We report a selective LC–MS/MS method for the simultaneous quantitative determinations of the adenosine A2a receptor antagonist NIR178 (NIR178) and its major metabolite NJI765 in human plasma. Sample preparation steps involved protein precipitation, sample evaporation and reconstitution using a plasma sample volume of 0.1 ml plasma. Separation was achieved in 10 min on an Acquity UPLC BEH C18 1.7 μm, 2.1 × 50 mm column heated at 60°C with a gradient elution at 0.6 ml/min mobile phase made of water and acetonitrile both acidified with 0.1% formic acid. The detection was performed in positive ion mode and quantification based on multiple reaction monitoring. The linear response range was 1.00–1,000 ng/ml using a 1/x2 weighting factor. The intra- and inter-day accuracies (bias %) and intra- and inter-day precisions (CV, %) obtained for NIR178 and NJI765 were within the acceptance criteria. The normalized NIR178 and NJI765 matrix factor calculated from six lots of normal, lipemic and hemolyzed plasmas ranged from 0.97 to 1.05. The normalized recoveries of both NIR178 and NJI765 compared with their internal standards were consistent and reproducible with a CV ≤8.0. This method was successfully applied to support pharmacokinetic studies in adult patients with cancer.  相似文献

The shape of a coherent non-misfitting FeC precipitate formed from solute atoms that interact with a dislocation stress field was studied using kinetic Monte Carlo simulation on a rigid lattice. The system studied is a model for binary alloys with an interstitial alloying element similar to Fe–C. The interaction with the dislocation comprises both a short-range chemical term and a long-range deformation field. The effect of each of these terms on precipitate shapes for different C supersaturations was investigated. A simple analytical model is proposed to rationalize the results obtained in the atomistic simulations.  相似文献

This study investigated the gas separation membranes made with gelatin, crosslinked with ferulic acid (FA) and blended with polyethylene glycol (PEG) 200, by using a solvent‐free procedure. Gas permeation properties (He, N2, O2, and CO2) of these “green membranes” were studied and discussed in relation with their structure. Differential scanning calorimetric measurements were carried out to determine the gelatin triple helical renaturation level. The lowest permeability values [He and CO2 permeability (4.5 × 10?2 Barrer) with CO2/O2 selectivity of 14.5] were reached with gelatin/PEG 200 uncrosslinked membranes showing the highest renaturation level (40%). Crosslinking with FA lead to less rigid and brittle materials than GTA and to 10 times more permeable membranes compared with uncrosslinked membranes. Membranes crosslinked with glutaraldehyde broke during gas permeation measurements. We investigated the photobiomodulation effects of 1072 nm infrared light on the natural immune response involved in anti-bacterial and wound healing processes. Thirty mice infected with MRSA on the skin were divided into two groups. The experimental group was treated with 1072 nm infrared light (irradiance: 20 mW/cm(2), fluence: 12 J/cm(2) for 10 min) at 2, 4, 8, 12, 24 h, 3 and 5 days after inoculation and the control group with sham light. Serial changes of the mRNA levels of TLR2, IL-1β, TNF-α, IL-6, iNOS, MCP-1, TGF-β, bFGF and VEGF were studied by real time RT-PCR and those of the expression level of VEGF, bFGF, TGF-β and NF-κB by immunohistochemistry. The mRNA levels of the cytokines involved in the early phase of anti-bacterial immune response (IL-1β, TNF-α, IL-6, MCP-1) increased significantly in the 1072 nm group, peaking between 12 and 24 h post-inoculation. These levels normalized after 3-5 days. Immunohistochemistry revealed a notably stronger expression of VEGF in the 1072 nm group from 8-h post-inoculation to 5-day post-inoculation. We concluded that 1072 nm infrared light had a photobiomodulation effect which resulted in an enhanced biological immune response to the bacterial infection by MRSA and also increased the expression of VEGF to a significant level.  相似文献
