Polymerase Chain Reaction-based Detection of Total and Specific Vibrio Species

The polymerase chain reaction (PCR) technique is widely used for efficient detection of food-borne pathogens because of speed and specificity. However, PCR methods have focused mostly on species-specific detection. In the present work, we describe a PCR-based method for the simultaneous detection of all Vibrio species because lots of them are notorious food-borne human pathogens. We then combined this total detection method with specific detection of Vibrio cholerae pathogen. Using a degenerate primer set based on the sequence of the potassium uptake gene, trkA, we were able to successfully detect all Vibrio species. Specific detection of V. cholerae was also possible using primer sets based on putative flagellin sequence. Importantly, simultaneous total and species-specific Vibrio detection was possible using all two primer sets in a multiplexed PCR strategy. Thus, the PCR method we have developed is applicable to both simultaneous and two-step detection of total and specific Vibrio species.     

Dual signaling of m-chloroperbenzoic acid by desulfurization of thiocoumarin

The chemosignaling of widely used peracid oxidant of m-chloroperbenzoic acid (mCPBA) by the selective desulfurization of thiocoumarin was investigated. Thiocoumarin was efficiently converted into its corresponding coumarin by the reaction with mCPBA, and resulted in a pronounced fluorescent turn-on type signaling. The conversion also provided a significant change in absorption behavior which allowed a ratiometric analysis. The effective signaling could be used as a convenient determination method for mCPBA in aqueous environment.

Signature invariants of covering links

We apply the theory of signature invariants of links in rational homology spheres to covering links of homology boundary links. From patterns and Seifert matrices of homology boundary links, we derive an explicit formula to compute signature invariants of their covering links. Using the formula, we produce fused boundary links that are positive mutants of ribbon links but are not concordant to boundary links. We also show that for any finite collection of patterns, there are homology boundary links that are not concordant to any homology boundary links admitting a pattern in the collection.

     

Spectroscopic Observation of the Hydroxy Position in Butanol Hydrates and Its Effect on Hydrate Stability

In this study, we investigate the crystal structures and phase equilibria of butanols+CH₄+H₂O systems to reveal the hydroxy group positioning and its effects on hydrate stability. Four clathrate hydrates formed by structural butanol isomers are identified with powder X‐ray diffraction (PXRD). In addition, Raman spectroscopy is used to analyze the guest distributions and inclusion behaviors of large alcohol molecules in these hydrate systems. The existence of a free OH indicates that guest molecules can be captured in the large cages of structure II hydrates without any hydrogen‐bonding interactions between the hydroxy group of the guests and the water‐host framework. However, Raman spectra of the binary (1‐butanol+CH₄) hydrate do not show the free OH signal, indicating that there could be possible hydrogen‐bonding interactions between the guests and hosts. We also measure the four‐phase equilibrium conditions of the butanols+CH₄+H₂O systems.
