The electrochemical behavior of hemin, an iron complex of porphyrin, on binding to DNA at a glassy carbon electrode (GCE) and in solution, is described. Hemin, which interacts with covalently immobilized calf thymus DNA, was detected by use of a bare GCE, a double-stranded DNA-modified GCE (dsDNA-modified GCE), and a single-stranded DNA-modified GCE (ssDNA-modified GCE), in combination with differential pulse voltammetry (DPV). The structural conformation of DNA was determined from changes in the voltammetric signals acquired on reduction of hemin. As a result of its large steric structure and anionic substitution on its porphyrin plane, hemin intercalates between the base pairs of dsDNA. A scan-rate study for hemin and the dsDNA-hemin complex were also performed to determine the electrochemical behavior of the complex. The partition coefficient was obtained from the peak currents measured when different concentrations of hemin were in the presence of dsDNA. By observing the oxidation signals of guanine, damage to DNA after reaction with hemin at the GCE surface was also detected. The electrochemical detection of hybridization between the covalently immobilized probe and its target sequence was detected by use of hemin. These results demonstrate the use of DNA biosensors in conjunction with hemin for electrochemical detection of hybridization and damage to DNA.  相似文献

DNA biosensors based on nucleic acid hybridization processes are rapidly being developed towards the goal of rapid and inexpensive diagnosis of genetic and infectious diseases. Electrochemical transducers are often being used for detecting the DNA hybridization event, due to their high sensitivity, small dimensions, low cost, and compatibility with microfabrication technology. In this study, an electrochemical biosensor for the voltammetric detection of DNA sequences related to the Hepatitis B virus (HBV) and TT virus (TTV) from polymerase chain reaction (PCR) amplified real samples is described for the first time. The biosensor relies on the immobilization of the 21- or 24-mer single stranded oligonucleotides (probe) related to the HBV and TTV sequences and hybridization of these oligonucleotides with their complementary sequences (target) at carbon paste electrode (CPE). The extent of hybridization between the probe and target sequences was determined by using square wave voltammetry (SWV) with moving average baseline correction and methylene blue (MB) as the hybridization indicator. As a result of the interaction between MB and the bound guanine bases of hybrid at CPE surface, the MB signal decreased, when it was compared with the MB signal, which was observed with probe modified CPE. The difference between the MB signals, obtained from the hybrid modified and the probe modified CPE is used to detect the DNA sequences of the infectious diseases from PCR amplified real samples. Numerous factors affecting the target hybridization and indicator binding reactions are optimized to maximize the sensitivity.  相似文献

Teucrium chamaedrys L. is an aromatic and medicinal plant used as traditional medicine. Aerial parts of the plant material were dried and extracted with hexane–dichloromethane (extract 1), ethyl acetate–dichloromethane (extract 2) and methanol–dichloromethane (extract 3) in a ratio of 1:1 at rt successively. The solvents were evaporated to give crude extracts. Extract 1 was suspended in water at 60°C then partitioned successively with hexane and ethyl acetate to give hexane and ethyl acetate portions. After the column chromatography (silica gel) of ethyl acetate extract, one new and four known compounds were isolated. The new compound was named as 1(12S,18R)-15,16-epoxy-2β,6β-dihydroxy-neo-cleroda-13(16),14-dien-20,l2-olide-l8,l9-hemiacetal (teuchamaedryn D) (4). The known compounds were teucrin A (1), dihydroteugin (2), teucroxide (3), syspirensin A (5). The chromatographic methods were also applied for extract 3 to isolate verbascoside (6) and teucrioside (7). The structure of isolated compounds was elucidated by spectroscopic methods including LC-TOF/MS, 1D NMR and 2D NMR.  相似文献

Adsorptive stripping differential pulse voltammetry (AdSDPV) was applied to the assay of sorafenib in human serum sample. Cyclic voltammetry at a carbon based screen printed electrode (SPE) permitted to detect the irreversible oxidation of SOR with formation of a new compound reversibly oxidized at a lower potential. Quantitative assays were realized using a chitosan/carboxylic acid functionalized multiwalled carbon nanotube modified glassy carbon electrode in 0.1 M phosphate buffer pH 7.0 in the presence of 50 % methanol. The AdSDPV method provided two linear responses within the concentration ranges 1×10?8–8×10?8 M and 1×10?7–8×10?7 M in serum with LOQ and LOD of 3.2×10?9 and 9.6×10?10 of lower linear range, respectively. The recovery of sorafenib in spiked serum was 97.5 %.  相似文献

