A double stranded DNA based fluorescence bioprobe for anticancer agent (doxorubicin) detection is described. This method provides a new way for sensitive DNA/drug interaction study by a homogeneous assay. The probe employs the long-wavelength intercalating fluorophore TOTO-3® (TT3). The anticancer agent, doxorubicin, which interacts with the DNA-TT3 complex, was indirectly measured by the decrease in the fluorescence intensity. Various oligonucleotides with different sequences were examined. Doxorubicin has preference for the oligonucleotide 5′AGCACG3′. Enhanced fluorescence observed for the TT3 intercalation with this oligonucleotide makes the DNA-dye complex a suitable bioprobe for doxorubicin detection by competitive assay. A home-built CCD camera setup was applied along with 384 well plate assay format for high throughput fluorescence imaging. The detection limit can be as low as 25 ng mL−1 with an upper limit of 100 μg mL−1. The recovery test with spiked serum sample shows that this method can be a potential routine method for therapeutic drug monitoring (TDM).  相似文献

Amphiphilic graft copolymers were prepared via the radical copolymerization of poly(ethylene oxide) (PEO) macromonomers with fluorocarbon or hydrocarbon acrylates in toluene with 2,2′‐azobisisobutyronitrile (AIBN) as an initiator. 1H NMR spectroscopy confirmed that the composition of the graft copolymers corresponded well to the monomer feed. For gel electrolytes prepared from the amphiphilic copolymers, the nature of the ionophobic parts of the amphiphilic graft copolymers had a great influence on the ion conductivity. Gel electrolytes based on graft copolymers containing fluorocarbon side chains showed significantly higher ion conductivity than electrolytes based on graft copolymers containing hydrocarbon groups. The ambient‐temperature ion conductivity was about 2.6 mS/cm at 20 °C for a gel electrolyte based on an amphiphilic graft copolymer consisting of an acrylate backbone carrying PEO and fluorocarbon side chains. Corresponding gels based on graft copolymers with PEO side chains and hydrocarbon groups showed an ambient‐temperature ion conductivity of about 1.2 mS/cm. The gel electrolytes contained 30 wt % copolymer and 70 wt % 1 M LiPF6 in an ethylene carbonate/γ‐butyrolactone (2/1 w/w) mixture. © 2001 John Wiley & Sons, Inc. An overview of the current trends within protein expression profiling is given where multidimensional separation of both gel- and liquid-phase techniques linked to mass spectrometry is viewed as a major route in the global proteome mapping. A clear trend in these biochemical developments is the effort to sequence and identify low-abundant protein expressions where assay miniaturization and integrated sample processing plays a central role. Two areas of miniaturization within the proteomics field are addressed: (i) sample cleanup and enrichment, and (ii) silicon microstructure developments for protein chip microarrays.  相似文献

We describe a Ti:sapphire-pumped picosecond optical parametric oscillator based on periodically poled RbTiOAsO(4) that is broadly tunable in the near to mid infrared. A 4.5-mm single-grating crystal at room temperature in combination with pump wavelength tuning provided access to a continuous-tuning range from 3.35 to 5microm , and a pump power threshold of 90 mW was measured. Average mid-infrared output powers in excess of 100 mW and total output powers of 400 mW in ~1-ps pulses were obtained at 33% extraction efficiency.  相似文献

Linear dichroism (LD) spectra are presented for naphthalene oriented in stretched polyethylene and polypropylene matrices at 77 K and 296 K. From the calculated spectrum LD(λ)/A(λ), where A(λ) is the corrected absorbance spectrum of the sample by unpolarized light, orientational parameters are calculated and component spectra, 235–315 nm, are resolved corresponding to polarization parallel to the long (B3u = x) and the short (B2u = y) axes in the molecular plane (D2h). The orientational parameters indicate different orientational mechanisms in polyethylene and polypropylene, but the resolving procedure yields mainly identical component spectra. It is suggested that the polarization (B3u) predominating in the 245–275 nm region isdue to a B1g vibronic perturbation of the 1B2u state.  相似文献

