An integrated microfluidic device capable of performing a variety of genetic assays has been developed as a step towards building systems for widespread dissemination. The device integrates fluidic and thermal components such as heaters, temperature sensors, and addressable valves to control two nanoliter reactors in series followed by an electrophoretic separation. This combination of components is suitable for a variety of genetic analyses. As an example, we have successfully identified sequence-specific hemagglutinin A subtype for the A/LA/1/87 strain of influenza virus. The device uses a compact design and mass production technologies, making it an attractive platform for a variety of widely disseminated applications. 相似文献

Topotecan is an antitumor agent with activity against a variety of cancers. We examined the steady-state and time-resolved fluorescence spectral properties of topotecan with one- and two-photon excitation. Topotecan was found to display a high two-photon cross section near 20 GM for wavelengths within the fundamental output of a Ti:sapphire laser, 800-880 nm. In frozen solution the anisotropies of topotecan are near the theoretical maxima for one-photon and two-photon excitation with colinear electronic transitions. The intensity and anisotropy decays of topotecan fluorescence were found to be homogeneous (single exponentials) in phosphate-buffered saline and propylene glycol. The steady-state and time-resolved data indicate that topotecan binds to a double-helical DNA oligomer d(AT)10 resulting in increased anisotropies and multiexponential intensity and anisotropy decays. Subnanosecond components in the anisotropy decay of the DNA-topotecan complex suggest loose binding of the drug to DNA. Loose binding of topotecan to DNA is also revealed by accessibility of topotecan to collisional quenching by iodide. 相似文献

The refined theory of the electroviscous lift forces is presented for the case when the separation distance between the particle and the wall is larger than the double-layer thickness. The theory is based on the lubrication approximation for motion of a long cylinder near a solid wall in creeping flow. The approximate analytical formula for the lift force valid for Pe=1 is derived and compared with the results of numerical calculations performed for an arbitrary Peclet number. The resulting electrokinetic lift force exceeds by several orders of magnitude one predicted by the previously developed theories of the lift force. The results for the electroviscous drag force indicate that when the double layer is thin compared with the particle size, the electroviscous drag is only a small correction (at most 10%) to the hydrodynamic drag force acting on a neutral particle moving near the wall. Microcalorimetric investigations have been carried out onthe interaction of the surfactants sodium cholate, sodium deoxycholate, tetradecyltrimethylammonium bromide, cetyl(hexadecyl)trimethylammonium bromide, and p-tert-octylphenoxy polyoxy-ethylene ether (Triton X-100) and the salts potassium iodide, sodium benzoate, sodium bromide, and sodium salicylate with the neutral polymer polyvinylpyrrolidone (PVP). The enthalpy of dilution of the surfactants has been measured in the absence and presence of the polymer and the results are compared to determine the effect of PVP on the micellization of the surfactants and the energetics of the process. As well, the micellization activity of the surfactants in the presence of the polymer has been studied by conductometric and fluorimetric methods. The dianthrimide method for the determination of boron in iron and low-alloy steels may be applied to nickel-base materials. The sample is dissolved, without any loss of boron, by hydrochloric and sulphuric acids and the resulting boric acid determined spectrophotometrically with dianthrimide.
