Seeds of Thevetia peruviana were screened for their antifungal photoactivity. Extracts obtained either with n-hexane or dichloromethane were fractionated by column chromatography and further analysed by thin-layer chromatography. All seed extracts and fractions were tested for inhibition of the fungus Cladosporium cucumerinum for the evaluation of photoactive inhibitory effects. Antifungal light-dependent activity was observed for some of the fractions and both crude extracts. The most photoactive fraction was analysed by capillary gas chromatography with mass spectrometry in order to identify its constituents. Two major groups of compounds were identified, terpenes and fatty acids and derivatives. Pulegone, linoleic acid and palmitic acid were the major compounds. Terpenes seem to be the major substances with antifungal photoactivity.  相似文献

The synthesis of eight bifunctional diketopiperazine (DKP) scaffolds is described; these were formally derived from 2,3-diaminopropionic acid and aspartic acid (DKP-1-DKP-7) or glutamic acid (DKP-8) and feature an amine and a carboxylic acid functional group. The scaffolds differ in the configuration at the two stereocenters and the substitution at the diketopiperazinic nitrogen atoms. The bifunctional diketopiperazines were introduced into eight cyclic peptidomimetics containing the Arg-Gly-Asp (RGD) sequence. The resulting RGD peptidomimetics were screened for their ability to inhibit biotinylated vitronectin binding to the purified integrins α(v)β(3) and α(v)β(5), which are involved in tumor angiogenesis. Nanomolar IC(50) values were obtained for the RGD peptidomimetics derived from trans DKP scaffolds (DKP-2-DKP-8). Conformational studies of the cyclic RGD peptidomimetics by (1)H?NMR spectroscopy experiments (VT-NMR and NOESY spectroscopy) in aqueous solution and Monte Carlo/Stochastic Dynamics (MC/SD) simulations revealed that the highest affinity ligands display well-defined preferred conformations featuring intramolecular hydrogen-bonded turn motifs and an extended arrangement of the RGD sequence [Cβ(Arg)-Cβ(Asp) average distance ≥8.8??]. Docking studies were performed, starting from the representative conformations obtained from the MC/SD simulations and taking as a reference model the crystal structure of the extracellular segment of integrin α(v)β(3) complexed with the cyclic pentapeptide, Cilengitide. The highest affinity ligands produced top-ranked poses conserving all the important interactions of the X-ray complex.  相似文献

[PtL(6)X] {X = Cl or NCS and L(6) = 5-mesityl-1,3-di(2-pyridyl)-benzene} display similar luminescence in solution but, in the solid state, the packing of the molecules is different, with short PtPt interactions for X = NCS, leading to a red-shifted emission band. A disposable screen-printed electrode was designed and evaluated for direct detection of chromium(VI) in batch and flow analysis. The carbon screen-printed electrode was modified with a graphite–epoxy composite. The optimal graphite–epoxy matrix contains 37.5% graphite powder, 12.5% diphenylcarbohydrazide, a selective compound for chromium(VI), and 50% epoxy resin. The principal analytical parameters of the potentiometric response in batch and flow analysis were optimized and calculated. The screen-printed sensor exhibits a response time of 20 ± 1 s. In flow analysis, the analytical frequency of sampling is 70 injections per hour using 0.1 M NaNO3 solution at pH 3 as the carrier, a flow rate of 2.5 mL·min−1, and an injection sample volume of 0.50 mL. The sensor shows potentiometric responses that are very selective for chromium(VI) ions and optimal detection limits in both static mode (2.1 × 10−7 M) and online analysis (9.4 × 10−7 M). The disposable potentiometric sensor was employed to determine toxicity levels of chromium(VI) in mineral, tap, and river waters by flow-injection potentiometry and batch potentiometry. Chromium(VI) determination was also carried out with successful results in leachates from municipal solid waste landfills.  相似文献

Proteins govern most aspects of cellular life and, through specific interfaces, are typically involved in intricate protein–protein interaction (PPI) networks and signaling pathways. Subtle up- or downregulation of key protein functions and PPIs results in disease; still, the preferred option to contrast the role of a protein in disease and healthy conditions alike remains its outright shutdown through orthosteric ligands that block its active site. Here, we explore subtler alternatives to modulate proteins and PPIs. Driven by a view of proteins as dynamic entities, we discuss ways to identify allosteric binding sites, which, when targeted by tailored ligands, can induce significant changes in the active site of a protein, and lead to agonistic or antagonistic effects. We also summarize the selective regulation of specific PPIs—either direct or allosteric—and show that effects can be stabilizing as well as destabilizing, depending on how the conformational equilibrium of a protein is shifted.  相似文献

Six new copper(II) complexes of formula [Cu(mu-cbdca)(H2O)]n (1) (cbdca = cyclobutanedicarboxylate), [Cu2(mu-cbdca)2(mu-bipy)2]n (2) (bipy = 4,4'-bipyridine), [Cu(mu-cbdca)(mu-bpe)]n (3) (bpe = 1,2-bis(4-pyridyl)ethane), [Cu(mu-cbdca)(bpy)]2 (4) (bpy = 2,2'-bipyridine), [Cu(terpy)(ClO4)]2(mu-cbdca).H2O (5) (terpy = 2,2':6',2' '-terpyridine), and [Cu(cbdca)(phen) (H2O)].2H2O (6) (phen = 1,10-phenanthroline) were obtained and structurally characterized by X-ray crystallography. Complex 1 is a two-dimensional network with a carboxylate bridging ligand in syn-anti (equatorial-equatorial) coordination mode. Complexes 2 and 3 are formed by chains through syn-anti (equatorial-apical) carboxylate bridges, linked to one another by the corresponding amine giving two-dimensional nets. Complexes 4 and 5 are dinuclear, with the copper ions linked by two oxo (from two different carboxylate) bridging ligands in 4 and with only one carboxylate showing the unusual bis-unidentate mode in complex 5. Complex 6 is mononuclear, with the carboxylate linked to copper(II) in a chelated form. Intermolecular hydrogen bonds and pi-pi stacking interactions build an extended two-dimensional network. Magnetic susceptibility measurements of complexes 1-5 in the temperature range 2-300 K show the occurrence of weak ferromagnetic coupling for 1 and 4 (J = 4.76 and 4.44 cm(-1), respectively) and very weak antiferromagnetic coupling for 2, 3, and 5 (J = -0.94, -0.67, and -1.61 cm(-1), respectively). Structural features and magnetic values are compared with those reported for the similar copper(II) malonate and phenylmalonate complexes.  相似文献

