We present a binocular adaptive optics vision analyzer fully capable of controlling both amplitude and phase of the two complex pupil functions in each eye of the subject. A special feature of the instrument is its comparatively simple setup. A single reflective liquid crystal on silicon spatial light modulator working in pure phase modulation generates the phase profiles for both pupils simultaneously. In addition, another liquid crystal spatial light modulator working in transmission operates in pure intensity modulation to produce a large variety of pupil masks for each eye. Subjects perform visual tasks through any predefined variations of the complex pupil function for both eyes. As an example of the system efficiency, we recorded images of the stimuli through the system as they were projected at the subject's retina. This instrument proves to be extremely versatile for designing and testing novel ophthalmic elements and simulating visual outcomes, as well as for further research of binocular vision.  相似文献

We recently developed a functional neuroimaging technique called encephalographic magnetic resonance imaging (eMRI). Our method acquires rapid single-shot gradient-echo echo-planar MRI (repetition time=47 ms); it attempts to measure an MR signal more directly linked to neuronal electromagnetic activity than existing methods. To increase the likelihood of detecting such an MR signal, we recorded concurrent MRI and scalp electroencephalography (EEG) during fast (20-200 ms), localized, high-amplitude (>50 μV on EEG) cortical discharges in a cohort of focal epilepsy patients. Seen on EEG as interictal spikes, these discharges occur in between seizures and induced easily detectable MR magnitude and phase changes concurrent with the spikes with a lag of milliseconds to tens of milliseconds. Due to the time scale of the responses, localized changes in blood flow or hemoglobin oxygenation are unlikely to cause the MR signal changes that we observed. While the precise underlying mechanisms are unclear, in this study, we empirically investigate one potentially important confounding variable — motion. Head motion in the scanner affects both EEG and MR recording. It can produce brief “spike-like” artifacts on EEG and induce large MR signal changes similar to our interictal spike-related signal changes. In order to explore the possibility that interictal spikes were associated with head motions (although such an association had never been reported), we had previously tracked head position in epilepsy patients during interictal spikes and explicitly demonstrated a lack of associated head motion. However, that study was performed outside the MR scanner, and the root-mean-square error in the head position measurement was 0.7 mm. The large inaccuracy in this measurement therefore did not definitively rule out motion as a possible signal generator. In this study, we instructed healthy subjects to make deliberate brief (<500 ms) head motions inside the MR scanner and imaged these head motions with concurrent EEG and MRI. We compared these artifactual MR and EEG data to genuine interictal spikes. While per-voxel MR and per-electrode EEG time courses for the motion case can mimic the corresponding time courses associated with a genuine interictal spike, head motion can be unambiguously differentiated from interictal spikes via scalp EEG potential maps. Motion induces widespread changes in scalp potential, whereas interictal spikes are localized and have a regional fall-off in amplitude. These findings make bulk head motion an unlikely generator of the large spike-related MR signal changes that we had observed. Further work is required to precisely identify the underlying mechanisms.  相似文献

It is important to consider radiation damage to crystals caused by data collection when solving structures and critical when determining protein function, which can often depend on very subtle structural characteristics. In this study the rate of damage to specific sites in protein crystals cooled at 100 K is found to depend on the energy of the incident X‐ray beam. Several lysozyme crystals were each subjected to 3–26 MGy of cumulative X‐ray exposure by collecting multiple data sets from each crystal at either 9 keV or 14 keV. The integrated electron density surrounding each S atom in the structure was calculated for each data set and the change in electron density was evaluated as a function of dose at the two energies. The rate of electron density decrease per cubic Å per MGy was determined to be greater at 14 keV than at 9 keV for cysteine sulfurs involved in disulphide bridges; no statistically significant differences in the decay rates were found for methionine sulfurs. These preliminary results imply that it might be possible to minimize certain types of specific radiation damage by an appropriate choice of energy. Further experiments studying a variety of photolabile sites over a wider range of energies are needed to confirm this conclusion.  相似文献

On a compact complex manifold of Kähler type, the energy E(Ω) of a Kähler class Ω is given by the squared L 2-norm of the projection onto the space of holomorphic potentials of the scalar curvature of any Kähler metric representing the said class, and any one such metric whose scalar curvature has squared L 2-norm equal to E(Ω) must be an extremal representative of Ω. A strongly extremal metric is an extremal metric representing a critical point of E(Ω) when restricted to the set of Kähler classes of fixed positive top cup product. We study the existence of strongly extremal metrics and critical points of E(Ω) on certain admissible manifolds, producing a number of nontrivial examples of manifolds that carry this type of metrics, and where in many of the cases, the class that they represent is one other than the first Chern class, and some examples of manifolds where these special metrics and classes do not exist. We also provide a detailed analysis of the gradient flow of E(Ω) on admissible ruled surfaces, show that this dynamical system can be extended to one beyond the Kähler cone, and analyze the convergence of solution paths at infinity in terms of conditions on the initial data, in particular proving that for any initial data in the Kähler cone, the corresponding path is defined for all t, and converges to a unique critical class of E(Ω) as time approaches infinity.  相似文献

We consider the numerical simulation of contact problems in elasticity with large deformations. The non-penetration condition is described by means of a signed distance function to the obstacle's boundary. Techniques from level set methods allow for an appropriate numerical approximation of the signed distance function preserving its non-smooth character. The emerging non-convex optimization problem subject to non-smooth inequality constraints is solved by a non-smooth multiscale SQP method in combination with a non-smooth multigrid method as interior solver. Several examples in three space dimensions including applications in biomechanics illustrate the capability of our methods.  相似文献

Confocal and EM analyses revealed that some female Wuchereria bancrofti, obtained from volunteers that received recommended diethylcarbamazine dose regimens, showed few or no embryos. Furthermore, inside the gravid uterus of female W. bancrofti treated with DEC we observed a finely granular, electron-dense material organised as strings of pearls, approximately 70 nm in maximal length surrounding intra-uterine microfilariae and apparently secreted by the embryo. Over the eggshells a similar material was also observed, possibly secreted by the uterine wall. The surface of intra-uterine microfilariae presented a material with identical electron-density to the scattered structures observed inside the egg. Similarly, the sheath of blood microfilariae of W. bancrofti also showed electron-dense projections, with shape and size similar to that observed inside the uterus.  相似文献

Structure elucidation of natural products including the absolute configuration is a complex task that involves different analytical methods like mass spectrometry, NMR spectroscopy, and chemical derivation, which are usually performed after the isolation of the compound of interest. Here, a combination of stable isotope labeling of Photorhabdus and Xenorhabdus strains and their transaminase mutants followed by detailed MS analysis enabled the structure elucidation of novel cyclopeptides named GameXPeptides including their absolute configuration in crude extracts without their actual isolation.  相似文献

Gold nanoparticles supported on TiO(2) (1.2 mol %) catalyze, for the first time under heterogeneous conditions, the cycloisomerization of a series of 1,6-enynes in high yields. In several cases, the product selectivity differs significantly as compared to homogeneous Au(I)-catalysis. Based on product analysis and stereoisotopic studies it is proposed that the major or exclusive pathway involves a 5-exo cyclization mode to form stereoselectively gold cyclopropyl carbenes that undergo a single cleavage pathway, in contrast to homogeneous Au-catalysis where the double cleavage pathway operates substantially.  相似文献
