In an attempt to generate nicotinic acetylcholine receptor (nAChR) ligands selective for the alpha4beta2 and alpha7 subtype receptors we designed and synthesized constrained versions of anabasine, a naturally occurring nAChR ligand. 2-(Pyridin-3-yl)-1-azabicyclo[2.2.2]octane, 2-(pyridin-3-yl)-1-azabicyclo[3.2.2]nonane, and several of their derivatives have been synthesized in both an enantioselective and a racemic manner utilizing the same basic synthetic approach. For the racemic synthesis, alkylation of N-(diphenylmethylene)-1-(pyridin-3-yl)methanamine with the appropriate bromoalkyltetrahydropyran gave intermediates which were readily elaborated into 2-(pyridin-3-yl)-1-azabicyclo[2.2.2]octane and 2-(pyridin-3-yl)-1-azabicyclo[3.2.2]nonane via a ring opening/aminocyclization sequence. An alternate synthesis of 2-(pyridin-3-yl)-1-azabicyclo[3.2.2]nonane via the alkylation of N-(1-(pyridin-3-ylethylidene)propan-2-amine has also been achieved. The enantioselective syntheses followed the same general scheme, but utilized imines derived from (+)- and (-)-2-hydroxy-3-pinanone. Chiral HPLC shows that the desired compounds were synthesized in >99.5% ee. X-ray crystallography was subsequently used to unambiguously characterize these stereochemically pure nAChR ligands. All compounds synthesized exhibited high affinity for the alpha4beta2 nAChR subtype ( K i < or = 0.5-15 nM), a subset bound with high affinity for the alpha7 receptor subtype ( K i < or = 110 nM), selectivity over the alpha3beta4 (ganglion) receptor subtype was seen within the 2-(pyridin-3-yl)-1-azabicyclo[2.2.2]octane series and for the muscle (alpha1betagammadelta) subtype in the 2-(pyridin-3-yl)-1-azabicyclo[3.2.2]nonane series. 相似文献

The surface morphology of hollow silica microspheres has influence on their applications. After a thorough investigation of the deposition of silica nanoparticles on polystyrene (PS) beads and the surface morphology and texture of the resultant hollow silica shells with scanning electron microscopy, transmission electron microscopy, and N2-sorption measurements, the influence of surfactant [cetyltrimethylammonium bromide (CTAB)] concentration on the surface morphology of hollow silica microspheres templated by PS beads is explained. Previously, CTAB was believed to turn the surface charge of PS beads from negative into positive so that negatively charged silica could be deposited on the PS template. Here, we show CTA+ cations preferentially assemble with silica species to form silica-CTA+ composite nanoparticles. Since the zeta potential of silica-CTA+ composite nanoparticles is smaller than that of pure silica nanoparticles, these composite nanoparticles encounter less repulsion when they are deposited on the surface of PS beads and close to each other. As more CTAB is added, the silica-CTA+ nanoparticles are less negatively charged, and more compact and smooth hollow silica microspheres are obtained. 相似文献

Ligand-stabilized platinum nanoparticles (Pt NPs) can be used to build well-defined three-dimensional (3-D) nanostructured electrodes for better control of the catalyst architecture in proton exchange membrane fuel cells (PEMFCs). Platinum NPs of 1.7 +/- 0.5 nm diameter stabilized by the water-soluble phosphine ligand, tris(4-phosphonatophenyl)phosphine (TPPTP, P(4-C6H4PO3H2)3), were prepared by ethylene glycol reduction of chloroplatinic acid and subsequent treatment of the isolated nanoparticles with TPPTP. The isolated TPPTP-stabilized Pt NPs were characterized by multinuclear magnetic resonance spectroscopy (31P and 195Pt NMR), high-resolution transmission electron microscopy (HRTEM), X-ray photoelectron spectroscopy (XPS), and extended X-ray absorption fine structure (EXAFS). The negatively charged TPPTP-Pt NPs were electrostatically deposited onto a glassy carbon electrode (GCE) modified with protonated 4-aminophenyl functional groups (APh). Multilayers were assembled via electrostatic layer-by-layer deposition with cationic poly(allylamine HCl) (PAH). These multilayer films are active for the key hydrogen fuel cell reactions, hydrogen oxidation (anode) and oxygen reduction (cathode). Using a rotating disk electrode configuration, fully mass-transport limited kinetics for hydrogen oxidation was obtained after 3 layers of TPPTP-Pt NPs with a total Pt loading of 4.2 microg/cm2. Complete reduction of oxygen by four electrons was achieved with 4 layers of TPPTP-Pt NPs and a total Pt loading of 5.6 microg/cm2. A maximum current density for oxygen reduction was reached with these films after 5 layers resulting in a mass-specific activity, i(m), of 0.11 A/mg(Pt) at 0.9 V. These films feature a high electrocatalytic activity and can be used to create systematic changes in the catalyst chemistry and architecture to provide insight for building better electrocatalysts. 相似文献

