bPAK-interacting exchange factor may regulate actin cytoskeleton through interaction with actin

p21-activated kinase (PAK)-interacting exchange factor (PIX) is known to be involved in regulation of Cdc42/Rac GTPases and PAK activity. PIX binds to the proline-rich region of PAK, and regulates biological events through activation of Cdc42/Rac GTPase. To further investigate the role of PIX we produced monoclonal antibodies (Mab) against bPIX. Three clones; N-C6 against N-terminal half and C-A3 and C-B7 against C- terminal half of bPIX were generated and characterized. N-C6 Mab detected bPIX as a major band in most cell lines. C-A3 Mab recognizes GIT-binding domain (GBD), but it does not interfere with GIT binding to bPIX. Using C-A3 Mab possible bPIX interaction with actin in PC12 cells was examined. bPIX Mab (C-A3) specifically precipitated actin of the PC12 cell lysates whereas actin Mab failed to immunoprecipitate bPIX. Co-sedimentation of PC12 cell lysates with the polymerized F-actin resulted in the recovery of most of bPIX in the cell lysates. These results suggest that bPIX may not interact with soluble actin but with polymerized F-actin and revealed that bPIX constitutes a functional complex with actin. These data indicate real usefulness of the bPIX Mab in the study of bPIX role(s) in regulation of actin cyoskeleton.     

Application of Functionalized Ag Nanoparticles for the Determination of Proteins at Nanogram Levels Using the Resonance Light Scattering Method

A novel method for the determination of proteins in aqueous solutions has been developed based on the enhancement of resonance light scattering (RLS) of Ag nanoparticles in the presence of proteins. Factors including acidity of the media, concentration of Ag hydrosol, reaction time, temperature, and interference of non-protein substances were investigated. Under the optimal conditions, with the enhanced RLS signals at 452nm, the linear ranges of calibration curves were 0–0.8µgmL^–1 for bovine serum albumin (BSA), 0–1.2µgmL^–1 for human serum albumin (HSA), and 0–2.5µgmL^–1 for human -IgG (-IgG), respectively. The detection limits were 1.3ngmL^–1 for BSA, 10ngmL^–1 for HAS, and 5.7ngmL^–1 for -IgG.This method has been applied to the analysis of synthetic samples and real human serum samples, and the results were in good agreement with those reported by the hospital, indicating that the method presented here is not only sensitive and simple, but also reliable and suitable for practical applications.     

4,4′‐Bipyridyl N,N′‐dioxide–3‐hydroxy‐2‐naphthoic acid (1/2)

4,4′‐Bipyridyl N,N′‐dioxide crystallizes with 3‐hydr­oxy‐2‐naphthoic acid to give a centrosymmetric three‐component adduct, C₁₀H₈N₂O₂·2C₁₁H₈O₃, which is engineered into a two‐dimensional layer structure by two kinds of π–π inter­actions. Weak C—H⋯O inter­actions further link the two‐dimensional structure into a three‐dimensional structure.     

The design of an irradiator for the continuous processing of liquid latex

This paper presents anew design concept for a gamma irradiation plant for the continuous processing of pumpable liquids. Typical applications of such a plant include

• * the irradiation vulcanisation of natural latex rubber

• * disinfection of municipal sewage sludge for agricultural use

• * sterilisation of liquids in the pharmaceutical and cosmetics industries

• * industrial processing of bulk liquids

The authors describe the design and operation of the latex irradiator now operating on a small production scale in Malaysia and proposed developments.

The design allows irradiation processing to be carried out under an inert or other gaseous environment.

State-of-the-art computer control system ensures the fully automatic processing operation needed by industrial computers.

Stability of neutral and charged Si50 cage and stacked structure and in comparison with Si60 structures

We have performed calculations on the fullerene cage and stacked structures of neutral and ionic Si₅₀ cluster by full-potential linear-muffin-tin-orbital molecular-dynamics (FP-LMTO-MD) method. It is found that the neutral and charged fullerene cages relax into distorted structures resembling puckered balls. Their electronic structures are remarkably similar besides their geometrical structures. However, the relaxed cages are still not so stable as the stacked structure built from tricapped trigonal prism. By comparison, for the fullerene cages and stacked structures of neutral and ionic Si₆₀ cluster, similar characteristics are found.     

New Clerodane Diterpenoids from Casearia membranacea

Bioassay‐guided fractionation of an AcOEt extract of Casearia membranacea resulted in the isolation of six new clerodane diterpenes, caseamembrins G–L ( 1 – 6 ). The structures of the new compounds, including their relative configurations, were established by an extensive study of their spectral data, especially 2D NMR. The cytotoxic activities of the isolated diterpenes against human oral epidermoid (KB), cervical epitheloid (Hela), and liver (Hep59T/VGH) carcinoma cell lines were investigated.     

Synthesis, characterization, photoluminescence and electroluminescence properties of new 1,3,4-oxadiazole-containing rhenium（I） complex Re（CO）3（Bphen）（PTOP）

A new 1,3,4-oxadiazole-contanining rhenium(I) complex,with the formula [Re(CO)_3(Bphen)(PTOP)],(Bphen=bathophe- nardine,PTOP=4-(5-p-tolyl-1,3,4-oxadiazd-2-yl)pyridine),is synthesized and characterized by elemental analysis,IR,~1H NMR, UV-vis and luminescence spectroscopy.The double-layer electroluminescence devices based on the Re(I) complex have been fabricated by spin-coating technique.The turn-on voltage,maximum efficiency,and brightness for green emission obtained from the devices are 9V,2.1cd/A and 165cd/m~2,respectively.     

Morphology and degradation of biodegradable poly(L-lactide-co-β-malic acid)

Poly(L -lactide-co-β-malic acid) was obtained after the hydrogenolysis of the copolymer of L -lactide (L-LA) and β-benzyl malate (MA). ¹H-NMR demonstrated that the composition of the copolymer was identical to the feeding dose. Thermal property study revealed the morphology of the copolymer could be changed from semi-crystalline to amorphous with increasing hydrophilic β-malic acid content. The copolymers with 8 and 13 mol% β-malic acid content possessed enough mechanical property for fabricating cell scaffold. Degradation result demonstrated that most of the hydrophilic β-malic acid units broke down from the copolymer within 1 week and the ester bond between L -lactide and β-malic acid hydrolyzed prior to the inner ester bond of poly(L -lactide) (PLLA).
