This paper demonstrates a method of using a pH meter to determine the micellar dissociation concentration (mdc) and the. critical micellar concentration (cmc) of eight bile salt samples: sodium cholate (NaC), sodium deoxycholate (NaDC), sodium glycocholate (NaGC), sodium glycodeoxycholate (NaGDC), sodium glyco-chenodeoxycholate (NaGCDC), sodium taurocholate (NaTC), sodium taurodeoxycholate (NaTDC) and sodium taurochenodeoxycholate (NaTCDC). The experiments were performed by diluting the bile salt solutions with freshly distilled water and following the pH changes with a hydrogen ion electrode at 25°C. One break appears in most of the pH-concentration plots of the bile salt solutions, signifying mdc. However, two breaks appear for NaC and NaDC samples, signifying mdc and cmc. The mdc and cmc values are in good agreement with values determined by surface tension and turbidity methods and with data reported in the literature. The method described in this paper is quick, simple and requiring no sample purification. It is the only method which can be used to determine mdc and cmc simultaneously.  相似文献

In this research, stimuli‐responsive porous/hollow nanoparticles were prepared by the self‐assembly method. First, chitosan‐graft‐poly(N‐isopropylacrylamide) (CS‐g‐PNIPAAm) copolymers were synthesized through polymerization of N‐isopropylacrylamide (NIPAAm) monomer in the presence of chitosan (CS) solution using ceric ammounium nitrate as the initiator. Then, the CS‐g‐PNIPAAm copolymers were dissolved in the acetic acid aqueous solution and heated to 40 °C to induce their self‐assembly. After CS‐g‐PNIPAAm assembled to form micelles, a cross‐linking agent was used to reinforce the structure to form nanoparticles. The molecular weight of grafted PNIPAAm on CS chains was changed to investigate its effect on the structure, morphology, thermo‐, and pH‐responsive properties of the nanoparticles. TEM images showed that a porous or hollow structure in the interior of nanoparticles was developed, depending on the medium temperature. The synthesized nanoparticles carried positive charges on the surface and exhibited stimuli‐responsive properties, and their mean diameter thus could be manipulated by changing the pH value and temperature of the environment. The nanoparticles showed a continuous release of the encapsulated doxycycline hyclate up to 10 days during an in vitro release experiment. These porous/hollow particles with environmentally sensitive properties are expected to be used in hydrophilic drug delivery system. © 2010 Wiley Periodicals, Inc. The IPI‐POSS‐modified epoxy resin (IPEP) was prepared from isocyanato‐propyldinethylsilyl‐isobutyl‐POSS (IPI‐POSS) and diglycidyl ether of bisphenol A epoxy resin. The steric hindrance of the IPEP bulky POSS side chain improved the curing activation energies. The POSS particles sizes were about 2–3 nm and dispersed uniformly. At lower IPEP concentration (POSS < 12 wt %), the glass transition temperatures (Tgs) of the IPEP nanocomposites increased from 118 to 170 °C. Ag nanoparticles with diameter in the range of 10–25 nm had been synthesized using a simple sucrose ester micellar-mediated method. Ag nanoparticles were formed by adding AgNO3 solution into the sucrose ester micellar solution containing sodium hydroxide at atmospheric condition after 24 h of aging time. Trace amount of dimethyl formamide (DMF) in the sucrose ester solution served as a reducing agent while NaOH acted as a catalyst. The produced Ag nanoparticles were highly stable in the sucrose ester micellar system as there was no precipitation after 6 months of storage. The as-synthesized Ag nanoparticles were characterized using transmission electron microscope (TEM), X-ray diffractometer (XRD), dynamic light scattering (DLS) and UV–vis spectroscopy (UV–vis). Formation mechanism of Ag nanoparticles in the micellar-mediated synthesis is postulated. The antibacterial properties of the Ag nanoparticles were tested against Methicillin-resistant Staphylococcus aureus (MRSA) (Gram-positive) and Aeromonas hydrophila (Gram-negative) bacteria. This work provides a simple and “green” method for the synthesis of highly stable Ag nanoparticles in aqueous solution with promising antibacterial property.  相似文献

The undecanuclear copper cluster Cu11(μ9‐Se)(μ3‐I)3[Se2P(OEt)2]6 1 , has been isolated along with Cu88‐Se)[Se2P(OEt)2]6 2 , from the reaction of NH4Se2P(OEt)2, Cu(CH3CN)4PF6, and Bu4NI in a molar ratio of 3:2:2 in diethyl ether. The molecular formulation of 1 was confirmed by elemental analysis, positive FAB mass spectrometry, multinuclear NMR (1H, 31P, and 77Se), and X‐ray diffraction. In cluster 1 eleven copper atoms adopt the geometry of a 3,3,4,4,4‐pentacapped trigonal prism with a selenium atom in the center. The coordination geometry for the central, nonacoordinated selenium atom is tricapped trigonal prismatic. In addition, the central core Cu11Se is further stabilized by three iodides and six dsep ligands. Besides, weak inter‐molecular Se···I interactions (3.949–3.972 Å) are uncovered and form a one dimensional polymeric chain.  相似文献

