To identify small molecules that selectively control hematopoietic stem cell differentiation, we performed an unbiased screen using primary human CD34(+) cells. We identified a plant-derived natural product, euphohelioscopin A, capable of selectively differentiating CD34(+) cells down the granulocyte/monocytic lineage. Euphohelioscopin A also inhibits proliferation and induces differentiation of the myeloid leukemia cell lines THP-1 and HL-60. Mechanistic studies revealed that euphohelioscopin A is an activator of protein kinase C (PKC), and that the promonocytic effects of this natural product are mediated by PKC activation. In addition to shedding insights into normal hematopoiesis, this work may ultimately facilitate the application of stem cell therapies to a host of myeloid dysfunctions. 相似文献

Speech produced in the presence of noise-Lombard speech-is more intelligible in noise than speech produced in quiet, but the origin of this advantage is poorly understood. Some of the benefit appears to arise from auditory factors such as energetic masking release, but a role for linguistic enhancements similar to those exhibited in clear speech is possible. The current study examined the effect of Lombard speech in noise and in quiet for Spanish learners of English. Non-native listeners showed a substantial benefit of Lombard speech in noise, although not quite as large as that displayed by native listeners tested on the same task in an earlier study [Lu and Cooke (2008), J. Acoust. Soc. Am. 124, 3261-3275]. The difference between the two groups is unlikely to be due to energetic masking. However, Lombard speech was less intelligible in quiet for non-native listeners than normal speech. The relatively small difference in Lombard benefit in noise for native and non-native listeners, along with the absence of Lombard benefit in quiet, suggests that any contribution of linguistic enhancements in the Lombard benefit for natives is small. 相似文献

A new technique to obtain the oscillator strength of select rare-earth optical transitions in nanostructured dielectric materials (nanophosphors) is presented. It is based on the experimentally observed nanophosphor lifetime dependence on the embedding medium. A constant oscillator strength and parity-allowed electric dipole transitions of the RE ion emission are assumed. The oscillator strength is obtained from the slope of the 1/τij vs. n(n2+2)2 plot, where τij is the radiative lifetime of transition between states i and j, and n is the index of refraction of the embedding medium. This paper extends the results in [D. Kurowicka, R.M. Cooke, A parametrization of positive definite matrices in terms of partial correlation vines, Linear Algebra Appl. 372 (2003) 225-251]. We show that a partial correlation vine represents a factorization of the determinant of the correlation matrix. We show that the graph of an incompletely specified correlation matrix is chordal if and only if it can be represented as an m-saturated incomplete vine, that is, an incomplete vine for which all edges corresponding to membership-descendents (m-descendents for short) of a specified edge are specified. This enables us to find the set of completions, and also the completion with maximal determinant for matrices corresponding to chordal graphs. 相似文献
