A critical aspect to understanding the molecular basis of Alzheimer's disease (AD) is the characterization of the kinetics of interconversion between the different species present during amyloid-β protein (Aβ) aggregation. By monitoring hydrogen/deuterium exchange in Aβ fibrils using electrospray ionization mass spectrometry, we demonstrate that the Aβ molecules comprising the fibril continuously dissociate and reassociate, resulting in molecular recycling within the fibril population. Investigations on Aβ40 and Aβ42 amyloid fibrils reveal that molecules making up Aβ40 fibrils recycle to a much greater extent than those of Aβ42. By examining factors that could influence molecular recycling and by running simulations, we show that the rate constant for dissociation of molecules from the fibril (k(off)) is much greater for Aβ40 than that for Aβ42. Importantly, the k(off) values obtained for Aβ40 and Aβ42 reveal that recycling occurs on biologically relevant time scales. These results have implications for understanding the role of Aβ fibrils in neurotoxicity and for designing therapeutic strategies against AD.  相似文献

A valveless microdevice has been developed for the integration of solid phase extraction (SPE) and polymerase chain reaction (PCR) on a single chip for the short tandem repeat (STR) analysis of DNA from a biological sample. The device consists of two domains--a SPE domain filled with silica beads as a solid phase and a PCR domain with an ~500 nL reaction chamber. DNA from buccal swabs was purified and amplified using the integrated device and a full STR profile (16 loci) resulted. The 16 loci Identifiler? multiplex amplification was performed using a non-contact infrared (IR)-mediated PCR system built in-house, after syringe-driven SPE, providing an ~80-fold and 2.2-fold reduction in sample and reagent volumes consumed, respectively, as well as an ~5-fold reduction in the overall analysis time in comparison to conventional analysis. Results indicate that the SPE-PCR system can be used for many applications requiring genetic analysis, and the future addition of microchip electrophoresis (ME) to the system would allow for the complete processing of biological samples for forensic STR analysis on a single microdevice.  相似文献

Tetramisole promotes the catalytic asymmetric intramolecular Michael addition-lactonization of a variety of enone acids, giving carbo- and heterocyclic products with high diastereo- and enantiocontrol (up to 99:1 dr, up to 99% ee) that are readily derivatized to afford functionalized indene and dihydrobenzofuran carboxylates. Chiral isothioureas also promote the catalytic asymmetric intermolecular Michael addition-lactonization of arylacetic acids and α-keto-β,γ-unsaturated esters, giving anti-dihydropyranones with high diastereo- and enantiocontrol (up to 98:2 dr, up to 99% ee).  相似文献

An operational modification of Tafel analysis, applied to the intermediate region of square wave voltammetric curves, devoted to the identification of corrosion products on archaeological metal, is described. This is based on the voltammetry of microparticles methodology using conventional abrasive conditioning of the electrode, as well as ‘one‐touch’ and layer‐by‐layer techniques. The proposed methodology is applied to the identification of copper and silver corrosion products in mediaeval silver‐copper coins from the Libertad street hoard in Valencia (Spain).  相似文献

Organoselenium compounds have already been reported to be good anticarcinogenic candidates. A new selenoquinazoline derivative, 2,4-bis(selenomethyl)quinazoline (compound 1), has been synthesized, spectroscopically characterized and its crystal structure has been studied. An intermolecular coupling between C2 and \( {\text{H}}_{5}^{\prime } \) in the Heteronuclear Multiple Bond Correlation (HMBC) experiment has been observed. Assuming that the head-to-tail overlap of parallel molecules (as identified by X-ray diffraction) remains in solution to give bimolecular entities, the π–π interaction enables heteronuclear coupling between the former atoms with a three-bond distance [C2···(π–π)···\( {\text{C}}_{5}^{\prime } \)\( {\text{H}}_{5}^{\prime } \)]. The crystal structure of compound 1 has been solved by X-ray diffraction. It crystallizes in triclinic system, space group P?1. Unit cell parameters are a = 7.4969(7) Å, b = 8.7008(8) Å, c = 10.1666(9) Å, α = 110.215(2)°, β = 90.354(2)°, γ = 115.017(1)°. Linear chains in crystals of compound 1 are generated by C–H···Se and Se···Se bonds between molecules. Furthermore, head-to-tail overlap of parallel molecules, in which π–π interactions can occur, is observed. Compound 1 exhibited a cytotoxic effect in all of the evaluated tumoral cell lines and showed a higher cytotoxic effect in colon and breast cancer cell lines than etoposide, which was used as a reference compound.  相似文献

