The extra chromosome 21 of Down syndrome (DS) or trisomy 21 patients contains an average of 40 extra copies of rRNA genes and the in vivo/in vitro regulation of the activity of these genes is not fully understood. The objective of this work was to compare the NORs expression pattern in interphase lymphocytes of DS patients with regular trisomy 21 and control individuals according to phytohemagglutinin (PHA) concentration (0.37, 0.75, 1.48 and 2.21 ml) per 100 ml of medium. Because the AgNOR staining is an indicator of the active rRNA genes, comparison of the image analysis values of the AgNOR area in 72 h cultivated lymphocytes for each concentration of PHA between DS patients (N=30) and controls (N=24) provided a plausible conclusion on the regulation of the extra rRNA genes in DS lymphocytes. The nucleolus organizer regions area/total nuclear area (NORa/TNa) was calculated using an in-house computer program. Fifty consecutive interphases per PHA concentration were analysed for each individual, for determination of the NORa/TNa. In contrast to healthy controls, NORa/TNa of lymphocytes from DS patient babies/children (0-8 years old) increased gradually in parallel with the PHA concentration in the culture medium: 10.44+/-1.72% for 0.37 ml of PHA, 11.74+/-1.93% for 0.75 ml of PHA, 13.25+/-2.03% for 1.48 ml of PHA and 13.43+/-2.08% for 2.21 ml of PHA per 100 ml of medium. Contrary to control cells (in which the NORa/TNa ratio according to PHA concentration in the culture medium remains constant), DS interphase lymphocytes in culture do not down-regulate their NOR expression. These results obtained from interphase NORs are consistent with the previous results obtained by evaluating the mean of AgNOR+ chromosome number in metaphase cells, also in relation to the mitogen concentration in the culture medium.  相似文献

The structural, elastic and thermodynamic characteristics of CeGa2 compound in the AlB2 (space group: P6/mmm) and the omega trigonal (space group: P-3m1) type structures are investigated using the methods of density functional theory within the generalized gradient approximation (GGA). The thermodynamic properties of the considered structures are obtained through the quasi-harmonic Debye model. The results on the basic physical parameters, such as the lattice constant, the bulk modulus, the pressure derivative of bulk modulus, the phase-transition pressure (P t ) from P6/mmm to P-3m1 structure, the second-order elastic constants, Zener anisotropy factor, Poisson’s ratio, Young’s modulus, and the isotropic shear modulus are presented. In order to gain further information, the pressure and temperature-dependent behavior of the volume, the bulk modulus, the thermal expansion coefficient, the heat capacity, the entropy, Debye temperature and Grüneisen parameter are also evaluated over a pressure range of 0–6 GPa and a wide temperature range of 0–1800 K. The obtained results are in agreement with the available experimental and the other theoretical values.  相似文献

Four new y-shaped fluorophores of 4- {4,5-[2,2'-Bis(2,4,6-trimethoxyphenyl)vinyl]-1-H-imidazole-2-yl}benzonitrile 1a, 2-phenyl-{4,5-[2,2'-Bis(2,4,6-trimethoxyphenyl)vinyl]-1-H-imidazole} 1b, 2- (9-anthryl)-{4,5-[2,2'-Bis(2,4,6-trimethoxyphenyl)vinyl] }-1-H-imidazole 1c and 2- (4-nitrophenyl) - {4,5-[2,2'-Bis(2,4,6-trimethoxyphenyl)vinyl] -1-H-imidazole 1d which bear an imidazole core, were synthesized for the first time via intermediate 1,6-Bis(2,4,6-trimethoxyphenyl)hexa-1,5-diene-3,4-dion with different aldehydes. The structures of the new derivatives were confirmed by (1)H NMR, (13)C NMR and FT-IR. The optical properties such as absorption and emission maxima, Stokes' shift and quantum yield values were investigated in solvents of toluene, tetrahydrofuran and acetonitrile. The products show intense emission maxima in the range of 440-630?nm. The imidazole derivatives exhibited excellent photostabilities.  相似文献