We report the extraordinary performance of carbon‐coated sodium super ion conductor (NASICON)‐type LiTi2(PO4)3 as an ideal host matrix for reversible insertion of both Li and Na ions. The NASICON‐type compound was prepared by means of a Pechini‐type polymerizable complex method and was subsequently carbon coated. Several characterization techniques such as XRD, thermogravimetric analysis (TGA), field‐emission (FE) SEM, TEM, and Raman analysis were used to study the physicochemical properties. Both guest species underwent a two‐phase insertion mechanism during the charge/discharge process that was clearly evidenced from galvanostatic and cyclic voltammetric studies. Unlike that of Li (≈1.5 moles of Li), Na insertion exhibits better reversibility (≈1.59 moles of Na) while experiencing a slightly higher capacity fade (≈8 % higher than Li) and polarization (780 mV) than Li. However, excellent rate capability profiles were noted for Na insertion relative to its counterpart Li. Overall, the Na insertion properties were found to be superior relative to Li insertion, which makes carbon‐coated NASICON‐type LiTi2(PO4)3 hosts attractive for the development of next‐generation batteries. 相似文献

Highly functionalized diaryl ethers were prepared by copper(II) acetate mediated O‐arylation reaction of phenols using trivalent organobismuthanes. The reaction is performed under simple conditions and tolerates a wide diversity of functional groups on the phenol and on the organobismuth reagent. Complexing polymer‐coated electrodes have been synthesized by oxidative electropolymerization of ethylenediamine tetra‐N‐(3‐pyrrole‐1‐yl)propylacetamide (monomer L ). The presence of four polymerizable pyrrole fragments on the same EDTA skeleton was thought to confer enhanced rigidity and controlled dimensionality to the resulting complexing materials, which were used for the electrochemical detection of Hg(II), Cu(II), Pb(II) and Cd(II) ions by means of the chemical preconcentration‐anodic stripping technique. The polyamide electrode material showed particularly a significant selectivity towards mercury ions, even in the presence of a large excess of other metal cations. Moreover, the use of imprinted polymer‐coated electrodes prepared by electropolymerization of L in the presence of metal cations turned out to significantly improve the detection limits, down to 5×10?10 mol L?1 for Hg(II) and Cu(II) species. 相似文献

The near‐infrared window of fluorescent heptamethine cyanine dyes greatly facilitates biological imaging because there is deep penetration of the light and negligible background fluorescence. However, dye instability, aggregation, and poor pharmacokinetics are current drawbacks that limit performance and the scope of possible applications. All these limitations are simultaneously overcome with a new molecular design strategy that produces a charge balanced and sterically shielded fluorochrome. The key design feature is a meso‐aryl group that simultaneously projects two shielding arms directly over each face of a linear heptamethine polyene. Cell and mouse imaging experiments compared a shielded heptamethine cyanine dye (and several peptide and antibody bioconjugates) to benchmark heptamethine dyes and found that the shielded systems possess an unsurpassed combination of photophysical, physiochemical, and biodistribution properties that greatly enhance bioimaging performance. 相似文献
