A simple, selective, rapid, precise and accurate reverse phase high pressure liquid chromatographic method has been developed for the simultaneous estimation of diclofenac sodium and rabeprazole sodium from pharmaceutical formulations. The method was developed using a HiQ SiL C18 (250 mm × 4.6 mm i.d.) column with a mobile phase consisting of methanol:water, (80:20 v/v), at a flow rate of 1.25 mL min?1. Detection was carried out at 284 nm. Indapamide was used as an internal standard. The developed method was validated for linearity, accuracy, precision, limit of detection and limit of quantitation. The proposed method can be used for the estimation of these drugs in combined dosage forms.  相似文献

Characterization of heterogeneous proteins as large as 150,000 u was performed by a quadrupole mass spectrometer by using electrospray ionization (ESI). We were able to determine not only the molecular weight, but the detailed heterogeneity for the large glycoproteins as well. The successful application was facilitated by the optimization of the instrument in the high mass-to-charge range up to m/z 4000, where the multiply charged envelopes of the 150,000-u glycoproteins were found. For the analysis of clinically important monoclonal antibodies mass spectral data acquired by this method were consistent with the carbohydrate analysis and were useful in resolving the monosaccharide data into glycoform variations. In the case of the characterization of other large, heterogeneous proteins such as elongation factor 3 and bovine serum albumin, the quadrupole ESI mass spectrometer provided adequate mass resolution and high mass measurement accuracy to discern the modification and degradation of the proteins.  相似文献

Summary Oximidobenzotetronic acid (OBTA) is proposed as a sensitive spectrophotometric reagent for the estimation of 0.5–3.0 ppm of copper(II) at 427 nm in 50% dioxan at pH 5.3–7.5. For the estimation of 2 ppm Cu(II), 1.3 ppm Ni(II), 1.3 ppm Co(II), 3.2 ppm Fe(II), 10.3 ppm Fe(III), 9.7 ppm Ce(IV), 300 ppm acetate, 160 ppm oxalate, 95 ppm tartrate, 50 ppm citrate, as well as Zn(II), Cd(II), Hg(II)) Pb(II), Mn(II), As(III) as well as (V), Th(IV), Be(II), Ce(III), La(III), V(V) and Mo(VI), even when present in large quantities, do not interfere. The interference due to 25 ppm Bi(III), 20 ppm Sb(III), 20 ppm Sn(II), 25 ppm Sn(IV) and 30 ppm W(VI) can be removed by the addition of 95 ppm tartrate ions.
Zusammenfassung Oximidobenzotetronsäure wurde als empfindliches Reagens zur spektrophotometrischen Bestimmung von 0,5 bis 3,0 ppm Kupfer(II) bei 427 nm in 50%iger Dioxanlösung bei pH 5,3 bis 7,5 vorgeschlagen. Die Anwesenheit von 1,3 ppm Ni(II), 1,3 ppm Co(II), 3,2 ppm Fe(II), 10,3 ppm Fe(III), 9,7 ppm Ce(IV), 300 ppm Acetat, 160 ppm Oxalat, 95 ppm Tartrat, 50 ppm Citrat sowie die Anwesenheit auch großer Mengen Zn(II), Cd(II), Hg(II), Pb(II), Mn(II), As(III) bzw. (V), Th(IV), Be(II), Ce(III), La(III), V(V) und Mo(VI) stören die Bestimmung von 2 ppm Cu(II) nicht. Der störende Einfluß von 25 ppm Bi(III), 20 ppm Sb(III), 20 ppm Sn(II), 25 ppm Sn(IV) und 30 ppm W(VI) kann durch Zusatz von 95 ppm Tartrat beseitigt werden.

A selective complexometric method is described for the determination of palladium, sodium nitrite being used as masking reagent. Palladium(II) in a given sample solution is initially cornplexed with an excess of EDTA and the surplus EDTA is titrated with zinc sulfate solution at pH 4.5–5.5 (acetic acid-sodium acetate buffer), using xylenol orange as indicator. An excess of sodium nitrite is then added, the mixture is shaken well and the EDTA released from the Pd-EDTA complex is titrated with a standard zinc sulfate solution. Results are obtained for 2.5–27.5 mg of Pd with relative errors 0.5% and standard deviations 0.05 mg. The interferences of various ions are studied. The method is applied for the determination of palladium(II) in alloys and complexes.  相似文献

Two efficient protocols for the synthesis of tert-butyl (5S,6R,2E, 7E)-5-[(tert-butyldimethylsilyl)oxy]-6-methyl-8-phenyl-2, 7-octadienoate, a major component of the cryptophycins, are reported. The first utilized the Noyori reduction and Frater alkylation of methyl 5-benzyloxy-3-oxopentanoate to set two stereogenic centers, which became the C16 hydroxyl and C1' methyl of the cryptophycins. The second approach started from 3-p-methoxybenzyloxypropanal and a crotyl borane reagent derived from (-)-alpha-pinene to set both stereocenters in a single step and provided the dephenyl analogue, tert-butyl (5S,6R,2E)-5-[(tert-butyldimethylsilyl)oxy]-6-methyl-2, 7-octadienoate, in five steps. This compound was readily converted to the 8-phenyl compound via Heck coupling. The silanyloxy esters were efficiently deprotected and coupled to the C2-C10 amino acid fragment to provide desepoxyarenastatin A and its dephenyl analogue. The terminal olefin of the latter was further elaborated via Heck coupling. Epoxidation provided cryptophycin-24 (arenastatin A).  相似文献

A complexo-titrimetric method for the determination of copper(II) in the presence of other metal ions is described, based on the selective masking ability of 2-mercaptoethanol towards copper(II). Copper and other ions in a given sample solution are initially complexed with an excess of EDTA and the surplus EDTA is titrated with zinc sulfate solution at pH 5.0–6.0 (hexamine), using xylenol orange as indicator. A known excess of 2-mercaptoethanol solution (10%) is then added, the mixture is shaken well and the released EDTA from the Cu-EDTA complex is titrated against standard zinc sulfate solution. The interferences of various ions are studied and the method is applied to the determination of copper in its ores, alloys and complexes. Reproducible and accurate results are obtained for 2.5–40 mg of Cu with relative errors 0.4% and standard deviations 0.04 mg.  相似文献
