Stimulated electronic Raman scattering (SERS) in atomic vapours provides a simple method of extending the tuning ranges of pulsed dye lasers well into the infrared region. The special advantages of this technique in comparison with other types of tunable infrared lasers are discussed, and are illustrated by describing a SERS system which uses a modest nitrogen laser-pumped dye laser ( 20 kW). This produces infrared radiation tunable from 2.67 to 3.47m by SERS in caesium vapour, which is contained in a heat pipe oven. Photon conversion efficiencies of up to 50% are obtained. The design of the heat pipe oven, operation of the system and optimization of experimental parameters are described in detail.  相似文献

In the further development and understanding of heme-copper O2-reduction chemistry inspired by the active-site chemistry in cytochrome c oxidase, we describe a dioxygen adduct, [(F8TPP)FeIII-(O22-)-CuII(TMPA)](ClO4) (3), formed by addition of O2 to a 1:1 mixture of the porphyrinate-iron(II) complex (F8TPP)FeII (1a) {F8TPP = tetrakis(2,6-difluorophenyl)porphyrinate dianion} and the copper(I) complex [(TMPA)CuI(MeCN)](ClO4) (1b) {TMPA = tris(2-pyridylmethyl)amine}. Complex 3 forms in preference to heme-only or copper-only binuclear products, is remarkably stable {t1/2 (RT; MeCN) approximately 20 min; lambda max = 412 (Soret), 558 nm; EPR silent}, and is formulated as a peroxo complex on the basis of manometry {1a/1b/O2 = 1:1:1}, MALDI-TOF mass spectrometry {16O2, m/z 1239 [(3 + MeCN)+]; 18O2, m/z 1243}, and resonance Raman spectroscopy {nu(O-O) = 808 cm-1; Delta16O2/18O2 = 46 cm-1; Delta16O2/16/18O2 = 23 cm-1}. Consistent with a mu-eta2:eta1 bridging peroxide ligand, two metal-O stretching frequencies are observed {nu(Fe-O) = 533 cm-1, nu(Fe-O-Cu) = 511 cm-1}, and supporting normal coordinate analysis is presented. 2H and 19F NMR spectroscopies reveal that 3 is high-spin {also muB = 5.1 +/- 0.2, Evans method} with downfield-shifted pyrrole and upfield-shifted TMPA resonances, similar to the pattern observed for the structurally characterized mu-oxo complex [(F8TPP)FeIII-O-CuII(TMPA)]+ (4) (known S = 2 system, antiferromagnetically coupled high-spin FeIII and CuII). M?ssbauer spectroscopy exhibits a sharp quadrupole doublet (zero field; delta = 0.57 mm/s, |DeltaEQ| = 1.14 mm/s) for 3, with isomer shift and magnetic field dependence data indicative of a peroxide ligand and S = 2 formulation. Both UV-visible-monitored stopped-flow kinetics and M?ssbauer spectroscopic studies reveal the formation of heme-only superoxide complex (S)(F8TPP)FeIII-(O2-) (2a) (S = solvent molecule) prior to 3. Thermal decomposition of mu-peroxo complex 3 yields mu-oxo complex 4 with concomitant release of approximately 0.5 mol O2 per mol 3. Characterization of the reaction 1a/1b + O2 --> 2 --> 3 --> 4, presented here, advances our understanding and provides new insights to heme/Cu dioxygen-binding and reduction.  相似文献

