Standing waves can exist as stable vibrating patterns in perfect structures such as spherical bodies, and inertial rotation of the body causes precession (Bryan's effect). However, an imperfection such as light mass anisotropy destroys the standing waves. In this paper, an imperfection is introduced in the form of light mass anisotropy for a vibrating, slowly rotating spherical body. Assuming this light mass imperfection throughout this paper, the effects of slow rotation and light isotropic viscous damping are considered in a system of variables consisting of the amplitudes of principal and quadrature vibrating patterns, the angle of the rotation of the vibrating pattern (called the precession angle) and the phase shift of the vibrating pattern. We demonstrate how a combination of both qualitative and quantitative analysis (using, inter alia, the method of averaging) predicts that the inertial angular rate does not influence changes with time in the amplitudes of the principal and quadrature vibrations or the phase shift. The light mass imperfection causes changes with time which appear to be of a damped oscillatory nature for both the quadrature component as well as the principal component. The precession angular rate appears to depend on the inertial angular rate as well as the quadrature component of the vibration but is not influenced by the damping factor. It is not directly proportional to the inertial angular rate as is the case for a perfect isotropically damped structure. If the quadrature component is not suppressed, then a “capture effect” appears to occur, namely that the precession angle will not grow at a constant rate but is “captured” and shows periodic behaviour. It is evident that the damping factor does not influence changes with time in the phase shift and that the mass imperfection substantially influences these changes. The phase shift appears to be negative, strictly decreasing and unbounded.  相似文献

Base-catalyzed depolymerization of black liquor retentate (BLR) from the kraft pulping process, followed by ultrafiltration, has been suggested as a means of obtaining low-molecular-weight (LMW) compounds. The chemical complexity of BLR, which consists of a mixture of softwood and hardwood lignin that has undergone several kinds of treatment, leads to a complex mixture of LMW compounds, making the separation of components for the formation of value-added chemicals more difficult. Identifying the phenolic compounds in the LMW fractions obtained under different depolymerization conditions is essential for the upgrading process. In this study, a state-of-the-art nontargeted analysis method using ultra-high-performance supercritical fluid chromatography coupled to high-resolution multiple-stage tandem mass spectrometry (UHPSFC/HRMSn) combined with a Kendrick mass defect-based classification model was applied to analyze the monomers and oligomers in the LMW fractions separated from BLR samples depolymerized at 170–210 °C. The most common phenolic compound types were dimers, followed by monomers. A second round of depolymerization yielded low amounts of monomers and dimers, while a high number of trimers were formed, thought to be the result of repolymerization.  相似文献

On-line hyphenation of extraction with chromatography has been explored in several different types of combinations. However, monitoring the complete process of a dynamic, continuous-flow extraction is not possible with any hyphenated system reported so far. The current work demonstrates that this challenging task can be effectively fulfilled by using a parallel sampling interface, which mimics the concept of comprehensive two-dimensional chromatography. In this study, pressurised hot water extraction was coupled on-line with ultra-high-performance liquid chromatography. The set-up was utilised in a kinetic study of dynamic pressurised hot water extraction of curcuminoids from turmeric powder. Compound-specific extraction curves were obtained, which clearly indicated the rate-limiting factors of the extraction processes under different conditions. Additionally, thermal degradation of curcumin during the extraction could also be demonstrated in some of the extractions.