The objective of this work is to compare the NORs expression in interphase nuclei in non-stimulated lymphocytes of DS patients and healthy controls. Because the AgNOR staining is the indicator of the active rRNA genes, comparison of the image analysis values of AgNORs area between DS's and healthy controls' interphase lymphocytes is considered to be sufficient to evaluate the level of rDNA activities in the two groups. The Nucleolus Organizer Regions area/Total Nuclear area (NORa/TNa) was calculated using a computer program designed by us. 100 consecutive NORa/TNa per individual were evaluated. We report that 24 DS children's peripheral lymphocytes show significantly higher NORa/TNa mean value (6.32 +/- 1.77%) than that of the 20 healthy controls' cells (5.31 +/- 1.34%) (2-tailed Mann-Whitney U test, z = 19.4, P = 0.000). The same is true for the nucleolus (AgNOR spot) number per nucleus. The mean value of nucleoli number per nucleus in DS lymphocytes was significantly higher than that of the controls: z = 14.6, P = 0.000. In conclusion, extra rRNA genes on the chromosome 21 are not down-regulated in DS patients' lymphocytes. Rather, extra NORs expressions in 'in vivo' condition contribute to the increase of AgNORs area and AgNOR spots number per nucleus. This is the first work on the comparison of NORs activities in resting (non-stimulated) interphase lymphocytes between DS and healthy controls.  相似文献

Determination of organic vapor sensing properties of α-Naphthylmethacrylate (α-NMA) monomer based Langmuir-Blodgett (LB) thin films was aimed in this study. LB thin film fabrication was performed on quartz glass and quartz crystal substrates in order to investigate the characterization and organic vapor properties of α-NMA materials by using UV-Visible, Atomic Force Microscopy (AFM) and Quartz Crystal Microbalance (QCM) techniques. π-A isotherm graph was taken and a suitable surface pressure value were primarily determined as 13?mN m?1 for successful α-NMA LB thin film fabrication. Transfer ratio value was found to be ≥ 0.93 for quartz glass and quartz crystal substrates. The typical frequency shift per layer was obtained as 16.93?Hz/layer and the deposited mass onto a quartz crystal was calculated as 271.30?ng/layer (1.02?ng mm?2). The sensing responses of α-NMA LB films against dichloromethane, chloroform, toluene and m-xylene were measured by QCM system. Dichloromethane created the maximum shift in the resonance frequency than other organic vapors used in this study. Results exhibited that α-NMA LB thin films were potential candidates for organic vapor sensing applications, especially high sensitive detection of dichloromethane at room temperature.  相似文献

Two new Schiff base-hydrazones bearing furan ring, (Z)-4-butoxy-N′-(furan-2-ylmethylene)benzohydrazide (IV) and (Z)-N′-(furan-2-ylmethylene)-4-(hexyloxy)benzohydrazide (V), as well as their Cu(II), Ni(II), and Zn(II) complexes have been synthesised and characterised. The DNA-binding and DNA-cleavage activities of both aroylhydrazone ligands and their transition metal complexes were examined using UV-VIS titration and agarose gel electrophoresis in the presence of an oxidative agent (H2O2). The results indicate that the copper complexes bind significantly to calf thymus DNA and effectively cleave pBR322 DNA whereas the nickel and zinc complexes interact slightly with DNA.  相似文献

