The total synthesis of phosphatidylinositol mannosides (PIMs), a key class of antigenic glycolipids found on the cell wall of Mycobacterium tuberculosis, is described. The synthetic strategy relied on a [4 + 3] glycosylation of tetramannoside 1 and pseudotrisaccharide 2, which allowed for convergent access to the glycan backbone of the phosphatidylinositol dimannoside (PIM2) and hexamannoside (PIM6). A short practical synthesis of tuberculostearic acid was achieved based on a copper-catalyzed cross-coupling reaction. Union of the glycan and lipid parts resulted in the first total synthesis of native PIM2 and PIM6.  相似文献

QS-21 is one of the most promising new adjuvants for immune response potentiation and dose-sparing in vaccine therapy given its exceedingly high level of potency and its favorable toxicity profile. Melanoma, breast cancer, small cell lung cancer, prostate cancer, HIV-1, and malaria are among the numerous maladies targeted in more than 80 recent and ongoing vaccine therapy clinical trials involving QS-21 as a critical adjuvant component for immune response augmentation. QS-21 is a natural product immunostimulatory adjuvant, eliciting both T-cell- and antibody-mediated immune responses with microgram doses. Herein is reported the synthesis of QS-21A(api) in a highly modular strategy, applying novel glycosylation methodologies to a convergent construction of the potent saponin immunostimulant. The chemical synthesis of QS-21 offers unique opportunities to probe its mode of biological action through the preparation of otherwise unattainable nonnatural saponin analogues.  相似文献

Over the past 15 years, research in the field of microfluidics has experienced rapid growth due to significant potential advantages such as low cost, short analysis times, and elimination of sources of contamination. Although etched and thermally bonded glass substrates have seen widespread use and offer solid performance, device fabrication still remains cumbersome. Recent advances in sacrificial layer microfabrication methods for microfluidics have overcome many disadvantages of conventional fabrication approaches. Phase-changing sacrificial layers have been implemented in making inexpensive and high-performance polymer microchips for electrophoretic analysis, protein focusing, and sample preconcentration. In addition, novel channel fabrication methods based on standard thin-film processes, which are readily integratable with microfabrication techniques used for electrical components, are being applied increasingly for the creation of microfluidic devices. These new sacrificial layer fabrication approaches will be instrumental in making low-cost and high-quality polymer microchips, and in interfacing electrical and fluidic systems on glass or semiconductor substrates.  相似文献

The hindered internal rotor states (n(K) = 0(0), 1(1), and 1(0)) of the CN-Ar complex with two quanta of CN stretch (v(CN) = 2), along with its ground state (v(CN) = 0), have been characterized by IR-UV double resonance and UV spectroscopy. Analysis of rotationally structured bands enable n(K) assignments and reveal perturbations due to Coriolis coupling between two closely spaced hindered rotor states, n(K) = 1(1) and 1(0). A deperturbation analysis is carried out to derive accurate rotational constants and their associated CN center-of-mass to Ar bond lengths as well as the magnitude of the coupling. The energetic ordering and spacings of the CN-Ar hindered rotor states provide a direct experimental probe of the angular dependence of the CN X (2)Σ(+) + Ar potential and permit radially averaged anisotropy parameters (V(10) = 5.2 cm(-1) and V(20) = 3.2 cm(-1)) to be determined. This analysis indicates a relatively flat potential about a linear N≡C-Ar configuration with a barrier to CN internal rotation of only ~12 cm(-1). The angular potentials determined from experiment and ab initio theory are in good accord, although theory predicts a higher barrier to CN internal rotation. A similar approach yields the infrared spectrum of H(2)-CN in the CN overtone region, which exhibits a rotationally resolved Σ ← Σ parallel band that is consistent with theoretical predictions for ortho-H(2)-CN.  相似文献

Control of the crystallization of conjugated polymers is of critical importance to the performance of organic electronics, such as organic photovoltaic devices, due to the effect on charge separation and transport, particularly for all‐polymer devices. The block copolymer poly(3‐dodecylthiophene)‐block‐poly(9,9‐dioctylfluorene) (P3DDT‐b‐PF), which has matched crystallization temperatures for each block, is used to study the effects of processing history on resulting crystallization. For longer annealing times and rapid quenching to room temperature, P3DDT crystals are preferred whereas for shorter annealing times and slower quenching, PF crystals are preferred. Both crystal forms are evidenced for long annealing time and slow quenching. Additionally, for room temperature annealing in the presence of a chloroform vapor, PF crystals are found in the PF β phase with the predominant crystal peak oriented perpendicular to the thermally annealed case. These results will provide guidance for optimizing annealing strategies for future donor/acceptor block copolymer photovoltaic devices. © 2014 Wiley Periodicals, Inc. J. Polym. Sci., Part B: Polym. We study degree sequences for simplicial posets and polyhedral complexes, generalizing the well-studied graphical degree sequences. Here we extend the more common generalization of vertex-to-facet degree sequences by considering arbitrary face-to-flag degree sequences. In particular, these may be viewed as natural refinements of the flag f-vector of the poset. We investigate properties and relations of these generalized degree sequences, proving linear relations between flag degree sequences in terms of the composition of rank jumps of the flag. As a corollary, we recover an f-vector inequality on simplicial posets first shown by Stanley.  相似文献

The de novo synthesis of an aceric acid thioglycoside building block and the total synthesis of the plant carbohydrate aceric acid are described via a highly convergent strategy. Aldol reaction of acetaldehyde and a protected tartaric acid derivative provided the open chain carbohydrate. Subsequent acid treatment yielded the aceric acid thioglycoside in 35% total yield over five steps. Oxidative cleavage of the thioketal in the open chain carbohydrate and basic hydrolysis of the methyl ester furnished fully deprotected aceric acid in 31% yield over six steps.  相似文献

The synthesis of a core arabinomannan (AM) oligosaccharide from Mycobacterium tuberculosis has been achieved using a convergent [6 + 6] glycosylation strategy and a defined set of building blocks. Dodecasaccharide 1, containing the key AM structural features of lipoarabinomannan (LAM), was obtained in excellent yield and selectivity from hexamannan 3 and hexaarabinan 5. This flexible synthetic strategy involves late-stage couplings and modifications, thus providing ready access to several different LAM fragments. The incorporation of a thiol linker at the reducing end of the oligosaccharide allows for the attachment of these compounds to microarrays and protein carriers.  相似文献

The peak set of a permutation records the indices of its peaks. These sets have been studied in a variety of contexts, including recent work by Billey, Burdzy, and Sagan, which enumerated permutations with prescribed peak sets. In this article, we look at a natural analogue of the peak set of a permutation, instead recording the values of the peaks. We define the “pinnacle set” of a permutation w to be the set {w(i):i is a peak ofw}. Although peak sets and pinnacle sets mark the same phenomenon for a given permutation, the behaviors of these sets differ in notable ways as distributions over the symmetric group. In the work below, we characterize admissible pinnacle sets and study various enumerative questions related to these objects.  相似文献
