Chemotherapeutic drugs for cancer treatment have been traditionally originated by the isolation of natural products from different environmental niches, by chemical synthesis or by a combination of both approaches thus generating semisynthetic drugs. In the last years, a number of gene clusters from several antitumor biosynthetic pathways, mainly produced by actinomycetes and belonging to the polyketides family, are being characterized. Genetic manipulation of these antitumor biosynthetic pathways will offer in the near future an alternative for the generation of novel antitumor derivatives and thus complementing current methods for obtaining novel anticancer drugs. Novel antitumor derivatives have been produced by targetted gene disruption and heterologous expression of single (or a few) gene(s) in another hosts or by combining genes from different, but structurally related, biosynthetic pathways ("combinatorial biosynthesis"). These strategies take advantage from the "relaxed substrate specificity" that characterize secondary metabolism enzymes.  相似文献

Novel complexes of type M2LCl4·nH2O (M: Ni, n = 4; M: Cu, n = 2.5 and M: Zn, n = 1.5; L: ligand resulted from 1,3-phenylenediamine, 3,6-diazaoctane-1,8-diamine, and formaldehyde one-pot condensation) were synthesized and characterized. The ligand was also isolated and characterized. The complexes features have been assigned from microanalytical, electrospray ionization tandem mass spectrometry, IR, UV–vis, 1H NMR, and EPR spectra as well as magnetic data at room temperature. Simultaneous thermogravimetric/dynamic scanning calorimetry/evolved gas analysis measurements were performed to evidence the nature of the gaseous products formed in each step. Processes as water elimination, fragmentation, and oxidative degradation of the organic ligand as well as chloride elimination were observed during the thermal decomposition. The final product of decomposition was metal(II) oxide except for copper complex where CuCl remained also in the oxide network. The complexes exhibited an improved antibacterial activity in comparison with the ligand concerning both planktonic as well as biofilm-embedded cells.  相似文献

A selective and sensitive method based on the ammonium derivatisation with o -phthaldialdehyde (OPA) and N -acetyl-cysteine (NAC) has been developed for ammonium determination in real water samples. The proposed procedure has been compared with ammonium reference methods such as Nessler reagent method and ammonium selective electrode. All procedures have been chemometrically tested and compared in terms of the main analytical properties. These procedures have been used to determine ammonium in unknown water samples. The OPA-NAC reagent method does not present any systematic error (proportional or constant), while Nessler reagent presents both of them for some samples assayed. The ammonium selective electrode is free of corrigible systematic errors, however presents amine interference. The OPA-NAC ammonium method is able to achieve a detection limit (LOD) of 0.07 mg/L in the sample, with a linear dynamic range up to 1.4 mg/L of ammonium.  相似文献

In this study, a sequential extraction method using water and methanol to recover ethylene glycol dinitrate or nitroglycol (EGDN) contained in Goma-2 ECO dynamite was developed. After, an HPLC method was used for the determination of EGDN in the two extracted phases. The analytical method was validated by evaluating its selectivity, sensitivity, linearity, and linear working concentration range, limit of detection and quantitation, precision (as repeatability and intermediate precision), accuracy, and robustness, providing appropriate values (i.e. RSD values for precision about 6% and accuracy about 100%). Finally, the EGDN content of a sample of the Goma-2-ECO dynamite was determined obtaining a concentration of 30.29%, which is in accordance with the manufacturer's specifications for this dynamite (25.7-31.4%).  相似文献

Two new cyclopeptides, perthamides E and F were isolated from the polar extracts of the sponge Theonella swinhoei. The new structures, featuring an unprecedented β-amino acid unit (AHMOA), were determined by interpretation of NMR and MS data. The absolute configuration of the AHMOA residue was proposed on the basis of quantum chemical calculation of NMR chemical shifts. Perthamides were proved to inhibit TNF-α and IL-8 release in primary human keratinocytes cells and therefore could represent potentially leads for the treatment of psoriasis.  相似文献

The multicomponent Amine-Aldehyde-Dienophile reaction is optimized employing benzyl or 4-methoxybenzylamine. The interest of the transformation consist in the synthesis of polysubstituted cyclohex-2-enylamines. The study of the scope of this AAD process is carried out, as well as the diastereoselective version, employing commercially available chiral benzylic amines and a maleimide with the chiral information at the N-substituent. VCD spectroscopy is a very useful tool for the determination of the absolute configuration of the isolated enantiomerically enriched compounds.  相似文献

Quinones were firstly used as dipolarophiles in a photoclick 1,3-cycloaddition with 2,5-diaryltetrazoles, as photoactivatable predipoles, providing a novel and efficient access to three types of pyrazole-fused quinones (indazoledione derivatives). Distinctive features of this protocol include the use of light as the unique reagent and readily available, stable, and easy to handle starting materials and good to excellent yields. Photophysical and electrochemical properties of the quinones and their potential application as photoredox catalysts are also detailed.  相似文献

Poly(ethylene glycol) diacrylate (PEGDA) of different molecular weights (Mn = 575 and 700) was used as crosslinking agent for the photopolymerization of 2-hydroxyethyl methacrylate (HEMA) in order to obtain HEMA/PEGDA-based hydrogels. Composites were synthesized in situ employing a new methodology that implies the addition of different quantities of silver nitrate aqueous solution to the monomer mixture with the finality to obtain hydrogels with different silver nanoparticles’ spatial density and distribution. Samples were characterized by thermal, optical, spectroscopic and structural/morphological methods. Thermal studies showed that the increase of PEGDA molecular weight and the AgNO3 concentration in the reaction mixture enhance the glass transition temperature and the thermal stability of the composites. This behavior could be related to the silver coordination with the polymer network. Infrared spectroscopy with Fourier transform and Raman analyses were realized in order to corroborate the sample chemical structure by the identification of specific functional groups. Surface hydrogel morphology was visualized with scanning electron microscopy analysis, detecting a homogeneous micro-porous surface for the samples obtained from high molecular weight PEGDA. Presence of silver nanoparticles was established by X-ray fluorescence spectroscopy and UV/Vis methods. In this last case, the characteristic silver nanoparticle plasmon was observed. Using Transmission Electron Microscopy it was possible to visualize a homogeneous spatial distribution of spherical silver nanoparticles with very narrow diameter distribution that rounds about 14–21 ± 5 nm. In general, the silver nanoparticle presence in the compounds enhances considerably their thermal/morphological characteristics.  相似文献

The mono- and binuclear hydride compounds fac-[ReH(CO)3L] (1a) and [{ReH(CO)4}2(μ-L)] (1b) have been prepared by reaction of [ReH(CO)5] with Ph2PN(CH3)(CH2)2N(CH3)PPh2 (L) under UV light. Protonation reactions of the hydride compound 1a with equimolar amounts of HSO3CF3 or HCl yielded the triflato or the chlorido compounds fac-[Re(OSO2CF3)(CO)3L] (2) and fac-[ReCl(CO)3L] (3), respectively. The compounds have been characterised by elemental analysis, IR and NMR spectroscopic data, and mass spectrometry. Their structures have been confirmed by X-ray crystallography.  相似文献