DNA hypermethylation is an epigenetic alteration and a promising biomarker for early prostate cancer detection. Simple, sensitive, easy to handle and rapid detection methodologies are imperative for point of care diagnostics especially for cancer. Herein, we describe for the first time a regenerable and compatible electrochemical biosensor for detection of Glutathione S‐Transferase P‐1 (GSTP‐1) gene hypermethylation related to prostate cancer via DNA hybridization onto the disposable Carbon and Multi Walled Carbon Nanotubes (MWCNT) Screen Printed Electrodes (SPEs). In the study, capture probes were adsorbed onto the SPEs by simple passive adsorption and then hybridization was achieved by sending the complementary target onto the probe‐modified electrodes. The selectivity of the biosensor was proved by control studies. Differential Pulse Voltammetry (DPV) technique was used to detect hybridization via guanine oxidation signals changes. The total time of the optimized method was nearly 1h, measurements took for less than 1 min, and the biosensor response was stable up to 40 days of storage period at 4 °C. The main advantages of the biosensor are very low detection limit (picomolar range) and capability of reusing the biosensor for at least 3 times after very simple regeneration process that is a unique property to reduce the cost of the assay. In addition, this is the first study that demonstrates the detection of GSTP‐1 hypermethylation electrochemically by using SPEs in order to create point of care diagnostics. The optimum parameters for the biosensor, as well as its future prospects to enhance the performance of DNA biosensors were also presented.  相似文献

The hitherto unknown complexes, [M2(CO)6(μ-CO)(μ-L)], [M = Cr; 1, Mo; 2, W; 3] and [M2(CO)6(μ-CO)(μ-L′)], [M = Cr; 4, Mo; 5, W; 6] have been synthesized by the photochemical reactions of photogenerated intermediate, M(CO)5THF (M = Cr, Mo, W) with thio Schiff base ligands, N,N′-bis(2-aminothiophenol)-1,4-bis(2-carboxaldehydephenoxy)butane (H 2 L) and N,N′-bis(2-aminothiophenol)-1,7-bis(2-formylphenyl)-1,4,7-trioxaheptane (H 2 L′). The complexes have been characterized by elemental analysis, LC-mass spectrometry, magnetic studies, FT-IR and 1H NMR spectroscopy. The spectroscopic studies show that H 2 L and H 2 L′ ligands are converted to benzothiazole derivatives, L and L′ after UV irradiation and coordinated to the central metal as bridging ligands via the central azomethine nitrogen and sulphur atoms in 1–6.  相似文献

A novel tetradentate azo‐Schiff base ligand (H2L) was synthesized by 2:1 molar condensation of an azo‐aldehyde and ethylenediamine. Its mononuclear Cu(II), Ni(II), Co(II) and Zn(II) complexes were prepared and their structures were confirmed using elemental analysis, NMR, infrared and UV–visible spectroscopies and molar conductivity measurements. The results suggest that the metal ion is bonded to the tetradentate ligand through phenolic oxygens and imine nitrogens of the ligand. The solid‐state structures of the azo‐Schiff base ligand and its Cu(II) complex were determined using single‐crystal X‐ray diffraction studies. The azo‐Schiff base ligand lies on a crystallographic inversion centre and thus the asymmetric unit contains half of the molecule. X‐ray data revealed that keto–amine tautomer is favoured in the solid‐state structure of the ligand. In the structure of the Cu(II) complex, the Cu(II) ion is coordinated to two phenolate oxygen atoms and two imine nitrogen atoms of the azo‐Schiff base ligand with approximate square planar geometry. The anticancer activity of the synthesized complexes was investigated for human cancer cell line (MCF‐7) and cytotoxicity of the synthesized compounds was determined against mouse fibroblast cells (L929). The ligand and its complexes were found to show antitumor activity. The synthesized metal complexes were optimized at the B3LYP/LANL2DZ level and a new theoretical formula for MCF‐7 cells was also derived.  相似文献

The electrochemical behavior of atorvastatin and amlodipine at a glassy carbon electrode has been studied using different voltammetric techniques. First derivative of the ratio voltammetric methods for determination of amlodipine and atorvastatin in tablets in the presence of the other compound has been described. This technique depends on the measuring of first derivative of the ratio voltammograms of each concentration as a function of the increased concentrations. DP and SW voltammetric methods depend on first derivative of the ratio‐voltammetry by measurements of the selected potentials for amlodipine and atorvastatin. The linear response was within the range of 4×10?6–1×10?4 M for amlodipine and 2×10?6–1×10?4 M for atorvastatin. The proposed methods have been extensively validated.  相似文献

Propane steam reforming was studied over Ni–Al2O3 catalysts that were prepared by a conventional impregnation (IM) method and a one-step sol–gel (SG) technique. Both Ni–Al2O3 catalysts showed similar initial activity. However, IM-Ni–Al2O3 deactivated severely with time-on-stream of propane steam reforming. The catalyst prepared using a SG technique demonstrated stable catalytic performance. The two catalysts also showed major differences in product distribution, with SG catalyst giving much higher yields of hydrogen. Catalysts were characterized with temperature-programmed reduction (TPR), X-ray photoelectron spectroscopy (XPS), temperature-programmed oxidation (TPO), transmission electron microscopy (TEM), X-ray diffraction (XRD) and Raman spectroscopy. It was revealed that, with sol–gel preparation, highly dispersed small Ni crystallites are formed with a strong interaction with the support. This is shown to be important for coke suppression and catalyst stability.  相似文献
