Summary The validity of a method for characterizing stationary phases for reversed-phase, liquid chromatography, based on the use of homologous series, has been evaluated. The method is based on a retention model which describes the dependence of the logarithm of the capacity factor on mobile phase composition and the carbon number of specific homologous series. A first-order as well as a second-order version of this model was investigated. The second-order model proved to be a significant improvement on the first-order model, even for smaller mobile-phase ranges. Nevertheless both models showed a significant lack of fit, reflecting the incompleteness of these models. Therefore, it is very questionable whether this method is suitable to describe HPLC-column characteristics like hydrophobicity and hydrophylicity. 相似文献

A Stokesian dynamics computer simulation based method is presented for the estimation of the bed porosity of slurry-packed capillary liquid chromatography (LC) columns. A colloidally well-described reversed-phase stationary phase-slurry liquid suspension was used as a model system. The applied simulation method takes into account the velocity of the slurry and colloidal interaction forces, as well as inter-particle hydrodynamic interactions. The predicted bed porosities suggest that a lower slurry velocity leads to a denser packing structure due to the increased effect of colloidal repulsion effects. The results of the simulations were compared with the external porosity and chromatographic performance of capillary LC columns that were packed at different filtration and compaction pressures. However, the trends that were observed in the experimental results suggest that hydrodynamic packing parameters have no or little effect on the chromatographic performance of capillary LC columns. Within the experimental parameter window, the chromatographic performance and the column porosity were not influenced by the filtration and compaction pressure, nor by the duration of the compaction process. 相似文献

Parkinson's disease (PD) is the most common neurodegenerative movement disorder. Although a subject of intense research, the etiology of PD remains poorly understood. Recently, several lines of evidence have implicated an intimate link between aberrations in the ubiquitin proteasome system (UPS) and PD pathogenesis. Derangements of the UPS, which normally functions as a type of protein degradation machinery, lead to alterations in protein homeostasis that could conceivably promote the toxic accumulation of proteins detrimental to neuronal survival. Not surprisingly, various cellular and animal models of PD that are based on direct disruption of UPS function reproduce the most prominent features of PD. Although persuasive, new developments in the past few years have in fact raised serious questions about the link between the UPS and PD. Here I review current thoughts and controversies about their relationship and discuss whether strategies aimed at mitigating UPS dysfunction could represent rational ways to intervene in the disease. Summary This paper focuses attention on the potentially larger signal-to-noise ratios produced by microbore columns in comparison
with conventional columns. The increased chromatographic signals by the application of microbore columns are due to the lower
chromatographic dilution of elution profiles which are proportional to the square of the column inner radius. Generally less
than 1μl sample should be injected into microbore systems to obtain the full benefit of the column performance. However, since
more sample can be loaded on conventional columns compared to microbore columns the advantage of improved signal-to-noise
ratio can only be realised in situations where very little sample is available. To inject more than 1μl sample, at the same
time avoiding extra band-broadening effects, suitable injection techniques must be available.
