The lack of a vascular network and poor perfusion is what mostly prevents three‐dimensional (3D) scaffolds from being used in organ repair when reconstruction of thick tissues is needed. Highly‐porous scaffolds made of poly(L ‐lactic acid) (PLLA) are prepared by directional thermally induced phase separation (dTIPS) starting from 1,4‐dioxane/PLLA solutions. The influence of polymer concentration and temperature gradient, in terms of imposed intensity and direction, on pore size and distribution is studied by comparison with scaffolds prepared by isotropic TIPS. The processing parameters are optimized to achieve an overall porosity for the 3D scaffolds of about 93% with a degree of interconnectivity of 91%. The resulting pore network is characterized by the ordered repetition of closely packed dendrite‐like cavities, each one showing stacks of 20 µm large side lamellar branches departing from 70 µm diameter vertical backbones, strongly resembling the vascular patterns. The in vitro biological responses after 1 and 2 weeks are evaluated from mesenchymal (bone marrow stromal) cells (MSC) static culturing. A novel vacuum‐based deep‐seeding method is set up to improve uniform cell penetration down to scaffold thicknesses of over 1 mm. Biological screenings show significant 3D scaffold colonization even after 18 h, while cellular retention is observed up to 14 d in vitro (DIV). Pore architecture‐driven cellular growth is accompanied by cell tendency to preserve their multi‐potency towards differentiation. Confluent tissues as thick as 1 mm were reconstructed taking advantage of the large perfusion enhanced by the highly porous microstructure of the engineered scaffolds, which could successfully serve for applications aimed at vascular nets and angiogenesis.

In this case report we studied alterations in mitochondrial proteins in a patient suffering from recurrent profound muscle weakness, associated with ethylmalonic-adipic aciduria, who had benefited from high dose of riboflavin treatment. Morphological and biochemical alterations included muscle lipid accumulation, low muscle carnitine content, reduction in fatty acid beta-oxidation and reduced activity of complexes I and II of the respiratory chain. Riboflavin therapy partially or totally reversed these symptoms and increased the level of muscle flavin adenine dinucleotide, suggesting that aberrant flavin cofactor metabolism accounted for the disease. Proteomic investigation of muscle mitochondria revealed decrease or absence of several flavoenzymes, enzymes related to flavin cofactor-dependent mitochondrial pathways and mitochondrial or mitochondria-associated calcium-binding proteins. All these deficiencies were completely rescued after riboflavin treatment. This study indicates for the first time a profound involvement of riboflavin/flavin cofactors in modulating the level of a number of functionally coordinated polypeptides involved in fatty acyl-CoA and amino acid metabolism, extending the number of enzymatic pathways altered in riboflavin-responsive multiple acyl-CoA dehydrogenase deficiency.  相似文献

An electronic tongue system based on the array of six metallic potentiometric sensors (metallic wires) was developed and utilized for discrimination of foodstuffs: several types of vinegar and fruit juices. Copper, tin, iron, aluminum, brass and stainless steel wires were included in the array and supplemented by pH glass electrode. The response of potentiometric metallic sensors towards various organic acids has been studied and possible sensitivity mechanisms were discussed. Overall potential changes of metallic sensors were exanimate as complex mixed signals influenced by several components presenting in analyte employing chemometric approach. The multisensor array of such a type can be useful for several applications since of simplicity in handling, low cost of sensors and easy measure procedure.  相似文献

A flow injection method has been developed for determination of silver. The method is based on a reduction reaction with sodium borohydride which leads to the formation of a colloidal species which is monitored at a wavelength of 390 nm.The reaction variables flow rate, sodium borohydride concentration and pH, which affect sensitivity, were investigated and their effects were established using a two-levels, three-factor experimental design. Further optimization of manifold variables (reaction coil and injection volume) allowed us to determine silver in the range 0.050-5.0 mg L−1 with a minimum detectable concentration of 0.050 mg L−1. Silver is added, as biocide, to drinking water for spacecrafts. The chemical species of silver, present in this kind of sample, were characterized by a procedure based on the selective retention of Ag+ onto a 2.2.2. cryptand based substrate followed by determination of the non-bound and bound (after elution) Ag+ by the FIA method. The method optimized was applied to a drinking water sample provided for the launch with the Automated Transfer Vehicle (ATV) module Jule Verne to the International Space Station (March 9, 2008).  相似文献

We study the cross sections for the production of neneural, intermediate mass Higgs boson in the process pptq′φ, pptWφ and ppbZ0φ in the Minimal Supersymmetric Standard Model (φ = H0, h0 and A0) at Supercollider energies. The additional heavy particles (t, W, Z) in the final state can be used for tagging purposes, increasing the signal to background ratio. These reactions are dominated by bg and bg fusion. Their relevance for Higgs particle searches is discussed taking into account the expected efficiencies and purities for b-tagging. We find that, for tan β = 30, the cross sections for ppbZ0φ are larger than 14 pb, over the whole intemediate range of MA0, for A0 and at least one of the other two Higgses. Therefore this reaction is an excellent candidate for the discovery of one or more MSSM Higgs particles.  相似文献

The synthesis of poly(butylene terephthalate) (PBT) nanocomposites by in situ polymerisation method using an organo-modified layered doubled hydroxide is described. 4-Sulfobenzoic acid potassium salt, sodium dodecyl sulfate and dimethyl 5-sulfo isophthalate sodium salt were used as intercalating compounds to improve clay exfoliation. The thermal and dynamic mechanical properties of the nanocomposites prepared were investigated and compared to those of montmorillonite-type nanocomposites prepared by similar synthetic route. The nanocomposites obtained, independent of the degree of exfoliation, showed better dynamic mechanical properties respect to PBT homopolymer while improvements in thermal stability were achieved when dimethyl 5-sulfo isophthalate was used as intercalating agent, highlighting the importance of the interactions of ionic groups covalently linked to the polymer with the charged clay platelets.  相似文献
