Three copper(II) complexes containing beta-cyclodextrin (betaCD) derivatives as ligands [mono-6-methylamino-6-deoxy-betaCD dithiocarbamate (CDTC), mono-6-histamino-6-deoxy-betaCD (CDHM) and mono-6-Nalpha-arginyl-6-deoxy-betaCD (CDARG)] have been studied by electron paramagnetic resonance. The spectra have been recorded at X- and S-bands and different temperatures and simulated to obtain the best set of magnetic parameters. In particular, the 300 K spectra are typical of the slow motion regime, as expected considering the high molecular weight of the ligands. Some structural characteristics of the complexes are proposed on the basis of dynamic and covalency parameters obtained from simulation.  相似文献

The aim of this study was to investigate the relationship between proteoglycans (PGs) and collagen fibrils at the early mineralization process of mantle dentin. Ten first molar dental germs of rats were removed and fixed in glutaraldehyde/formaldehyde in cacodylate buffer and post-fixed in osmium tetroxide. The samples were dehydrated and embedded in epoxy resin. Ultrathin sections were contrasted and analyzed in TEM before and after treatment with EDTA, chondroitinases AC and ABC. After EDTA treatment, a electrondense substance associated with collagen fibril was removed, and did not stain again. A high magnification of these areas showed globular structures with 15 nm diameter surrounding collagen fibrils. In advanced mineralization areas, collagen fibrils showed a banded pattern and at high magnification the fibrils presented a light 10 nm ring inside and a dark 10 nm ring outside. After chondroitinase treatment, the electrondense substance associated with collagen fibrils was removed, showing a banded pattern of clear and dark areas along them. From morphological data, the authors proposed a model of interaction between PGs and collagen fibrils, where glicosaminoglycans chains are inside the fibrils, while the protein core remains outside. That stereochemical arrangement would start the crystal nucleation.  相似文献

MP2 and CCSD(T) ab initio calculations have been carried out to elucidate geometrical structure and vibrational frequencies of representative lanthanide trihalides LnX(3) (Ln = La, Lu; X = F, Cl) explicitly including temperature, anharmonic, inert-gas matrix, and spin-orbit effects. The results have been compared with gas-phase electron diffraction, gas-phase IR measurements, and IR spectra of molecules trapped in inert-gas matrices. On the Born-Oppenheimer surface LaCl(3), LuF(3), and LuCl(3) adopt trigonal planar (D(3)(h)()) geometry while LaF(3) assumes a slightly pyramidal (C(3)(v)()) structure. Because of normal-mode anharmonicities, the resulting thermal average bond angles are considerably lower than the equilibrium ones, while vibrationally averaged bond lengths are predicted to be longer. The inert-gas matrix effects, modeled by the coordination of two inert-gas molecules LnX(3).IG(2) (IG = Ne, Ar, Xe, and N(2)), are substantial and strongly depend on the polarizability of coordinating particles. Coordinating inert-gas units always favor the tendency of LnX(3) molecules to adopt planar structure and induce noticeable frequency shifts.  相似文献

Mechanism of Sintering of Highly Dispersed Pt on Al2O3 in Oxygen Sintering of highly dispersed Pt on alumina in O2 consists in a slow thermal decomposition of a PtIV surface compound, followed by a fast coalescence of the highly mobile, zerovalent Pt, formed during the decomposition. Sintering according to this mechanism is inhibited by O2. The slower Ostwald ripening, promoted by O2, comes into effect only after complete decomposition of the PtIV surface compound.  相似文献

The efficient and rapid detection of bioactive compounds in complex matrices of different origins (natural or synthetic) is a key step in the discovery of molecules with potential application in therapy. Among them, molecules able to interact with nucleic acids can represent important targets.In this study, an optical DNA biosensor, based on surface plasmon resonance (SPR) transduction, has been studied in its potential application as new analytical device for drug screening. This device was applied to the analysis of pure synthetic or natural molecules and also to some fractions obtained by chromatographic separation of an extract of Chelidonium majus L. (great celandine), a plant containing benzo[c]phenanthridinium alkaloids having intercalating properties.The ability of these molecules to interact with the double stranded nucleic acid (dsDNA) immobilised on the sensor surface has been investigated. The optical sensing relies on the SPR-based bench instrument Biacore X™ and represents an example of multiuse sensor. The results obtained demonstrate the potential application of this device for the rapid screening of bioeffective compounds. The characteristics of the biosensor offer the possibility to be coupled to chemical analysis as in hyphenated technologies.  相似文献

The three novel pyridine‐containing 12‐membered macrocyclic ligands 1 – 3 were synthesized. The coordinating arms are represented by three acetate moieties in 1 and 3 and by one acetate and two phosphonate moieties in 2 . In all three ligands, the acetate arm in the distal position is substituted, with a benzyl group in 1 and 2 and with an arylmethyl moiety in 3 . The relaxivities r1p (20 MHz, 25°) of GdIII complexes are: GD?1 , r1p=8.3 mM ?1 s?1; GD?2 , r1p8.1 mM ?1 s?1; Gd?3 , r1p10.5 mM ?1 s?1. 1H‐NMRD and 17O‐NMR T2 data show that Gd?1 and Gd?3 contain two H2O molecules in the inner sphere, whereas the presence of two phosphonate arms allows the coordination of only one H2O molecule in Gd?2 . Interestingly, the exchange lifetime of coordinated H2O in the three complexes is similar in spite of the difference in the coordination number of the GdIII ion (i.e., 9 in Gd?1 and Gd?3 , and 8 in Gd?2 ). 1H‐Relaxometric measurements at different pH and in the presence of lactate and oxalate were carried out to get some insight into the formation of ternary complexes from Gd?1 and Gd?3 . Finally, it was found that binding to human‐serum albumin (HSA) of Gd?1 and Gd?2 , though weak, yields limited relaxivity enhancements, likely as a consequence of effects on the hydration sphere caused by donor atoms on the surface of the protein.  相似文献

The present work aimed to characterize the molecular relationships between structure and function of the seed storage protein β-vignin, the vicilin storage protein of cowpea (Vigna unguiculata, l. Walp) seeds. The molecular characterization of β-vignin was carried out firstly by assessing its thermal stability, under different conditions of pH and ionic strength, by thermal shift assay (TSA) using SYPRO Orange fluorescent dye. Secondly, its aggregation propensity was evaluated using a combination of chromatographic and electrophoretic techniques. Two forms of β-vignin were considered: the native form purified from mature quiescent seeds, and a stable breakdown intermediate of 27 kDa produced while seeds germinate. TSA is a useful tool for determining and following over time the structural changes that occur to the protein during germination. The main result was the molecular characterization of the 27 kDa intermediate breakdown polypeptide, which, to the best of our knowledge, has never been described before. β-vignin seems to retain its trimeric conformation despite the evident degradation of its polypeptides.  相似文献