Three novel donor–π-bridge–donor (D -π-D) hole-transporting materials (HTMs) featuring triazatruxene electron-donating units bridged by different 3,4-ethylenedioxythiophene (EDOT) π-conjugated linkers have been synthesized, characterized, and implemented in mesoporous perovskite solar cells (PSCs). The optoelectronic properties of the new dumbbell-shaped derivatives (DTTXs) are highly influenced by the chemical structure of the EDOT-based linker. Red-shifted absorption and emission and a stronger donor ability were observed in passing from DTTX-1 to DTTX-2 due to the extended π-conjugation. DTTX-3 featured an intramolecular charge transfer between the external triazatruxene units and the azomethine–EDOT central scaffold, resulting in a more pronounced redshift. The three new derivatives have been tested in combination with the state-of-the-art triple-cation perovskite [(FAPbI3)0.87(MAPbBr3)0.13]0.92[CsPbI3]0.08 in standard mesoporous PSCs. Remarkable power conversion efficiencies of 17.48 % and 18.30 % were measured for DTTX-1 and DTTX-2 , respectively, close to that measured for the benchmarking HTM spiro-OMeTAD (18.92 %), under 100 mA cm−2 AM 1.5G solar illumination. PSCs with DTTX-3 reached a PCE value of 12.68 %, which is attributed to the poorer film formation in comparison to DTTX-1 and DTTX-2 . These PCE values are in perfect agreement with the conductivity and hole mobility values determined for the new compounds and spiro-OMeTAD. Steady-state photoluminescence further confirmed the potential of DTTX-1 and DTTX-2 for hole-transport applications as an alternative to spiro-OMeTAD.  相似文献

This paper is concerned with the isogeometric analysis (IGA) of composite laminates under cylindrical bending. Non-uniform rational B-splines (NURBS) are employed as basis functions for both geometric and computational implementations. In order to account for multiple domains, each lamina is modeled as a single NURBS patch. This multipatch representation corresponds to decomposition of the computational domain (composite laminate) into non-overlapping subdomains. As NURBS patches are discontinuous across their boundaries, a standard FEA-like procedure does not work for multipatch IGA; an additional numerical technique is required for coupling NURBS patches. Therefore, in this paper, one of the discontinuous Galerkin (DG) methods, namely symmetric interior penalty Galerkin formulation, is employed to allow for discontinuities. For numerical calculations, a composite laminate with stacking sequences $$0^{\circ }{/}90^{\circ }$$ and $$0^{\circ }{/}90^{\circ }{/}0^{\circ }$$, respectively, is adopted. The stresses are calculated along the thickness of the composite laminate, subjected to a sinusoidal load, and they are compared with the analytical solutions. It is shown that DG–IGA gives a better approximation in comparison with the standard IGA.  相似文献

Type II photoinitiated self‐condensing vinyl polymerization for the preparation of hyperbranched polymers is explored using 2‐hydroxyethyl methacrylate (HEMA) or 2‐(dimethylamino)ethyl methacrylate (DMAEMA), and methyl methacrylate as hydrogen donating inimers and comonomer, respectively, in the presence of benzophenone and camphorquinone under UV and visible light. Upon irradiation at the corresponding wavelength, the excited photoinitiator abstracts hydrogen from HEMA or DMAEMA leading to the formation of initiating radicals. Depending on the concentration of inimers, type of the photoinitiator, and irradiation time, hyperbranched polymers with different branching densities and cross‐linked polymers are formed.

The reliable generation of quasi-homogeneous autoignition inside a combustor fed by a continuous air flow would represent a milestone in realizing pressure gain combustion in gas turbines. In this work, the ignition distribution inside a stratified fuel–air mixture is analyzed. The ability of precise and reproducible injection of a desired fuel profile inside a convecting air flow is verified by applying tunable diode laser absorption spectroscopy in non-reacting measurements. High-speed, static pressure sensors and ionization probes allow for simultaneous detection of the flame and pressure rise at several axial positions in reactive measurements with dimethyl ether as fuel. A second, exchangeable combustion tube enables optical observation of OH* intensity in combination with pressure measurements. Experiments with three arbitrary fuel profiles show a set of ignition distributions that vary in shape, homogeneity, and the number of simultaneous autoignition events. Although the measurements show notable variation, a significant and reproducible influence of the fuel injection on the ignition distribution is observed. Results show that uniform autoignition leads to a coupling of the reaction front with the pressure rise and, therefore, induces a greater aerodynamic constraint than non-uniform ignition distributions, which are dominated by propagating deflagration fronts.  相似文献