The equilibrium adsorption behavior of two n-alkyl-beta-D-glucosides (octyl (C8G1) and decyl (C10G1)) and four n-alkyl-beta-D-maltosides (octyl (C8G2), decyl (C10G2), dodecyl (C12G2), and tetradecyl (C14G2)) from aqueous solution on a titania surface, as measured by ellipsometry, has been investigated. The main focus has been on the effect of changes in the alkyl chain length and headgroup polymerization, but a comparison with their adsorption on the silica/water and air/water interfaces is also presented. Some comparison with the corresponding adsorption of ethylene oxide surfactants, in particular C10E6 and C12E6, is given as well. For all alkyl polyglucosides, the maximum adsorbed amount on titania is reached slightly below the critical micelle concentration (cmc), where it levels off to a plateau and the amount adsorbed corresponds roughly to a bilayer. However, there is no evidence that this is the actual conformation of the surfactant assemblies on the surface, but the surfactants could also be arranged in a micellar network. On hydrophilic silica, the adsorbed amount is a magnitude lower than on titania, corresponding roughly to a layer of surfactants lying flat on the surface. A change in the alkyl chain length does not result in any change in the plateau molar adsorbed amount at equilibrium; however, the isotherm slope for the alkyl maltosides increases with increasing chain length. Headgroup polymerization on the other hand affects the adsorbed amount. The alkyl glucosides start adsorbing at lower bulk concentrations than the maltosides and equilibrate at higher adsorbed amounts above the cmc. When compared with the ethylene oxide (EO) surfactants, it is confirmed that the EO surfactants hardly adsorb on titania, since the measured changes in the ellipsometric angles are within the noise level. The catalytic performance of porous silicon (PS) micro enzyme reactors (muIMER) is strongly dependent on the PS matrix morphology for enzyme immobilisation. PS was achieved in the muIMER by anodisation in a HF-ethanol mixture. PS etching of structured silicon surfaces commonly results in an inhomogeneous pore formation. The deep channel microreactors described herein have previously suffered from these phenomena, yielding non-optimised muIMERs. In order to obtain a homogeneous PS layer on the deep microreactor channel walls, different reactor geometries (channel wall thicknesses of 50 and 75 mum) were anodised at 10 and 50 mA cm(-2) for anodisation times ranging between 0 and 50 min. The muIMERs were evaluated by immobilising two types of enzymes, glucose oxidase (GOx) and trypsin, and the resulting catalytic turnover was monitored by a colorimetric assay. It was found that reactors with a homogeneous PS matrix displayed improved performance. The trypsin muIMERs were used to digest a protein, beta-casein, in an on-line format and the digest was analysed by MALDI-TOF MS. The importance of tailoring the muIMER geometry and the PS-matrix is crucial for the protein digestion. Successful protein identification after only 12 s. digestion was demonstrated for the best reactor, 75 mum channel wall, 25 mum channel width, anodised at 50 mA cm(-2) for 10 min.  相似文献

In this paper, a novel strategy for multicomponent analysis of two classes of pesticides such as triazines (atrazine and simazine) and phenoxyalkanoic acids (2,4-dichlorophenoxy acetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), 4-chlorophenoxyacetic acid (CPOAc), phenoxyacetic acid (POAc)) employing immuno-arrays is demonstrated. The approach is based on cross-reactive arrays of specific antibody pairs coupled to chemometric pattern recognition. The monoclonal antibody pairs employed in this work (atrazine-simazine and 2,4-D) are specific towards a set of analytes and preclude a particular set of others present in the sample matrix. Antibody pairs of atrazine, simazine, and 2,4-D are used to discriminate and quantify analyte of interest. Atrazine was quantified in presence of trace concentration of simazine and that of 2,4-D. The combinatorial cross-reactivity of antibody pairs towards simazine, atrazine and 2,4-D is used to distinguish among different classes of analytes and their influence on the signal suppression in immuno-techniques. These sensors exclude recognition by carbamates such as carbaryl and carbofuran.  相似文献

We report on the optical properties of single isolated silver nanodisks and pairs of disks fabricated by electron beam lithography. By systematically varying the disk size and surface separation and recording elastic scattering spectra in different polarization configurations, we found evidence for extremely strong interparticle interactions. The dipolar surface plasmon resonance for polarization parallel to the dimer axis exhibited a red shift as the interdimer separation was decreased; as expected from previous work, an extremely strong shift was observed. The scattering spectra of single particles and pairs separated by more than one particle radius can be well described by the coupled dipole approximation (CDA), where the particles are approximated as point dipoles using a modified dipole polarizability for oblate spheroids. For smaller particle separations (d < 20 nm), the simple dipole model severely underestimates the particle interaction, indicating the importance of multipolar fields and finite-size effects. The discrete dipole approximation (DDA), which is a finite-element method, describes the experimental results well even at d < 20 nm, including particles that have metallic bridges.  相似文献