Cost-effective and rapid monitoring of Salmonella in the meat production chain can contribute to food safety. The objective of this study was to validate an easy-to-use pre-PCR sample preparation method based on a simple boiling protocol for screening of Salmonella in meat and carcass swab samples using a real-time PCR method. The protocol included incubation in buffered peptone water, centrifugation of an aliquot, and a boiling procedure. The validation study included comparative and interlaboratory trials recommended by the Nordic Organization for Validation of Alternative Microbiological Methods (NordVal). The comparative trial was performed against a reference method (NMKL 187, 2007) and a PCR method previously approved by NordVal with a semiautomated magnetic bead-based DNA extraction step using 122 artificially contaminated samples. The LOD was found to be 3.0, 3.2, and 3.4 CFU/sample for the boiling, magnetic bead-based, and NMKL 187 methods, respectively. When comparing the boiling method with the magnetic beads, the relative accuracy (AC), relative sensitivity (SE), and relative specificity (SP) were 98, 102, and 98%, respectively (Cohen's kappa index 0.95). When comparing results obtained by the boiling to the culture-based method, the AC, SE, and SP were found to be 98, 102, and 98%, respectively (kappa index 0.93). In the interlaboratory trial including valid results from 11 laboratories, apart from two false-positive samples by the boiling method combined with PCR, no deviating results were obtained (SP, SE, and AC were 100, 95, and 97%, respectively). This test is under implementation by the Danish meat industry, and can be useful for screening of large number of samples in the meat production, especially for fast release of minced meat with a short shelf life.  相似文献

Pressurized liquid extraction is a “green” technology for the extraction of nutraceuticals from foods and herbal plants. This review discusses the extraction principles and the optimization of the extraction parameters that improves the extraction efficiency. The use of different solvent mixtures and other extraction additives to enhance the efficiency of the extraction are discussed. Dynamic mode of extraction in Pressurized liquid extraction, and the use of combined and hyphenated sample preparation and analytical techniques are presented. This work discusses how different studies used Pressurized liquid extraction to enrich phenolic compounds, lignans, carotenoids, oils and lipids, essential oils and other nutraceuticals from foods and herbal plants.  相似文献

Molecular imprinted polymer produced using quercetin as the imprinting compound was applied for the extraction of flavonol aglycones (quercetin and kaempferol) from Moringa oleifera methanolic extracts obtained using heated reflux extraction method. Identification and quantification of these flavonols in the Moringa extracts was achieved using high performance liquid chromatography with ultra violet detection. Breakthrough volume and retention capacity of molecular imprinted polymer SPE was investigated using a mixture of myricetin, quercetin and kaempferol. The calculated theoretical number of plates was found to be 14, 50 and 8 for myricetin, quercetin and kaempferol, respectively. Calculated adsorption capacities were 2.0, 3.4 and 3.7 μmol/g for myricetin, quercetin and kaempferol, respectively. No myricetin was observed in Moringa methanol extracts. Recoveries of quercetin and kaempferol from Moringa methanol extracts of leaves and flowers ranged from 77 to 85% and 75 to 86%, respectively, demonstrating the feasibility of using the developed molecularly imprinted SPE method for quantitative clean‐up of both of these flavonoids. Using heated reflux extraction combined with molecularly imprinted SPE, quercetin concentrations of 975 ± 58 and 845 ± 32 mg/kg were determined in Moringa leaves and flowers, respectively. However, the concentrations of kaempferol found in leaves and flowers were 2100 ± 176 and 2802 ± 157 mg/kg, respectively.  相似文献

Phlorotannins are bioactive polyphenols in brown macroalgae that make these algae interesting as healthy food. Specific phlorotannins are, however, seldom identified, and extracts from different species are often only analysed for total phenolic content (TPC). In this study, our focus was to identify phlorotannin molecules from Saccharina latissima and Ascophyllum nodosum (a species rich in these compounds) using ultra-high-performance liquid chromatography coupled to high-resolution tandem mass spectrometry (UHPLC-HRMS2). Water and ethanol (30 and 80% v/v) were used at solid:liquid ratios, extraction times and temperatures, proposed to result in high TPC in extracts from other species. The S. latissima extracts, however, did not allow phlorotannin detection by either UHPLC-UV/Vis or UHPLC-HRMS2, despite a TPC response by the Folin–Ciocalteu assay, pinpointing a problem with interference by non-phenolic compounds. Purification by solid phase extraction (SPE) led to purer, more concentrated fractions and identification of four phlorotannin species in A. nodosum and one in S. latissima by UHPLC-HRMS2, using extracts in ethanol 80% v/v at a solid:liquid ratio of 1:10 for 20 h at 25 °C with an added 10 h at 65 °C incubation of remaining solids. The phlorotannin with the formula C12H10O7 (corresponding to bifuhalol) is the first identified in S. latissima.  相似文献
