The optimal blends of six compounds that should be present in culture media used in recombinant protein production were determined by means of artificial neural networks (ANN) coupled with crossed mixture experimental design. This combination constitutes a novel approach to develop a medium for cultivating genetically engineered mammalian cells. The compounds were collected in two mixtures of three elements each, and the experimental space was determined by a crossed mixture design. Empirical data from 51 experimental units were used in a multiresponse analysis to train artificial neural networks which satisfy different requirements, in order to define two new culture media (Medium 1 and Medium 2) to be used in a continuous biopharmaceutical production process. These media were tested in a bioreactor to produce a recombinant protein in CHO cells. Remarkably, for both predicted media all responses satisfied the predefined goals pursued during the analysis, except in the case of the specific growth rate (μ) observed for Medium 1. ANN analysis proved to be a suitable methodology to be used when dealing with complex experimental designs, as frequently occurs in the optimization of production processes in the biotechnology area. An epoxy-based monolith has been developed for use as hydrophilic support in bioseparation. This monolith is produced by self-polymerization of polyglycerol-3-glycidyl ether in organic solvents as porogens at room temperature within 1 h. One receives a highly cross-linked structure that provides useful mechanical properties. The porosity and pore diameter can be controlled by varying the composition of the porogen. In this work, an epoxy-based monolith with a high porosity (79%) and large pore size (22 μm) is prepared and used in affinity capturing of bacterial cells. These features allow the passage of bacterial cells through the column. As affinity ligand polymyxin B is used, which allows the binding of gram-negative bacteria. The efficiency of the monolithic affinity column is studied with Escherichia coli spiked in water. Bacterial cells are concentrated on the column at pH 4 and eluted with a recovery of 97 ± 3% in 200 μL by changing the pH value without impairing viability of bacteria. The dynamic capacity for the monolithic column is nearly independent of the flow rate (4 × 109 cells/column). Thereby, it is possible to separate and enrich gram-negative bacterial cells, such as E. coli, with high flow rates (10 mL/min) and low back pressure (<1 bar) in a volume as low as 200 μL compatible for real-time polymerase chain reaction, microarray formats, and biosensors. 相似文献
The biosynthesis of serotonin requires aromatic substrates to be bound in the active sites of the enzymes tryptophan hydroxylase and aromatic amino acid decarboxylase. These aromatic substrates are held in place partially by dispersion and induction interactions with the enzymes' aromatic amino acid residues. Mutations that decrease substrate binding can result in a decrease in serotonin production and thus can lead to depression and related disorders. We use optimized crystal structures of these two enzymes to examine pair-wise electronic interaction energies between aromatic residues in the active sites and the aromatic ligands. We also perform in silico mutations on the aromatic residues to determine the change in interaction energies as mutations occur. Our second-order Moller-Plessett perturbation theory calculations show that drastic changes in interaction energy can occur and, in light of our previous work, we are able to use these data to offer predictions on the loss of protein function and on the possibility of disease upon mutation. We also examine local and gradient corrected density functional theory methods to evaluate their ability to predict these induction/dispersion-dominated interaction energies. We find that the hybrid B3LYP cannot model these interactions well, whereas the GGA HCTH407 offers largely qualitatively correct results, and the local functional SVWN quantitatively mimics the MP2 results rather well. 相似文献

Photon-correlation spectroscopy has been used to measure the diffusion coefficient D and first-mode intramolecular relaxation time τ1 of polystyrene in a theta solvent, cyclohexane at 34.5°C. Measurements were made on five narrow fractions (w = 2.88 × 106 to 9.35 × 106) as a function of concentration c, in the dilute regime. D varied linearly with c, D = Do (I + kDc), with Do = (1.4 ± 0.2) × 10?4 cm2 s?1. Although the values obtained for τ1 were reproducible to within 5%, no systematic variation with c was detected. The results are fitted by the relation τ1 = (7.7 ? 0.3) × 10?8 μs, which agrees well with the theoretical expression of Zimm for the non-draining bead-and-spring model, modified for the light-scattering case. 相似文献
Following a bottom-up approach to nanomaterials, we present a rational synthetic route from hexacyanometalates [M(CN)(6)](3-) (M=Cr(III), Co(III)) cores to well-defined heptanuclear complexes. By changing the nature of the metallic cations and using a localised orbital model it is possible to control and to tune the ground state spin value. Thus, with M=Cr(III), d(3), S=3/2, three heptanuclear species were built and characterised by mass spectrometry in solution, by single-crystal X-ray diffraction and by powder magnetic susceptibility measurements, [Cr(III)(CNbondM'L(n))(6)](9+) (M'=Cu(II), Ni(II), Mn(II), L(n)=polydentate ligand), showing spin ground states S(G)=9/2 [Cu(II)], with ferromagnetic interactions J(Cr,Cu)=+45 cm(-1), S(G)=15/2 [Ni(II)] and J(Cr,Ni)=+17.3 cm(-1), S(G)=27/2 [Mn(II)], with an antiferromagnetic interaction J(Cr,Mn)=-9 cm(-1), (interaction Hamiltonian H=-J(Cr,M) [S(Cr)Sigma(i)S(M)(i)], i=1-6). With M=Co(III), d(6), S=0, the heptanuclear analogues [Co(III)(CN-M'L(n))(6)](9+) (M'=Cu(II), Ni(II), Mn(II)) were similarly synthesised and studied. They present a singlet ground state and allow us to evaluate the weak antiferromagnetic coupling constant between two next-nearest neighbours M'-Co-M'. 相似文献
When the source of a tone moves with respect to a listener's ears, dichotic (or interaural) phase and amplitude modulations (PM and AM) are produced. Two experiments investigated the psychophysical characteristics of dichotic linear ramp modulations in phase and amplitude, and compared them with the psychophysics of diotic PM and AM. In experiment 1, subjects were substantially more sensitive to dichotic PM than diotic PM, but AM sensitivity was equivalent in the dichotic and diotic conditions. Thresholds for discriminating modulation direction were smaller than detection thresholds for dichotic AM, and both diotic AM and PM. Dichotic PM discrimination thresholds were similar to detection thresholds. In experiment 2, the effects of ramp duration were examined. Sensitivity to dichotic AM and PM, and diotic AM increased as duration was increased from 20 ms to 200 ms. The functions relating sensitivity to ramp duration differed across the stimuli; sensitivity to dichotic PM increased more rapidly than sensitivity to dichotic or diotic AM. This was also reflected in shorter time-constants and minimum integration times for dichotic PM detection. These findings support the hypothesis that the analysis of dichotic PM and AM rely on separate mechanisms. 相似文献
