A composite polymer carbon fiber electrode modified with Nafion and cellulose acetate is described.The modified electrode discriminates against both anionic reactants and big molecular organic compounds.The bilayer configuration is prepared in two steps.First,the carbon fiber electrode is coated with Nafion,then followed by air evaporation of the solvent,the electrode is dipped in a cellulose acetate solution and hydrolyzed for a selected time.The permeability of the film is explored by use of rotating disk electrode measurements.Parameters affecting the film electrochemistry are investigated.The resulting electrodes show high selectivity and stability in body fluids.For in-vivo voltammetry,the composite polymer modified electrode has been used for detection of the oxidative current of neurotransmitter dopamine in rat brain,while it inhabits the oxidation of anionic neurotransmitter metabolites and some electroactive compounds.  相似文献

A joint approach combining free‐energy calculations and calcium‐imaging assays on the broadly tuned human 1G1 olfactory receptor is reported. The free energy of binding of ten odorants was computed by means of molecular‐dynamics simulations. This state function allows separating the experimentally determined eight agonists from the two non‐agonists. This study constitutes a proof‐of‐principle for the computational deorphanization of olfactory receptors.  相似文献

Recombinant human erythropoietin (rhEPO) has been extensively used as a pharmaceutical product for treating anemia. Glycosylation of rhEPO affects the biological activity, immunogenicity, pharmacokinetics, and in-vivo clearance rate of rhEPO. Characterization of the glycosylation status of rhEPO is of great importance for quality control. In this study, we established a fast and comprehensive approach for reliable characterization and relative quantitation of rhEPO glycosylation, which combines multiple-enzyme digestion, hydrophilic-interaction chromatography (HILIC) enrichment of glycopeptides, and tandem mass spectrometry (MS) analysis. The N-linked and O-linked intact glycopeptides were analyzed with high-resolution and high-accuracy (HR–AM) mass spectrometry using an Orbitrap. In total, 74 intact glycopeptides from four glycosylation sites at N24, N38, N83, and O126 were identified, with the simultaneous determination of peptide sequences and glycoform compositions. The extracted ion chromatograms based on the HR–AM data enabled relative quantification of glycoforms. Our results could be extended to quality control of rhEPO or could help establish detection approaches for glycosylation of other proteins. Graphical Abstract

The one‐dimensional coordination polymer catena‐poly[diaqua(sulfato‐κO)copper(II)]‐μ2‐glycine‐κ2O:O′], [Cu(SO4)(C2H5NO2)(H2O)2]n, (I), was synthesized by slow evaporation under vacuum of a saturated aqueous equimolar mixture of copper(II) sulfate and glycine. On heating the same blue crystal of this complex to 435 K in an oven, its aspect changed to a very pale blue and crystal structure analysis indicated that it had transformed into the two‐dimensional coordination polymer poly[(μ2‐glycine‐κ2O:O′)(μ4‐sulfato‐κ4O:O′:O′′:O′′)copper(II)], [Cu(SO4)(C2H5NO2)]n, (II). In (I), the CuII cation has a pentacoordinate square‐pyramidal coordination environment. It is coordinated by two water molecules and two O atoms of bridging glycine carboxylate groups in the basal plane, and by a sulfate O atom in the apical position. In complex (II), the CuII cation has an octahedral coordination environment. It is coordinated by four sulfate O atoms, one of which bridges two CuII cations, and two O atoms of bridging glycine carboxylate groups. In the crystal structure of (I), the one‐dimensional polymers, extending along [001], are linked via N—H...O, O—H...O and bifurcated N—H...O,O hydrogen bonds, forming a three‐dimensional framework. In the crystal structure of (II), the two‐dimensional networks are linked via bifurcated N—H...O,O hydrogen bonds involving the sulfate O atoms, forming a three‐dimensional framework. In the crystal structures of both compounds, there are C—H...O hydrogen bonds present, which reinforce the three‐dimensional frameworks.  相似文献

The practical application of polyaniline-poly(sodium 4-styrenesulfonate) (PANI-PSS) composite particles has been held back by the low stability of their dispersed state in water. In this work, we present a general oxidation approach to prepare PANI-PSS composite nanoparticles that can form highly stable colloids in water or buffer over a wide range of pH from 1 to 11. We demonstrate that the size of the PANI-PSS composite particles can be controlled by the acidity of precursor solutions. It is hypothesized that the number of negatively charged sites on PSS, which can be affected by the acidity of the precursor solutions, plays an important role in determining the size of the PANI-PSS composite particles and the stability of corresponding colloids in water.  相似文献

A series of azidoaryl- and azidoalkyl(diphenyl)oxazole scaffolds were warranted for biofilm inhibition studies. Cyclization of azidoaryl- or azidoalkyl esters of benzoin with ammonium acetate in acetic acid gives 2-azidoaryl- or 2-azidoalkyl-4,5-diphenyloxazoles. The azidoaryl esters are prepared from the corresponding azidocarboxylic acids/acid chlorides while the azidoalkyl esters are prepared from the corresponding haloalkyl esters.  相似文献

When pyrazolate ligands with thioether chelate arms are used in cyclic coinage metal pyrazolates [Au(μ-pz)](n), the inner gold ring can be framed with an outer silver ring to give novel heterometallic double-crowned complexes [AuAg(μ-L(x))(BF(4))](4). They feature short intramolecular in-plane Ag-Au interactions, are stable as octanuclear species in solution, and show promising luminescence properties.  相似文献

Xanthomonas campestris polysaccharide in the solid state is stable to 225°C in air and 250°C in inert atmosphere. In solution, even at moderate temperatures, the polymer undergoes hydrolytic degradation via the glycosidic linkages, and occurrence of main-chain scission results in lower solution viscosity. In solution, the polymer can exist in ordered and disordered conformations. In distilled water at temperatures ≤ 50°C, the polymer exists in the disordered conformation. In the presence of salt, acid, or base the polymer exists in the ordered conformation. In the ordered conformation the polymer exhibits a far greater hydrolytic stability. The higher stability of the ordered conformation is especially demonstrated when the polymer is aged in acid or base solutions. Contrary to the expected lower stability of the glycosidic linkages in acid or base than in water, Xanthomonas campestris polysaccharide shows higher stability in these media.  相似文献
