[7-13C]benzopyrene-7,8-diol and trans-7,8-dihydro[10-13C]benzopyrene-7,8-diol are reported, together with the one-bond carbon-carbon coupling constants between C-4 and C-5 in selected 4,5-disubstituted benzopyrenes. Values for the directly bonded coupling constants and long-range coupling constants are similar to those reported previously for other aromatic and aliphatic systems. Substituent effects on carbon-carbon coupling are compared for similarly substituted cyclic and acyclic systems.  相似文献
