High-throughput ligand-based proton NMR screening performed in the presence of a spy molecule and a control molecule is a valuable tool for identifying drug leads. A limitation of the technique is represented by the severe overlap encountered in the screening of large chemical mixtures. An approach for overcoming this overlap problem is the use of multi-selective R(1) filtered and COSY or TOCSY experiments. Application of this methodology to compounds binding to the Sudlow site I of human serum albumin is presented. The screening is performed by simply monitoring the intensity of two signals. The precise measurement of the relative intensity of the two resonances permits determination of the binding constant of the NMR-hit. For a simple competition binding mechanism, the rapidly-derived NMR binding constants are in good agreement with the values derived from full-titration ITC and fluorescence spectroscopy measurements.  相似文献

In this paper are presented the production methods for very "high specific activity" radionuclides (HSA-RN) of vanadium, manganese and thallium which have been developed in our laboratories for labelling different chemical forms of these elements present in the echo-systems in ultra-trace amounts, for metallo-toxicological and bio-kinetic studies. Use was made of both cyclotron and thermal nuclear reactor. If the nuclear reaction product has atomic number different from irradiated target, it is possible separating the radioactive nuclide from irradiated target, without addition of isotopic carrier. This kind of radionuclide is named No Carrier Added, NCA, and his specific activity, As is very high and can reach values close to the theoretical Carrier Free one, CF. The experimental determination of specific activity, chemical and radiochemical purities is mandatory for all these kinds of applications.  相似文献

In a recent series of papers, Miller and co-workers were able to show that His(pi-Me)-based, terminally protected peptides are potent catalysts of the asymmetric acyl transfer reaction, useful for the kinetic resolution of alcohols. In a structure-supporting solvent, one of the most active compounds, an Aib-containing tetrapeptide, is folded in a doubly intramolecularly H-bonded beta-hairpin motif incorporating a type-II' beta-turn conformation. In this work, we have expanded the study of the Miller tetrapeptide by examining a set of analogues and shorter sequences (dipeptide amides), characterized by chiral C(alpha)-tetrasubstituted alpha-amino acids of diverging bulkiness and optical configuration. Peptide synthesis in solution, conformational analysis by FT-IR absorption and (1)H NMR techniques, and screening of catalytic activity as well have been performed. Our results confirm the close relationship between the beta-hairpin 3D-structure and the catalytic activity of the peptides. A tetrapeptide analogue slightly more selective than the Miller compound has been found. However, the terminally protected, industrially more appealing, dipeptide amides are poorly effective.  相似文献

Chromatographic evaluations of a C18 dimethylurea phase in 150 mm x 3.9 mm HPLC columns were performed using the Tanaka and Engelhardt test mixtures. The applicability of the new C18 dimethylurea phase was also evaluated with a mixture of some herbicides and their metabolites. An artificial aging procedure was also performed by passing a potassium phosphate mobile phase buffered at pH 7.0 through C18 50 mm x 3.9 mm dimethylurea columns. The column stability was evaluated by means of the chromatographic parameters obtained for the separation of some compounds from the Neue test mixture, using apolar, polar and highly basic analytes.  相似文献

Perusal of literature data and some new results concerning syndiotactic-specific polymerization of styrene suggest a reaction mechanism accounting for the steric control. Key features of the proposed mechanism are stereorigid ηn coordination of the growing chain end and diastereoselective coordination of the monomer imposed by direct interactions with the ancillary ligand of the metal complex, a pseudotetrahedral chiral Ti(III) or Zr(III) cation, which inverts its configuration after every syndiospecific insertion step.  相似文献

A luminescent bis(salicylaldiminato)zinc(II) Schiff-base complex, 1, is characterized by a concentration dependent second-order nonlinear optical response, related to the degree of aggregation of the complex in a dichloromethane solution. The formation of the monomeric adduct, by addition of a Lewis base, such as pyridine, to concentrated solutions of 1, leads to a switch-on of the quadratic hyperpolarizability. The phototransformation of phenol in aqueous solution was studied with different quinoid compounds, which are usually detected on atmospheric particulate matter: 2-ethylanthraquinone (EtAQ), benzanthracene-7,12-dione (BAD), 5,12-naphthacenequinone (NQ), 9,10-anthraquinone (AQ), and 2,6-dihydroxyanthraquinone (DAQ). All the studied quinones were able to sensitise the phototransformation of phenol. Under blue-light irradiation the approximated, polychromatic quantum yields for phenol photodegradation were in the order AQ > BAD > EtAQ > NQ > DAQ. Quantum mechanical calculations showed that AQ and DAQ have a very different spin distribution in the triplet state (largely located on the carbonyl oxygen and delocalised over the aromatic ring, respectively) that could account for the difference in reactivity. The spin distribution of EtAQ is similar to that of AQ. Under simulated sunlight, EtAQ induced the highest rate of phenol degradation. Radiation-excited EtAQ would oxidise both ground-state EtAQ and phenol; a kinetic model that excludes the ˙OH radical and singlet oxygen as reactive species is supported by the experimental data. Quinones were also able to oxidise nitrite to nitrogen dioxide, thereby inducing phenol nitration. Such a process is a potential source of nitrogen dioxide and nitrophenols in the atmospheric aerosols.  相似文献

The main objective of the study is the accurate modeling of the bubble pressure and of the vapor phase concentration in associating hydrocarbon + alcohol mixtures and the correct comparison with results from the literature. A relatively simple equation of state is used and comparison is done considering various factors that affect the accuracy of the results, so fair and correct conclusions can be drawn. The mathematical complexity of the model, the type and amount of basic properties and the number of adjustable parameters used by the model, among other factors are discussed. The Peng-Robinson equation of state including the Wong-Sandler mixing rules was used. This combination of equations of state and mixing rules have not yet been applied in a systematic way to alcohol + hydrocarbon mixtures at low and moderate pressure, as done in this work, although other complex equation of state models have been used for some selected systems. It is concluded that simple and well-founded models can correlate equilibrium data in these complex mixtures with similar accuracy than other more sophisticated models.  相似文献