This work describes the performance of poly(methyl methacrylate) (PMMA) microfluidic DNA purification devices with embedded microfabricated posts, functionalized with chitosan. PMMA is attractive as a substrate for creating high surface area (SA) posts for DNA capture because X-ray lithography can be exploited for extremely reproducible fabrication of high SA structures. However, this advantage is offset by the delicate nature of the posts when attempting bonding to create a closed system, and by the challenge of functionalizing the PMMA surface with a group that invokes DNA binding. Methods are described for covalent functionalization of the post surfaces with chitosan that binds DNA in a pH-dependent manner, as well as for bonding methods that avoid damaging the underlying post structure. A number of geometric posts designs are explored, with the goal of identifying post structures that provide the requisite surface area without a concurrent rise in fluidic resistance that promotes device failure. Initial proof-of-principle is shown by recovery of prepurified human genomic DNA (hgDNA), with real-world utility illustrated by purifying hgDNA from whole blood and demonstrating it to be PCR-amplifiable.  相似文献

The catalytic activity and enantioselectivity in the kinetic resolution of (±)-1-naphthylethanol with a range of structurally related 3,4-dihydropyrimido[2,1-b]benzothiazole-based catalysts is examined. Of the isothiourea catalysts screened, (2S,3R)-2-phenyl-3-isopropyl substitution proved optimal, giving good levels of selectivity in the kinetic resolution of a number of secondary alcohols (S values up to >100 at ~50% conversion). Low catalyst loadings (0.10-0.25 mol%) of the optimal isothiourea can be used to generate enantiopure alcohols (>99% ee) in good yields.  相似文献

The double cyclocondensation of symmetric pyridyl bis(oxoacids) 2b and 3b with (R)-phenylglycinol stereoselectively gave access to bis-phenylglycinol-derived oxazolopyrrolidine 9 and oxazolopiperidone 10, respectively. Application of the stereocontrolled cyclocondensation reaction to phenyl bis-γ-oxoacid 4b provided 11, which was converted to the corresponding enantiopure di(pyrrolidinyl)benzene 22. The absolute configuration of the new stereogenic centers generated in the key cyclocondenstion step was unambiguously established by X-ray crystallographic analysis.  相似文献

Simple and effective multi-component one-pot aldol addition/protection reactions of β-ketoesters to a series of aldehydes in the presence Me(3)SiCl and i-Pr(2)EtN have been described. The analysis of the scope of the reaction revealed a dramatic dependence of the reactivity on the substrates used. Thus the effect of a catalytic amount of DMF and different reaction conditions was widely investigated. Further transformations of the aldol adducts were particularly useful to give valuable diols and compounds with quaternary stereocenters, while X-ray structural analysis gave also important stereochemical information about this challenging reaction.  相似文献

The reaction of [8,8-(PPh(3))(2)-nido-8,7-RhSB(9)H(10)] (1) with PR(3) in a 1:2 ratio affords mixtures that contain the mono-substituted bis-PR(3)-ligated rhodathiaboranes [8,8-(PPh(3))(L)-nido-8,7-RhSB(9)H(10)] [L = PMe(2)Ph (5), PMe(3) (6)] and the corresponding tris-PR(3)-ligated compounds [8,8,8-(L)(3)-nido-8,7-RhSB(9)H(10)] [L = PMe(2)Ph (7), PMe(3) (8)]. These latter species are more conveniently prepared from the reaction of 1 with three equivalents of the monodentate phosphines, PMe(2)Ph and PMe(3). Reaction between 1 and PMePh(2) in a 1:2 ratio yields the disubstituted rhodathiaborane [8,8-(PMePh(2))(2)-nido-8,7-RhSB(9)H(10)] (4), whereas the use of three equivalents of phosphine leads to the formation of B-ligated eleven-vertex [8,8,8-(PMePh(2))(2)(H)-nido-8,7-RhSB(9)H(9)-9-(PMePh(2))] (9). Compounds 4-9 have been characterized by NMR spectroscopy, and the structures of 8 and 9 confirmed by X-ray diffraction analyses. The characterization of the cluster compounds has been aided by the use of DFT calculations on some of the species. Variable-temperature NMR studies have demonstrated a lability of the PMePh(2) ligands in compounds 4 and 9, providing mechanistic insights about the ligand substitutional chemistry in these eleven-vertex rhodathiaboranes.  相似文献
