The pH-selective interaction between the immunoglobulin G (IgG) fragment crystallizable region (Fc region) and the neonatal Fc receptor (FcRn) is critical for prolonging the circulating half-lives of IgG molecules through intracellular trafficking and recycling. By using directed evolution, we successfully identified Fc mutations that improve the pH-dependent binding of human FcRn and prolong the serum persistence of a model IgG antibody and an Fc-fusion protein. Strikingly, trastuzumab-PFc29 and aflibercept-PFc29, a model therapeutic IgG antibody and an Fc-fusion protein, respectively, when combined with our engineered Fc (Q311R/M428L), both exhibited significantly higher serum half-lives in human FcRn transgenic mice than their counterparts with wild-type Fc. Moreover, in a cynomolgus monkey model, trastuzumab-PFc29 displayed a superior pharmacokinetic profile to that of both trastuzumab-YTE and trastuzumab-LS, which contain the well-validated serum half-life extension Fcs YTE (M252Y/S254T/T256E) and LS (M428L/N434S), respectively. Furthermore, the introduction of two identified mutations of PFc29 (Q311R/M428L) into the model antibodies enhanced both complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity activity, which are triggered by the association between IgG Fc and Fc binding ligands and are critical for clearing cancer cells. In addition, the effector functions could be turned off by combining the two mutations of PFc29 with effector function-silencing mutations, but the antibodies maintained their excellent pH-dependent human FcRn binding profile. AbstractWe demonstrate the electrical properties of nano energy harvesters (NEHs) with various textiles for smart textiles that can be applied to the next generation wearable electronics. Output voltages and currents of NEHs with various fabrics, such as a cotton, rayon and wool that have different triboelectricity were measured. Cotton, rayon and wool shows the maximum output voltages of 1.250, 3.313 and 4.063 V, respectively. In addition, output currents of those textiles were 0.75, 4.4, 1.063 μA, respectively. Wool, in particular, which has the highest triboelectricity of 350 V exhibits the highest output voltage. 相似文献

Dyglomera® is an aqueous ethanol extract of the fruit pods of Dichrostachys glomerata, a Cameroonian spice. Several studies have shown its anti-diabetic and anti-obesity effects. However, the underlying mechanisms for such effects remain unclear. Thus, the objective of this study was to investigate the anti-obesity effect of Dyglomera® and its underlying mechanisms in mice with high-fat diet-induced obesity and 3T3-L1 adipocytes. Our results revealed that Dyglomera® inhibited adipogenesis and lipogenesis by regulating AMPK phosphorylation in white adipose tissues (WATs) and 3T3-L1 adipocytes and promoted lipolysis by increasing the expression of lipolysis-related proteins. These results suggest that Dyglomera® can be used as an effective dietary supplement for treating obesity due to its modulating effect on adipogenesis/lipogenesis and lipolysis. 相似文献

Nanotube/nanoparticle hybrid structures are prepared by forming Au and Pt nanoparticles on the sidewalls of single-walled carbon nanotubes. Reducing agent or catalyst-free electroless deposition, which purely utilizes the redox potential difference between Au3+, Pt2+, and the carbon nanotube, is the main driving force for this reaction. It is also shown that carbon nanotubes act as a template for wire-like metal structures. An electrochemical synthesis strategy for the production of nanostructured films was developed by combining self-assembly of surfactant-inorganic aggregates at solid-liquid interfaces and an electrodeposition process. Through this approach high quality nanostructured ZnO films were cathodically deposited from a plating solution containing 0.1 wt % of sodium dodecyl sulfate (SDS). The resulting ZnO films possess lamellar structures with two different repeat distances, d001 = 31.7 A and d001* = 27.5 A, both of which feature well-defined long range order. Due to kinetically controlled surfactant-inorganic assembly during the deposition process, the film exhibits a wide distribution of the stacking directions of the ZnO layers, which will allow facile access of the guest molecules and analytes to the interlayers. The synthetic mechanism used here can be generalized to generate nanostructured films of other semiconducting and metallic materials with architectures that cannot be assembled by other means. 相似文献

Thermal decomposition of dimethyl azo(bisisobutyrate) in a solution containing C(60) produced 1,4- and 1, 16-di(2-carbomethoxy-2-propyl)-1,x-dihydro[60]fullerenes in yields of 21% and 27%, respectively, based on reacted C(60). The structure of this first 1,16-dialkyl-1,16-dihydro[60]fullerene was assigned from (13)C 2D INADEQUATE NMR spectra. The 1,16-isomer has first and second electrochemical reduction potentials shifted positively by 0. 18 V relative to those of the 1,4-isomer. From the close similarity of all spectral, chromatographic, and electrochemical data, the previously unassigned isomer of 1,x-di(2-cyano-2-propyl)-1, x-dihydro[60]fullerene, which was obtained from azo(bisisobutyronitrile) and C(60), is also a 1,16-isomer. 相似文献

An ultra high molecular weight polyethylene was irradiated with the electron beam at dose levels ranging from 100 kGY to 1 MGy. The microstructures of the irradiated samples were characterized by FTIR, gel fraction measurement, DSC, and small‐ and wide‐angle X‐ray scattering. For the mechanical properties, a static tensile test and creep experiment were also performed. The crosslinking and the crystal morphology changes were the main microstructural changes to influence the mechanical properties. It was found that 250 kGy appeared to be the optimal dose level to induce crosslinks in the amorphous area and recrystalliztion in the crystal lamellae. At doses above 250 kGy, the electron beam penetrates into the crystal domains, resulting in crosslinks in the crystal domains and reduction in the crystal size and crystallinity. The static mechanical properties (modulus, strength) and the creep resistance were enhanced by the electron beam irradiation. The behavior of a medium energy (~1 kJ) pulse-power-driven vacuum spark is shown to depend on the electrode material and form of the anode in otherwise similar conditions of operation. The dynamical evolution of the discharge is followed from its soft X-ray emission. The electrode materials compared are titanium and aluminum with a form of anode that is tubular or conical. The use of a tubular anode favors a more uniform sheath and a better formation of a dense Z-pinch and the ensuing hot spots are found to be at least twice as bright. The position of the brightest spots differs according to the material and the electrode shape, and is found to coincide with the shape of the sheath. The energy output is measured and the density of the plasma in the hot spots may be calculated 相似文献
