Atomic force microscopy probing of cell elasticity   总被引:3,自引:0,他引:3  
Atomic force microscopy (AFM) has recently provided the great progress in the study of micro- and nanostructures including living cells and cell organelles. Modern AFM techniques allow solving a number of problems of cell biomechanics due to simultaneous evaluation of the local mechanical properties and the topography of the living cells at a high spatial resolution and force sensitivity. Particularly, force spectroscopy is used for mapping mechanical properties of a single cell that provides information on cellular structures including cytoskeleton structure.

This entry is aimed to review the recent AFM applications for the study of dynamics and mechanical properties of intact cells associated with different cell events such as locomotion, differentiation and aging, physiological activation and electromotility, as well as cell pathology. Local mechanical characteristics of different cell types including muscle cells, endothelial and epithelial cells, neurons and glial cells, fibroblasts and osteoblasts, blood cells and sensory cells are analyzed in this paper.  相似文献

Atomic force microscopy (AFM) was used to study surface layers of fixed intact erythrocytes. Advantages of simultaneous analysis of surface topography and lateral force maps in the investigation of cytoskeleton structure were shown. Fractal analysis was applied to the lateral force maps of erythrocyte surfaces to evaluate the complexity of the cytoskeleton. Peroxynitrite was used as an oxidant to induce changes in the cytoskeleton structure of intact erythrocytes. Peroxynitrite action on whole blood leads to local abnormalities in the erythrocyte cytoskeleton structure, as well as cytoskeleton reorganization in protruded regions of crenated erythrocytes.  相似文献

The high data point density measurements of 1H→11B cross-polarization (CP) kinetics upon magic-angle spinning (MAS) of [bmim][BF4] confined in mesoporous SBA-15 and MCM-41 were carried out. The complex shaped 11B CP MAS signals were observed in both silica and decomposed into two Lorentz components. This points towards the possibility of bimodal distribution of [bmim][BF4] in the studied confinements. The convergence of classical and non-classical spin coupling models was deduced processing CP kinetic curves. A good fit of the theoretical curves to the experimental data was achieved using both models without any non-random deviations between theory and experiment to appear. The convergence of spin coupling models was discussed in terms of relatively high mobility of BF4 ? anion respect to the cation and the dynamics of anions in pores. These factors delete the borders between spin clusters. The spin diffusion along the pore surfaces in MCM-41 is more than twice faster than in SBA-15.  相似文献

The method of spin-wave resonance has been used to detect in multilayer (Co98P2/Co95P5) N structures a modification of the exchange spin wave spectrum due to the formation of the first, second, and third Brillouin zones in a one-dimensional magnon crystal formed by a periodic modulation of the exchange. Investigation of surface domain walls motion in Co-rich magnetic microwires has been performed in circular and axial magnetic fields. The dc axial magnetic field acceleration of the domain wall motion related to the influence of the axial field on the structure of the moving domain wall has been discovered. Pulsed axial magnetic field induced unidirectional motion of surface domain wall also has been found.  相似文献

We investigate the impact of rotational diffusion on the electrodynamic coupling of fluorescent dye molecules (oscillating electric dipoles) to a tunable planar metallic nanocavity. Fast rotational diffusion of the molecules leads to a rapidly fluctuating mode density of the electromagnetic field along the molecules' dipole axis, which significantly changes their coupling to the field as compared to the opposite limit of fixed dipole orientation. We derive a theoretical treatment of the problem and present experimental results for rhodamine 6G molecules in cavities filled with low and high viscosity liquids. The derived theory and presented experimental method is a powerful tool for determining absolute quantum yield values of fluorescence.  相似文献