trans, trans‐Farnesyl diphosphate (FPP) serves as a universal substrate for a large family of sesquiterpene cyclases that are responsible for biosynthesis of more than 300 structurally diverse sesquiterpenes in nature. A new FPP substrate analogue, 12‐fluoro‐farnesylphosphonophosphate (12‐F‐F‐CH2PP), was synthesized in this paper for applications on kinetic and mechanistic studies of the enzyme family. Trichodiene synthase (TS), a sesquiterpene cyclase, catalyzes the conversion of trans, trans‐farnesyl diphosphate (FPP) to trichodiene. 12‐F‐F‐CH2PP was tested as a potential inhibitor of TS. Inactivation and inhibition kinetic experiments showed that 12‐F‐F‐CH2PP was not a mechanism‐based inactivator for TS; instead, a mixed‐type reversible inhibition was observed with inhibition constants Ki1 = 2.33 ± 0.50 μM and Ki2 = 25.80 ± 7.70 μM, values close to those previously determined for farnesylphosphonophosphate, Ki1 = 3.25 μM and Ki2 = 9.10 μM. Although 12‐F‐F‐CH2PP did not irreversibly inactivate TS, this new analogue serves as a potential active‐site directed inactivator and mechanistic probe of other sesquiterpene cyclases and FPP‐utilizing enzymes, which utilize FPP as a common acyclic substrate.  相似文献

The micellization of sodium dodecyl sulfate (SDS) in different glycol-water solvent mixtures was studied using the isothermal titration calorimetric (ITC) technique. At the same time, microcalorimetric titrations were also carried out to monitor the binding interaction of SDS and poly(ethylene oxide) (PEO) in the presence of different cosolvents. The demicellization of SDS in mixtures of water and cosolvents is different from that in water due to the reduction in solvent polarity and charge interaction of surfactants. The critical micelle concentration (cmc) first decreases with the addition of a small amount of cosolvents and then increases at higher cosolvent concentrations. The thermodynamics of surfactant micellization can be analyzed using the solubility parameters of solvent mixtures. For the binding interaction between SDS and PEO in different solvent mixtures, the dehydration process at low SDS concentrations is replaced by the chain solubilization process with decreasing solvent polarity. With further reduction in the solvent polarity, the binding interaction between SDS and PEO becomes weak and no aggregates can be formed beyond a certain glycol concentration. The binding interaction between SDS and PEO in different solvent mixtures was analyzed and ascribed to the effects of PEO solubility and hydrophobicity of SDS.  相似文献

Cryogenic single‐particle photoluminescence (PL) spectroscopy has been used with great success to directly observe the heterogeneous photophysical states present in a population of luminescent particles. Cryogenic electron tomography provides complementary nanometer scale structural information to PL spectroscopy, but the two techniques have not been correlated due to technical challenges. Here, we present a method for correlating single‐particle information from these two powerful microscopy modalities. We simultaneously observe PL brightness, emission spectrum, and in‐plane excitation dipole orientation of CdSSe/ZnS quantum dots suspended in vitreous ice. Stable and fluctuating emitters were observed, as well as a surprising splitting of the PL spectrum into two bands with an average energy separation of 80 meV. In some cases, the onset of the splitting corresponded to changes in the in‐plane excitation dipole orientation. These dynamics were assigned to structures of individual quantum dots and the excitation dipoles were visualized in the context of structural features.  相似文献

A series of mononuclear and binuclear cyclometalated platinum(II) complexes containing new terdentate meta-bis(2-pyridoxy)benzene ligands: 3,5-bis(2-pyridoxy)toluene (L1H) and 3,5-bis(2-pyridoxy)-2-dodecylbenzene (L2H): [Pt(L1)Cl] (1), [Pt(L2)Cl] (2), [Pt(L1)(CH3CN)](ClO4) (3), {[Pt(L1)]2(μ-dppm)}(ClO4)2 (4), {[Pt(L2)]2(μ-dppm)}(ClO4)2 (5), {[Pt(L1)]2(μ-pyrazole)}(ClO4) (6), {[Pt(L2)]2(μ-pyrazole)}(ClO4) (7), {[Pt(L1)]2(μ-imidazole)}(ClO4) (8) and {[Pt(L2)]2(μ-imidazole)}(ClO4) (9), have been synthesized and characterized. These ligands are coordinated to platinum(II) in a “pincer”-like manner and the presence of pyridyl donors enhances the availability of the ligand π orbitals for electronic transition. Spectroscopic properties of these cyclometalated complexes were studied. While the non-coplanar nature of the ligands hinders ligand-ligand and metal-metal interactions in these cyclometalated complexes, the presence of long hydrocarbon side chain on ligand L2H seems to alleviate such hindrance. Intermolecular π-π, and possibly Pt-Pt interactions were observed in complex 2 at high concentration.  相似文献

Photodynamic therapy (PDT) with lysosome-targeted photosensitizers induces the intrinsic pathway of apoptosis via the cleavage and activation of the BH3-only protein Bid by proteolytic enzymes released from photodisrupted lysosomes. To investigate the role of Bid in apoptosis induction and the role of damaged lysosomes on cell killing by lysosome-targeted PDT, we compared the responses of wild type and Bid-knock-out murine embryonic fibroblasts toward a mitochondrion/endoplasmic reticulum-binding photosensitizer, Pc 4, and a lysosome-targeted sensitizer, Pc 181. Whereas apoptosis and overall cell killing were induced equally well by Pc 4-PDT in both cell lines, Bid−/− cells were relatively resistant to induction of apoptosis and to overall killing following PDT with Pc 181, particularly at low PDT doses. Thus, Bid is critical for the induction of apoptosis caused by PDT with the lysosome-specific sensitizers, but dispensable for PDT targeted to other membranes.  相似文献
