This study presents two simple and rapid methods for the quantification of therapeutic mAbs based on LC. Two mAbs (bevacizumab and infliximab) in plasma samples were purified using magnetic beads immobilized with a commercially-available idiotype antibody for each mAb. Purified mAbs were separated with HT-RPLC and detected with their native fluorescence. Using immunoaffinity beads, each mAb was selectively purified and detected as a single peak in the chromatogram. The HT-RPLC achieved good separation for the mAbs with sharp peaks within 20 min. The calibration curves of the two mAbs ranged from 1 to 20 μg mL−1 (bevacizumab) and 1–10 μg mL−1 (infliximab), and they had strong correlation coefficients (r2 > 0.998). The LOD of bevacizumab and infliximab was 0.07 and 0.15 μg mL−1, and the LLOQ of bevacizumab and infliximab was 0.12 and 0.25 μg mL−1, respectively. Thus, the sensitivities were sufficient for clinical analysis. Immunoaffinity purification with HT-RPLC produced a selective and accurate bioanalysis without an LC-MS/MS instrument. Both methods could become general-purpose analytical methods and complement the results obtained with conventional LBA.  相似文献

In this study, Co-Al2O3 nano-composite films containing parallel-oriented cobalt nano-cylinders with length-to-diameter aspect ratios of ca. 4000 are synthesized to acquire a uniaxial magnetization property. The Co nano-cylinders were electrodeposited into anodized aluminum oxide films with numerous nano-channels under a low over-potential of less than 0.1 V using an acidic aqueous solution containing CoCl2 at 75 °C. The long axis of Co nano-cylinders, which are electrodeposited at cathode potential of ?0.58 V vs. Ag/AgCl, coincides with the c axis of hexagonal close packed (hcp)-Co. The Co-Al2O3 nano-composite films show a uniaxial magnetization behavior due to the large shape anisotropy of Co nano-cylinders and the large magneto-crystalline anisotropy of the hcp-Co crystal structure. We achieved coercivity and squareness values of up to 2.4 and 1.0 kOe, respectively. This study shows an effective way to produce nano-composite films of altered coercivities.  相似文献

The first examples of air‐stable 20π‐electron 5,10,15,20‐tetraaryl‐5,15‐diaza‐5,15‐dihydroporphyrins, their 18π‐electron dications, and the 19π‐electron radical cation were prepared through metal‐templated annulation of nickel(II) bis(5‐arylamino‐3‐chloro‐8‐mesityldipyrrin) complexes followed by oxidation. The neutral 20π‐electron derivatives are antiaromatic and the cationic 18π‐electron derivatives are aromatic in terms of the magnetic criterion of aromaticity. The meso N atoms in these diazaporphyrinoids give rise to characteristic redox and optical properties for the compounds that are not typical of isoelectronic 5,10,15,20‐tetraarylporphyrins.  相似文献

Conducting polymers can be synthesized by irreversible diradical monomer polymerization. A reversible version of this reaction consisting of the formation/dissociation of σ‐dimers and σ‐polymers from a stable quinonoidal diradical precursor is described. The reaction reversibility is made by a quinonoidal molecule which changes its structure to an aromatic species by forming weak and long intermolecular C?C single bonds. The reaction provokes a giant chromic effect of about 2.5 eV. The two opposite but complementary quinonoidal and aromatic tautomers provide the Janus faces of the reactants and products which produces the observed chromic effect. A reaction mechanism is proposed to explain the variety of final products starting with structurally very similar reactants. These reversible reactions, covering an unusual regime of weak covalent supramolecular bonding, yield products which might be envisaged as novel molecular and polymeric soft matter phases.  相似文献

The fire and explosion of the Bituminization Demonstration Facility at PNC Tokai Works were occurred on March 11th, 1997. As soon as the fire occurred, environmental monitoring was started. The influence of accidental release was recognized from some samples of airborne dust and surface soil on site. No influence was recognized from the samples off site except the airborne dust collected at O-arai (20 km south). From Cladonia rangiferina were isolated two novel abietane diterpenoids, hanagokenols A (1) and B (2). Also in this investigation, four known abitetane diterpenoids (3-6), four known labdane diterpenoids (7-10), one known isopimarane diterpenoid (11), and six known aromatic compounds were isolated. These structures were elucidated primarily through extensive NMR experiments. Hanagokenol A (1) was a unique abietane diterpene having an ether linkage between C-6 and C-18 of sugiol. Hanagokenol B (2) is also a unique secoabietane diterpene, having gamma-lactone which occurred by cleavage and subsequently oxidation between C-6/C-7 of 12-hydroxydehydroabietinol. Furthermore, all the isolated compounds (1-17) were tested for the antimicrobial activity against methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococci (VRE).  相似文献
