An improved method has been developed for the determination of the major neutral steroids (cholesterol and 5 beta-cholestan-3 beta-ol) and unconjugated bile acids (deoxycholic acid and lithocholic acid) in human faeces, using capillary gas chromatography with flame ionization detection. The freeze-dried faecal sample was subjected to a two-stage Soxhlet extraction followed by an aqueous alkali-organic solvent partition step to separate neutral steroids from bile acids. The neutral steroids were analysed as their trimethylsilyl ether derivatives on an OV-1 capillary column. The bile acids were further purified on a Sep-Pak C18 cartridge and then fractionated on a Sep-Pak SIL cartridge. Unconjugated bile acids were analysed as their methyl ester-trimethylsilyl ether derivatives also on an OV-1 capillary column. Quantitation of neutral steroids and unconjugated bile acids was achieved by reference to appropriate internal standards, added to the faecal extract immediately after the Soxhlet extraction stage. The method is being used in a study of the effect of diet on the metabolic activity of human gut flora.  相似文献

The use of oil-soluble radical generators for emulsion polymerization is considered. When radicals are formed in pairs within particles of a seed latex, the occurrence of radical desorption leads to complex kinetics. Use of radical balances and realistic simplifications produce a method for a relatively simple calculation of radical populations in the particles. Examples are restricted, for illustration purposes, to cases where the average number of radicals per particle, n , is less than one. Very low rates of radical desorption cause a substantial increase in n . As the desorption rate increases, the value of n decreases. n increases slowly with radical generation rate. Large decreases in the chain termination rate coefficient (which may accompany a gel effect) can have a negligible effect on the value of n . Radical distributions obtained from oil-soluble initiators can be slightly broader than those expected from the use of water-soluble initiators. The kinetics of emulsion polymerization will be similar for the two types of radical generator when radicals from oil-soluble generators can desorb from the polymer particles.  相似文献

Analysis of complex mixtures is a common challenge in natural products research. Quantitative nuclear magnetic resonance spectroscopy offers analysis of complex mixtures at early stages and with benefits that are orthogonal to more common methods of quantitation, including ultraviolet absorption spectroscopy and mass spectrometry. Several experiments were conducted to construct a methodology for use in analysis of extracts of fungal cultures. A broadly applicable method was sought for analysis of both pure and complex samples through use of an externally calibrated method. This method has the benefit of not contaminating valuable samples with the calibrant, and it passed scrutiny for line fitting and reproducibility. The method was implemented to measure the yield of griseofulvin and dechlorogriseofulvin from three fungal isolates. An isolate of Xylaria cubensis (coded MSX48662) was found to biosynthesize griseofulvin in the greatest yield, 149 ± 8 mg per fermentation, and was selected for further supply experiments. The batch emulsion polymerization of butyl acrylate in the presence of sodium lauryl sulphate as emulsifier and potassium persulphate as initiator was investigated. The effects of emulsifier concentration, initiator concentration, and monomer/water ratio on the kinetic features were studied. The kinetic data showed that at the conditions studied, the number of particles is proportional to [KPS]0.39 and [SLS]0.54. The number of particles did not practically vary with monomer concentration at the high range of monomer and emulsifier concentrations. At low emulsifier concentration, particle coagulation occurred in the course of reaction, which increased with monomer concentration. Particle nucleation was found to occur during Interval III of the batch process if undissociated micelles exist. It was also confirmed that the zero-one kinetics system can better fit the experimental results, compared to the pseudobulk kinetics. © 1999 John Wiley & Sons, Inc. Green chemistry can strongly attract students to chemistry. We, therefore, developed a green chemistry educational game that motivates students at the undergraduate and advanced high school levels to consider green chemistry and sustainability concerns as they design a hypothetical, chemical product. The game is intended for incorporation into any chemistry course for majors and non-majors that teaches sustainability and/or the Principles of Green Chemistry at the undergraduate level. The game is free of charge and encourages students to think like professional chemical designers and to develop a chemical product with respect to function and improved human and environmental health. This computer simulation has been assessed by educators and can be seamlessly integrated into an existing curriculum.  相似文献

Herein, we describe the isolation and characterization of a UnyLinker dimer which, if left uncontrolled, can become incorporated in oligonucleotide products. The dimer is formed as a result of an unusual intermolecular osmium-catalyzed etherification. We demonstrate that by simply replacing H2O2 as the co-oxidant with NMO, none of the UnyLinker dimer impurity is formed.  相似文献

Classification is concerned with the development of rules for the allocation of observations to groups, and is a fundamental problem in machine learning. Much of previous work on classification models investigates two-group discrimination. Multi-category classification is less-often considered due to the tendency of generalizations of two-group models to produce misclassification rates that are higher than desirable. Indeed, producing “good” two-group classification rules is a challenging task for some applications, and producing good multi-category rules is generally more difficult. Additionally, even when the “optimal” classification rule is known, inter-group misclassification rates may be higher than tolerable for a given classification model. We investigate properties of a mixed-integer programming based multi-category classification model that allows for the pre-specification of limits on inter-group misclassification rates. The mechanism by which the limits are satisfied is the use of a reserved judgment region, an artificial category into which observations are placed whose attributes do not sufficiently indicate membership to any particular group. The method is shown to be a consistent estimator of a classification rule with misclassification limits, and performance on simulated and real-world data is demonstrated.  相似文献