A new class of α-galactosylceramide (αGC) nonglycosidic analogues bearing galacto-configured aminocyclitols as sugar surrogates have been obtained. The aminocyclohexane having a hydroxyl substitution pattern similar to an α-galactoside is efficiently obtained by a sequence involving Evans aldol reaction and ring-closing metathesis with a Grubbs catalyst to give a key intermediate cyclohexene, which has been converted in galacto-aminocyclohexanes that are linked through a secondary amine to a phytoceramide lipid having a cerotyl N-acyl group. Natural Killer T (NKT) cellular assays have resulted in the identification of an active compound, HS161, which has been found to promote NKT cell expansion in vitro in a similar fashion but more weakly than αGC. This compound stimulates the release of Interferon-γ (IFNγ) and Interleukin-4 (IL-4) in iNKT cell culture but with lower potency than αGC. The activation of Invariant Natural Killer T (iNKT) cells by this compound has been confirmed in flow cytometry experiments. Remarkably, when tested in mice, HS161 selectively induces a very strong production of IFN-γ indicative of a potent Th1 cytokine profile. Overall, these data confirm the agonist activity of αGC lipid analogues having charged amino-substituted polar heads and their capacity to modulate the response arising from iNKT cell activation in vivo.  相似文献

Here we describe the identification and preliminary characterization of a new class of pyrrolo(imidazo)quinoxaline hydrazones as florescent probes for Aβ(1-42) fibrils. All the newly developed compounds were able to bind amyloid fibrils formed in vitro and some of them displayed an increase of their fluorescence upon binding. When tested on brain tissue preparations presenting Aβ deposits, the described hydrazones selectively stained amyloid structures and did not display aspecific binding. The hydrazones did not show antifibrillogenic activity and electron microscopy analysis revealed that they do not interfere with fibrils structure. The described pyrrolo(imidazo)quinoxalines could be useful for studying amyloid structures in vitro. Moreover, their experimentally proven ability to cross the blood-brain barrier in mouse opens the possibility of developing these compounds as potential amyloid imaging agents for in vivo applications.  相似文献

Gold(I) and silver(I) complexes of 1-methyl-5-thio-tetrazole (1) have been prepared and the coordination chemistry of this ligand toward metal-phosphine frameworks has been explored. As indicated by IR and Raman data, ligand 1 is deprotonated and the resulted anion acts as a bidentate (S,N)-tetrazole-5-thiolato unit in the new gold(I) complexes, [Au(SCN(4)Me)(PPh(3))] (2), [{Au(SCN(4)Me)}(2)(μ-dppm)] (3), and [{Au(SCN(4)Me)}(2)(μ-dppe)] (4), while it is coordinated only through the sulfur atom as its neutral tetrazole-5-thione form in the silver(I) derivative, [Ag(HSCN(4)Me)(PPh(3))](2)(OTf)(2) (5). Further characterization of the new compounds was performed using multinuclear ((1)H, (13)C, (31)P, (19)F) NMR spectroscopy, mass spectrometry, and DSC measurements. Single-crystal X-ray diffraction studies revealed basically linear P-M-S arrangements in complexes 3-5. The bidentate (S,N) coordination pattern results in a T-shaped (S,N)PAu core in 3 and 4, whereas, in 5, a similar coordination geometry is achieved in the dimer association based on S-bridging ligand 1. Herein, weak (C)H···Au and (C)H···Ag agostic interactions were observed. An intramolecular Au···Au contact occurs in 3, while in 4 intermolecular aurophilic bonds lead to formation of a chain polymer. An intermolecular Ag···Ag contact is also present in the dimer unit of 5. Low-temperature (31)P NMR data for 5 evidenced the presence of monomer and dimer units in solution. Theoretical calculations on model of the complexes 2 and 4 are consistent with the geometries found by X-ray diffraction studies.  相似文献

The guanidine unit in the guise of 2-aminoimidazole in the new structural motif 2-arylamino-1,3-diaza[3]ferrocenophane 4 acts as a binding site for anions. The electrochemical behavior of this compound has been studied by cyclic voltammetry (CV) and differential pulse voltammetry (DPV) and was found to exhibit a quasi reversible oxidation peak, associated to the Fe(II)/Fe(III) redox couple (Ep = 440 mV), and a non-reversible oxidation wave (Ep = 817 mV), probably associated to the oxidation of the C═N unit present in the guanidine bridge. Recognition of AcO(-), PhCO(2)(-), F(-), Cl(-), and Br(-) anions by the free receptor and the less basic anions Br(-), Cl(-), and NO(3)(-) by its monoprotonated form takes place by unusual redox-ratiometric measurements and spectroscopic ((1)H NMR and UV-vis) changes.  相似文献
