This work compares several different methods of site-specific analysis of glycoproteins using electrospray mass spectrometry. The glycoprotein, oLHalpha (ovine luteinizing hormone, alpha-subunit) was chosen as an appropriate example protein for these studies because of its biological relevance and extreme microheterogeneity. More than 20 unique glycoforms were detected for this glycoprotein at the Asn(56) site of oLHalpha. The carbohydrates present at this site affect receptor binding affinity, so understanding the great variety in the composition of these carbohydrates is important in studying ligand binding interactions. MS data was acquired on a quadrupole ion trap, a triple quadrupole, and a quadrupole time of flight mass spectrometer, and carbohydrate composition at the Asn(56) site of oLHalpha was determined using these instruments. Additionally, neutral loss and precursor ion scanning modes were also used to identify the glycoforms present, and these techniques were compared to the standard MS data. Of the three instruments compared in the study, the qTOF mass spectrometer achieved the lowest sample consumption, but all three instruments were useful in profiling the glycopeptide composition.  相似文献

Interest in nanocellulose-based coatings for packaging applications has been growing due to their excellent oil and gas barrier properties combined with their sustainable, recyclable, biodegradable, and non-toxic nature. Coating of nanocellulose materials such as microfibrillated cellulose (MFC) on paper/paperboard is challenging compared to traditional paper coating materials due to excessively high viscosity and yield stress of MFC suspensions at rather low solids content, typically below 5%. Possessing large amounts of water and a distinct rheological behavior such suspensions set tough demands on the substrate to be coated. It is important to understand and quantify substrate requirements in order to coat these suspensions successfully and achieve a satisfactory coating quality. A custom-built slot geometry is used herein to enable coating of highly viscous MFC suspensions on different paper-based substrates in a roll-to-roll process. The impact of substrate properties, such as surface chemistry and surface energy, surface roughness and surface porosity, and water absorption capacity on MFC coatability and coating quality is reported. Coating adhesion to the substrate was quantified with surface strength testing of MFC coated substrates. Various techniques, such as Scanning Electron Microscopy, IGT print penetration tests, and air permeability tests were employed for measuring coating coverage and surface porosity. MFC coating was found to adhere best to a highly hydrophilic surface, whereas the most uniform and defect-free film at low coat weights was formed on a smooth surface. It was also found that the MFC coat weight needed for full coverage, and therefore potentially good barrier, needs to exceed the surface roughness volume of the substrate. Water absorption capacity of the substrate also determines the final MFC coating quality obtained. The results clearly highlight the role of paper-based substrate for successful and effective coating of the micro and nanocellulose suspension.  相似文献

In telescopic homotopy theory, a space or spectrum is approximated by a tower of localizations , , taking account of -periodic homotopy groups for progressively higher . For each , we construct a telescopic Kuhn functor carrying a space to a spectrum with the same -periodic homotopy groups, and we construct a new functor left adjoint to . Using these functors, we show that the th stable monocular homotopy category (comprising the th fibers of stable telescopic towers) embeds as a retract of the th unstable monocular homotopy category in two ways: one giving infinite loop spaces and the other giving ``infinite -suspension spaces.' We deduce that Ravenel's stable telescope conjectures are equivalent to unstable telescope conjectures. In particular, we show that the failure of Ravenel's th stable telescope conjecture implies the existence of highly connected infinite loop spaces with trivial Johnson-Wilson -homology but nontrivial -periodic homotopy groups, showing a fundamental difference between the unstable chromatic and telescopic theories. As a stable chromatic application, we show that each spectrum is -equivalent to a suspension spectrum. As an unstable chromatic application, we determine the -localizations and -localizations of infinite loop spaces in terms of -localizations of spectra under suitable conditions. We also determine the -localizations and -localizations of arbitrary Postnikov -spaces.
