Carbohydrate antigens with subterminal fucosylation have been implicated in the development and progression of several cancers, including hepatocellular carcinoma (HCC). Fluorescent sensors targeting fucosylated carbohydrate antigens could potentially be used for diagnostic and other applications. We have designed and synthesized a series of 26 diboronic acid compounds as potential fluorescent sensors for such carbohydrates. Among these compounds, 7q was able to fluorescently label cells expressing high levels of sLex (HEPG2) within a concentration range of 0.5 to 10 microM. This compound (7q) did not label cells expressing Lewis Y (HEP3B), nor cells without fucosylated antigens (COS7). This represents the first example of a fluorescent compound labeling cells based on cell surface carbohydrate structures.  相似文献

The interaction between energetically asymmetric hydrophilic and hydrophobic surfaces has fundamental and practical importance in both industrial and natural colloidal systems. The interaction forces between a hydrophilic silica sphere and a silanated, hydrophobic glass plate in N-methyl-2-pyrrolidone (NMP)-water binary mixtures were measured using atomic force microscopy (AFM). A strong and long-range attractive force was observed in pure water and was attributed to the formation of capillary bridges associated with nanoscale bubbles initially present on the hydrophobic surface. When NMP was added, the capillary force and corresponding pull-off force became less attractive, which was explained readily in terms of the surface wettability by the binary solvent mixture. Similar to the case of symmetric (two hydrophilic) surfaces, the range of attraction between the asymmetric surfaces was maximized at around 30 vol % NMP, which is consistent with the formation of a thick adsorbed macrocluster layer on the hydrophilic silica surface.  相似文献

2,3‐Diaryl‐substituted maleic anhydrides were prepared by a modified one‐pot synthesis of Perkin condensation using mixed sodium salts of arylglyoxylic acid and arylacetic acid with acetic anhydride in 1,4‐dioxane. The treatment of these anhydrides with ammonium bicarbonate, or methanolic hydrazine, offered the corresponding 2,3‐diaryl‐substituted maleimides and maleic hydrazides (4,5‐diaryl‐substituted 1,2‐dihydropyridazine‐3,6‐dione), respectively. Evidence obtained from NMR, UV, and mass spectra suggest that 2,3‐diaryl‐substituted maleic hydrazides do not exhibit monolactim forms. Ring contraction of the diaryl‐substituted maleic hydrazide by nitrosation led to the formation of the corresponding maleimide. Interconversion between the corresponding maleic hydrazide and maleimide was observed following equilibrium reaction. Our experiment proposes that the chemistry of 2,3‐diaryl‐substituted maleic hydrazides rarely involves the function of ethylene moiety and resembles that of succinic hydrazine. J. Several investigators have emphasized the potential value of quantitative relaxation times in the assessment of diseases. In performing such measurements using the spin-echo technique, we have encountered several anomalous results, whereby the intensity of the organ parenchyma on second-echo images is greater than on first echo images. This is most likely a result of respiratory motion, and it occurs only rarely. Several volunteers were studied before and after exercise to see if respiratory motion could reproduce the anomalous intensity reverse; a reversal of intensities in renal parenchyma was observed in two of five individuals. We conclude that respiratory motion artifacts will seriously limit quantitative magnetic resonance imaging of the upper abdomen if respiratory gating during imaging is not used.  相似文献

The first total synthesis of the indole alkaloid mersicarpine is reported. Key steps include a beta-dicarbonyl radical cyclization, as well as an oxidation of the benzopyrrole moiety to establish the masked 1,2-dicarbonyl functionality. A piloted, partially premixed, liquid-fueled swirl burner is operated at high pressure (1 MPa). High-speed (6 KHz) stereoscopic PIV is used to investigate the characteristics of the stagnation line separating the pilot jet and the central recirculation zone (CRZ) with varying pilot-main ratio and global equivalence ratio. The mean curvature of the stagnation line displayed a large spatial scale pattern that was present for all operating conditions. All three components of velocity, in-plane shear, and swirling strength are conditioned upon the instantaneous stagnation line. Mean distributions of the velocity normal to the stagnation line show that velocity is oriented towards the CRZ when the stagnation line is found nearer the centerline of the combustor. The conditioned out-of-plane velocity (w) shows a distinct concentration of large mean and fluctuation RMS values towards the center of the measurement domain. Varying fuel flow does not significantly change this spatial structure, only the magnitudes of the w statistics. The in-plane shear stress was the largest for the pilot biased condition as a stronger shear layer develops. For the leanest flame, large fluctuation RMS values of shear stress were confined to a region where the pilot jet begins to interact more heavily with the main jet. Operating with less pilot fuel flow enhanced the mean conditional swirling strength indicating that the pilot shear layer was shedding more intense eddies. Disregarding spatial relations, a scatter plot of w, shear stress, and swirling strength displayed trends between the variables. The largest swirling strength values coincide with highest magnitude shear stresses and the widest range of w. These conditioned statistics highlight how certain aspects of the combustor flow field are invariant with fuel distribution. This is desirable for aeropropulsive combustors that must maintain stable ignition from a range of conditions from landing/take-off to cruise.  相似文献

LC-ion trap mass spectrometry was used to screen and confirm 38 compounds from a variety of drug classes in four species of fish: trout, salmon, catfish, and tilapia. Samples were extracted with acetonitrile and hexane. The acetonitrile phase was evaporated, redissolved in water and acetonitrile, and analyzed by gradient chromatography on a phenyl column. MS2 or MS3 spectra were monitored for each compound. Qualitative method performance was evaluated by the analysis over several days of replicate samples of control fish, fish fortified with a drug mixture at 1 ppm, 0.1 ppm and 0.01 ppm, and fish dosed with a representative from each drug class. Half of the 38 drugs were confirmed at 0.01 ppm, the lowest fortification level. This included all of the quinolones and fluoroquinolones, the macrolides, malachite green, and most of the imidazoles. Florfenicol amine, metronidazole, sulfonamides, tetracyclines, and most of the betalactams were confirmed at 0.1 ppm. Ivermectin and penicillin G were only detectable in the 1 ppm fortified samples. With the exception of amoxicillin, emamectin, metronidazole, and tylosin, residue presence was confirmed in all the dosed fish.  相似文献
