This study aims to evaluate the relevance of portable Raman and portable mid-Fourier transform infrared (FTIR) reflectance instruments in monitoring the synthetic treatments applied on plaster substrates, a crucial issue in a conservation work. Some polymeric consolidants and protectives have a relatively short life owing to their degradation, and after some years the surface should be retreated. It follows that any information about the presence and composition of the products applied, their chemical transformations and their distribution on the surfaces is essential. For these purposes, conservation scientists should seek and test new in situ methods, and this is of utmost importance especially in the case of buildings, considering their large dimensions and consequent extensive mapping. The effectiveness of portable Raman and portable mid-FTIR reflectance instruments has been compared by analysing a set of laboratory specimens prepared and treated with variable amounts of products belonging to three classes of polymers; the spectroscopic investigation highlighted, for the first time, the limits and the advantages of portable Raman and portable mid-FTIR reflectance instruments in the detection of small amounts of products commonly employed for the conservation of plasters.  相似文献

Numerical modeling of actual structural systems is a very complex task mainly due to the lack of complete knowledge on the involved parameters. Simplified assumptions on the uncertain geometry, material properties and boundary conditions make the numerical model response differ from the actual structural response. Improvements of the finite element (FE) models to obtain accurate response predictions can be achieved by vibration based FE model updating which uses experimental measures to minimize the differences between the numerical and experimental modal features (i.e. natural frequencies and mode shapes). Within this context, probabilistic model updating procedures based on the Bayes’ theorem were recently proposed in the literature in order to take into account the uncertainties affecting the structural parameters and their influence on the structural response. In this paper, a novel framework to efficiently estimate the posterior marginal PDF of the selected model parameters is proposed. First, the main dynamic parameters to be used for model updating are identified by ambient vibration tests on an actual structural system. Second, a first numerical FE model is developed to perform initial sensitivity analysis. Third, a surrogate model based on polynomial chaos is calibrated on the initial FE model to significantly reduce computational costs. Finally, the posterior marginal PDFs of the chosen model parameters are estimated. The effectiveness of the proposed method is demonstrated using a FE numerical model describing a curved cable-stayed footbridge located in Terni (Umbria Region, Central Italy).

N6-Isopentenyladenosine (i6A) is a naturally occurring modified nucleoside displaying in vitro and in vivo antiproliferative and pro-apoptotic properties. In our previous studies, including an in silico inverse virtual screening, NMR experiments and in vitro enzymatic assays, we demonstrated that i6A targeted farnesyl pyrophosphate synthase (FPPS), a key enzyme involved in the mevalonate (MVA) pathway and prenylation of downstream proteins, which are aberrant in several cancers. Following our interest in the anticancer effects of FPPS inhibition, we developed a panel of i6A derivatives bearing bulky aromatic moieties in the N6 position of adenosine. With the aim of clarifying molecular action of N6-benzyladenosine analogs on the FPPS enzyme inhibition and cellular toxicity and proliferation, herein we report the evaluation of the N6-benzyladenosine derivatives’ (compounds 2a–m) effects on cell viability and proliferation on HCT116, DLD-1 (human) and MC38 (murine) colorectal cancer cells (CRC). We found that compounds 2, 2a and 2c showed a persistent antiproliferative effect on human CRC lines and compound 2f exerted a significant effect in impairing the prenylation of RAS and Rap-1A proteins, confirming that the antitumor activity of 2f was related to the ability to inhibit FPPS activity.  相似文献

The reactivity of [NaL·ClO2] cluster anions (L = ClOx; x = 0–3) with sulphur dioxide has been investigated in the gas phase by ion–molecule reaction experiments (IMR) performed in an in-house modified Ion Trap mass spectrometer (IT-MS). The kinetic analysis revealed that SO2 is efficiently oxidised by oxygen-atom (OAT), oxygen-ion (OIT) and double oxygen transfer (DOT) reactions. The main difference from the previously investigated free reactive ClO2 is the occurrence of intracluster OIT and DOT processes, which are mediated by the different ligands of the chlorite anion. This gas-phase study highlights the importance of studying the intrinsic properties of simple reacting species, with the aim of elucidating the elementary steps of complex processes occurring in solution, such as the oxidation of sulphur dioxide.  相似文献

Tritordeum results from the crossbreeding of a wild barley (Hordeum chilense) species with durum wheat (Triticum turgidum spp. turgidum). This hexaploid crop exhibits agronomic and rheological characteristics like soft wheat, resulting in an innovative raw material to produce baked goods. We applied a gel-based proteomic approach on refined flours to evaluate protein expression differences among two widespread tritordeum cultivars (Aucan and Bulel) taking as the reference semolina and flour derived from a durum and a soft wheat cvs, respectively. The products of in vitro digestion of model breads were analyzed to compare bio-accessibility of nutrients and mapping tritordeum bread resistant peptides. Significant differences among the protein profiles of the four flours were highlighted by electrophoresis. The amino acid bio-accessibility and the reducing sugars of tritordeum and wheat breads were comparable. Tritordeum cvs had about 15% higher alpha-amino nitrogen released at the end of the duodenal simulated digestion than soft wheat (p < 0.05). Bulel tritordeum flour, bread and digested bread had about 55% less R5-epitopes compared to the soft wheat. Differences in protein expression found between the two tritordeum cvs reflected in diverse digestion products and allergenic and celiacogenic potential of the duodenal peptides. Proteomic studies of a larger number of tritordeum cvs may be successful in selecting those with good agronomical performances and nutritional advantages.  相似文献
