We present a new technique for the direct measurement of the two-point field correlation function of a monochromatic light source based on carrier-encoded lateral shearing interferometry. The technique provides a higher dynamic range and a significant reduction of the amount of data compared with previously demonstrated methods. Reducing agents are potential inhibitors of the microbial growth. We have shown recently that dithiothreitol (DTT), NaBH(4) and H(2) can modify the proton motive force of resting cells of Escherichia coli by increasing the membrane protons permeability [Eur. J. Biochem. 262 (1999) 595]. In the present work, the effect of reducing agents on the resting cells of Lactococcus lactis ssp. cremoris, a species widely employed in dairy processes was investigated. DTT did not affect the acidification nor the DeltapH, in contrast to the effect previously reported on E. coli. The DeltaPsi was slightly increased (30 mV) at low pH (pH 4) in the presence of 31 mM DTT or 2.6 mM NaBH(4). In the case of Na(2)S(2)O(4), small amounts (0.9 mM) drastically decreased the acidification range and this product was shown to abolish the DeltapH. These results are discussed in terms of the diversity of action of the chemical reagents and strain sensitivity. 相似文献

Acetylcholinesterase is one of the most widely used and studied enzymes. Not only does this enzyme regulate neurotransmission (and thus play a key role in neurodegenerative processes) but it is also a prime target for pest control agents and warfare agents. Above all, due to its particularly high turnover rate, acetylcholinesterase is among the most efficient reporter enzymes yet described (for use as enzymatic tracer in immunoassays, for instance). However, its activity is detected through a colorimetric reagent, the Ellman reagent, which displays low detection limits and is often subject to background perturbations. In the course of our search for a more sensitive detection assay, we describe here a first-generation 1,2-dioxetane chemiluminescent probe, based on chemically induced electron exchange luminescence, which has an approximately 10 times lower detection limit than the Ellman colorimetric assay (2.5 x 10(-19) mol for Electrophorus electricus AChE in its tetrameric form). 相似文献

We present the first experimental results on the magnetorheology of suspensions of non-Brownian magnetic ellipsoidal particles.
These particles are made of spherical iron particles linked by polymers and are called polymerized chains. Steady shear, oscillatory
shear, and oscillatory squeeze rheological tests have been performed. The rheological properties of the suspension of polymerized
chains have been compared with those of the suspension of spherical iron particles. In shear flow, both suspensions develop
nearly the same yield stress, while in squeeze flow, the yield stress is several times higher for the suspension of polymerized
chains. We show that the squeezing force of a suspension of spherical particles is an increasing function of the magnetic
field intensity at low magnetic fields but decreases dramatically at higher fields. Surprisingly, this phenomenon, attributed

We follow the damage process of high-density polyethylene during tensile tests. We simultaneously track changes in the density and average orientation of cavities using incoherent light transport. At the same time, we measure the true strain with a video-extensometer and the heat with an infrared imager. We see that the damage process has two major separate steps. First, a globally isotropic nucleation and growth of cavities occurs up to a deformation of about 1.1. Then, at higher deformations, cavities stop growing. Instead, they progressively orient and elongate along the tensile axis. The transition between these two damage processes seems to be related to strong physical and geometrical constraints, also probed through a typical thermal signature. 相似文献