A single-stage ion mirror in a time-of-flight (TOF) mass spectrometer (MS) can perform first order velocity focusing of ions initially located at a start focal plane while second order velocity focusing can be achieved using a double-stage reflectron. The situation is quite different when an ion source extraction field is taken into account. In this case which is common in any practical matrix-assisted laser desorption/ionization (MALDI) TOF-MS a single-stage reflectron, for example, cannot perform velocity focusing at all. In this paper an exact, analytic solution for an electric field inside a one-dimensional reflectron has been found to achieve universal temporal focusing of ions having an initial velocity distribution. The general solution is valid for arbitrary electric field distributions in the upstream (from the ion source to the reflectron) and downstream (from the reflectron to an ion detector) regions and in a decelerating part of the reflectron of a reflectron TOF mass spectrometer. The results obtained are especially useful for designing MALDI reflectron TOF mass spectrometers in which the initial velocity distribution of MALDI ions is the major limiting factor for achieving high mass resolution. Using analytical expressions obtained for an arbitrary case, convenient working formulas are derived for the case of a reflectron TOF-MS with a dual-stage extraction ion source. The special case of a MALDI reflectron TOF-MS with an ion source having a low acceleration voltage (or large extraction region) is considered. The formulas derived correct the effect of the acceleration regions in a MALDI ion source and after the reflectron before detecting ions.  相似文献

We introduce a new mixed finite element for solving the 2- and 3-dimensional wave equations and equations of incompressible flow. The element, which we refer to as P1DP2, uses discontinuous piecewise linear functions for velocity and continuous piecewise quadratic functions for pressure. The aim of introducing the mixed formulation is to produce a new flexible element choice for triangular and tetrahedral meshes which satisfies the LBB stability condition and hence has no spurious zero-energy modes. The advantage of this particular element choice is that the mass matrix for velocity is block diagonal so it can be trivially inverted; it also allows the order of the pressure to be increased to quadratic whilst maintaining LBB stability which has benefits in geophysical applications with Coriolis forces. We give a normal mode analysis of the semi-discrete wave equation in one dimension which shows that the element pair is stable, and demonstrate that the element is stable with numerical integrations of the wave equation in two dimensions, an analysis of the resultant discrete Laplace operator in two and three dimensions on various meshes which shows that the element pair does not have any spurious modes. We provide convergence tests for the element pair which confirm that the element is stable since the convergence rate of the numerical solution is quadratic.  相似文献

The higher order fields present in the quadrupole ion trap may have beneficial effects such as increases in mass resolution in the mass-selective instability or resonance ejection modes of operation, but may also result in losses of ions due to nonlinear resonances. In this work, the reduction in ion intensities observed in the mass spectra of polyethylene glycol (PEG 1000) has been utilized to monitor the ion losses resulting from these higher order fields during the rf voltage scans in both the forward and reverse directions. Extensive ion losses were observed in reverse rf voltage scans at q z=0.64 (a z=0), which corresponds to octopole resonance at β z=1/2. The losses depended upon rf voltage scan rate and ion mass being greater for lower scan rates and lower masses. For ions of m/z 877, losses of up to 60% of the stored ions were observed at low scan rates (<1×104 Da/s), but were minimal at higher scan rates. Thus, it is possible to avoid such losses during reverse scans by scanning the region q z=0.64 at rates in excess of 4×104 Da/s. In forward rf voltage scans, ion storage was considerably more reliable, with significant losses observed only at very high scan rates near the region q z=0.78 (hexapole resonance at β z=2/3).  相似文献

Mammalian ribonucleotide reductase (mRR) is a potential target for cancer intervention. A series of lactam-bridged cyclic peptide inhibitors (1-9) of mRR have been synthesized and tested in previous work. These inhibitors consist of cyclic and linear regions, causing their mass spectral characterization to be a challenge. We determined the fragmentation mechanism of cyclic peptides 1-9 using an ion-trap mass spectrometer equipped with an ESI source. Low-energy collision-induced dissociation of sodiated cyclic peptides containing linear branches follows a general pathway. Fragmentation of the linear peptide region produced mainly a and b ions. The ring peptide region was more stable and ring opening required higher collision energy, mainly occurring at the amide bond adjacent to the lactam bridge. The sodium ion, which bound to the carbonyl oxygen of the lactam bridge, acted as a fixed charge site and directed a charge-remote, sequence-specific fragmentation of the ring-opened peptide. Amino acid residues were cleaved sequentially from the C-terminus to the N-terminus. Our findings have established a new way to sequence cyclic peptides containing a lactam bridge based on charge-remote fragmentation. This methodology will permit unambiguous identification of high-affinity ligands within cyclic peptide libraries.  相似文献
