CD99 plays a critical role in the diapedesis of monocytes, T cell differentiation, and the transport of MHC molecules. Engagement of CD99 by agonistic monoclonal antibodies has been reported to trigger multifactorial events including T cell activation as well as cell-cell adhesion during hematopoietic cell differentiation. In this study, to identify the functional domains participating in the cellular events, we mapped the epitopes of CD99, which are recognized by two agonistic CD99 monoclonal antibodies, DN16 and YG32. Using recombinant fusion proteins of GST with whole or parts of CD99, we found that both antibodies interact with CD99 molecules independently of sugar moieties. DN16 mAb detected a linear epitope located in the amino terminal region of CD99 while YG32 mAb bound another linear epitope in the center of the extracellular domain. To confirm that the identified epitopes of CD99 are actually recognized by the two mAbs, we showed the presence of physical interaction between the mAbs and the fusion proteins or synthetic peptides containing the corresponding epitopes using surface plasmon resonance analyses. The dissociation constants of DN16 and YG32 mAbs for the antigen were calculated as 1.27 x 10(-7) and 7.08 x 10(-9) M, respectively. These studies will help understand the functional domains and the subsequent signaling mechanism of CD99.  相似文献

Summary. Upper critical solution temperatures (UCSTs) for liquid–liquid demixing in a set of mixtures of linear alkanes (pentane (N 1=5) to pentacontane (N 1=50)) with an oligostyrene (1241amu, N 2=12) are reported. We find strong correlation between the Hildebrand solubility parameters of the alkanes and the UCST. Correlations are developed which enable predictions concerning the miscibility of mixtures of compounds with longer chains.  相似文献

Background

One of the signatures of Alzheimer's disease is the accumulation of aggregated amyloid protein, Aβ, in the brain. Aβ arises from cleavage of the Amyloid Precursor protein by β and γ secretases, which present attractive candidates for therapeutic targeting. Two β-secretase candidates, ASP-1 and ASP-2, were identified as aspartic proteases, both of which cleave the amyloid precursor at the β-site. These are produced as immature transmembrane proteins containing a pro-segment.

Results

ASP-2 expressed in HEK293-cells cleaved the Swedish mutant amyloid precursor at different β-sites at different pHs in vitro. Recent reports show that furin cleaves the pro-peptide of ASP-2, whereas ASP-1 undergoes auto-catalysis. We show that purified recombinant ASP-2 cleaves its own pro-peptide at ph 5 but not pH 8.5 as seen by mass spectrometry, electrophoresis and N-terminal sequencing.

Conclusion

We suggest that ASP-2 processing as well as activity are influenced by pH, and hence the cellular localisation of the protein may have profound effects on the production of Aβ. These factors should be taken into consideration in the design of potential inhibitors for these enzymes.  相似文献

Zusammenfassung Ein zweites spektrochemisches Verfahren zur Bestimmung von Silber, Blei, Zinn, Wismut, Tellur, Arsen und Antimon in Eisen und Stahl wurde entwickelt. Auch hier wird die spektrographische Analyse nach Auflösen der Probe in Salpetersäure und Eindampfen der Lösung zur Trockne durchgeführt. Die leichtflüchtigen Elemente werden ohne Puffer durch Anwendung eines inhomogenen Magnetfeldes und einer zweckmäßigen Elektrodenform störungsfrei und mit guter Reproduzierbarkeit bestimmt. Dabei sind die unteren Bestünmungsgrenzen gleich oder niedriger als die anderer spektrochemischer Methoden, die bisher beschrieben wurden. Das Verfahren zeichnet sich durch Schnelligkeit und Einfachheit aus.

A second spectrochemical method for the determination of silver, lead, tin, bismuth, tellurium, arsenic and antimony in iron and steel has been developed. Here also the spectrographic analysis is to be carried out after dissolution of the sample in nitric acid and evaporation of the solution to dryness. Without buffer, the easily volatile elements are determined free of interferences and with good reproducibility by using an inhomogeneous magnetic field and electrodes of an appropriate form. The lower limits of determination are the same or lower than those obtained by any other spectrochemical method described. The method is characterised by rapidity and simplicity of procedure.

The method of similarity analysis in the study of differential equations is extended to study the variability of parameters in a physical system. The analysis provides an insight into the meaning of physical similarity, which usually means the possibility of a reduction in the number of physical parameters characterizing the system. This paper reports the effect of annealing on conduction mechanisms in amorphous silicon films. High field transport is mainly by the Poole-Frenkel mechanism in unannealed films of amorphous silicon. As they are annealed, the conduction meachnism changes from Poole-Frenkel to space charge conduction. The films are fully annealed at 120°C for 15 h and any more annealing for longer periods does not change the IV characteristics. From the measured values of threshold voltage at which space charge conduction sets in, the density of localized states around the Fermi level is calculated to be about 1017–1018/eV · cm3 which agrees very well with our results from other experimental techniques.  相似文献

COVID-19, resulting from infection by the SARS-CoV-2 virus, caused a contagious pandemic. Even with the current vaccines, there is still an urgent need to develop effective pharmacological treatments against this deadly disease. Here, we show that the water and ethanol extracts of the root and rhizome of Polygonum cuspidatum (Polygoni Cuspidati Rhizoma et Radix), a common Chinese herbal medicine, blocked the entry of wild-type and the omicron variant of the SARS-CoV-2 pseudotyped virus into fibroblasts or zebrafish larvae, with IC50 values ranging from 0.015 to 0.04 mg/mL. The extracts were shown to inhibit various aspects of the pseudovirus entry, including the interaction between the spike protein (S-protein) and the angiotensin-converting enzyme II (ACE2) receptor, and the 3CL protease activity. Out of the chemical compounds tested in this report, gallic acid, a phytochemical in P. cuspidatum, was shown to have a significant anti-viral effect. Therefore, this might be responsible, at least in part, for the anti-viral efficacy of the herbal extract. Together, our data suggest that the extracts of P. cuspidatum inhibit the entry of wild-type and the omicron variant of SARS-CoV-2, and so they could be considered as potent treatments against COVID-19.  相似文献

The way cells communicate is not fully understood. However, it is well-known that extracellular vesicles (EVs) are involved. Researchers initially thought that EVs were used by cells to remove cellular waste. It is now clear that EVs function as signaling molecules released by cells to communicate with one another, carrying a cargo representing the mother cell. Furthermore, these EVs can be found in all biological fluids, making them the perfect non-invasive diagnostic tool, as their cargo causes functional changes in the cells upon receiving, unlike synthetic drug carriers. EVs last longer in circulation and instigate minor immune responses, making them the perfect drug carrier. This review sheds light on the latest development in EVs isolation, characterization and, application as therapeutic cargo, novel drug loading techniques, and diagnostic tools. We also address the advancement in plant-derived EVs, their characteristics, and applications; since plant-derived EVs only recently gained focus, we listed the latest findings. Although there is much more to learn about, EV is a wide field of research; what scientists have discovered so far is fascinating. This paper is suitable for those new to the field seeking to understand EVs and those already familiar with it but wanting to review the latest findings.  相似文献
