Summary A flow-injection manifold with mixing chamber has been developed for the determination of phosphate by reaction with molybdate and crystal violet. The insoluble blue dye salt is kept in colloidal solution with poly(vinyl alcohol) and measured at 560 nm. The injection rate is 100 h–1. The detection limit is 0.01 g PO 4 3– /ml based on 250 l injection volumes. The system has been applied to the determination of phosphate in serum samples and after appropriate mineralisation to organic compounds and to plant materials.  相似文献

We demonstrate a versatile microfabricated electrophoresis platform, incorporating arrays of integrated on-chip electrodes, heaters, and temperature sensors. This design allows a range of different sieving gels to be used within the same device to perform separations involving both single- and double-stranded DNA over distances on the order of 1 cm. We use this device to compare linear and cross-linked polyacrylamide, agarose, and thermo-reversible Pluronic-F127 gels on the basis of gel casting ease, reusability, and overall separation performance using a 100 base pair double-stranded DNA ladder as a standard sample. While cross-linked polyacrylamide matrices provide consistently high-quality separations in our system over a wide range of DNA fragment sizes, Pluronic gels also offer compelling advantages in terms of the ability to remove and reload the gel. Agarose gels offer good separation performance, however, additional care must be exercised to ensure consistent gel properties as a consequence of the need for elevated gel loading temperatures. We also demonstrate the use of denaturing cross-linked polyacrylamide gels at concentrations up to 19% to separate single-stranded DNA fragments ranging in size from 18 to 400 bases in length. Primers differing by 4 bases at a read length of 30 bases can be separated with a resolution of 0.9-1.0 in under 20 min. This level of performance is sufficient to conduct a variety of genotyping assays including the rapid detection of single nucleotide polymorphisms (SNPs) in a microfabricated platform. The ability to use a single microelectrophoresis system to satisfy a wide range of separation applications offers molecular biologists an unprecedented level of flexibility in a portable and inexpensive format.  相似文献

Dilute aqueous dispersions of colloidal polystyrene latex spheres were flocculated by adding a nonadsorbing polymer sample, poly(acrylic acid). The structural compactness of the flocs thus formed was characterized in terms of their mass fractal dimension using the small-angle static light scattering technique. It was found that with low poly(acrylic acid) concentrations and thus weak depletion attraction forces, the dispersion medium viscosity had a marked effect on the floc structure. An increase in the viscosity led to formation of denser flocs. This was revealed in three sets of depletion flocculation experiments: (a) adjusting the background electrolyte concentration at a fixed level of poly(acrylic acid), (b) using water and 30% (w/w) glycerol as the respective solvents, and (c) inducing latex flocculation with two poly(acrylic acids) of different molecular weights at the respective critical polyacid concentrations. Direct force measurements were made with atomic force microscopy to isolate the influence of viscosity on floc structure from that of interparticle interaction energies. We conclude that the formation of denser flocs with increasing medium viscosity can be attributed to the reduced diffusivity of particles in the solution. The latter resulted in an enhanced rate of floc restructuring (through relaxation of attached particles) relative to floc growth.  相似文献

We prove that a diffeomorphism possessing a homoclinic point with a topological crossing (possibly with infinite order contact) has positive topological entropy, along with an analogous statement for heteroclinic points. We apply these results to study area-preserving perturbations of area-preserving surface diffeomorphisms possessing homoclinic and double heteroclinic connections. In the heteroclinic case, the perturbed map can fail to have positive topological entropy only if the perturbation preserves the double heteroclinic connection or if it creates a homoclinic connection. In the homoclinic case, the perturbed map can fail to have positive topological entropy only if the perturbation preserves the connection. These results significantly simplify the application of the Poincaré-Arnold-Melnikov-Sotomayor method. The results apply even when the contraction and expansion at the fixed point is subexponential.The first author was partially supported by a Sloan Foundation Fellowship and an N.S.F. grant. Pyrrolidyl triazenes prepared by interception of the diazonium transient in the Sandmeyer reaction of amines can serve as useful intermediates in the iodination and radioiodination of aryl rings. Decomposition of such triazenes in the presence of iodide is acid-catalyzed and is sensitive to choice of solvent, acid, and triazene structure. A condition variation study by HPLC of four different solvent systems and two non-nucleophilic acids was carried out on the123I iodination of the triazenes of p-nitroaniline, p-anisidine and p-toluidine. This method has proven useful in radiolabeling of two pharmaceutical analogs which were not amenable to labeling through the classic Sandmeyer method.  相似文献
