A fast method using high-performance liquid chromatography based on two monolithic columns has been developed for the simultaneous determination of isoflavones extracted from soybeans and derived foods. The 12 main isoflavones were resolved in 10 min in two coupled monolithic columns working at 35 °C using a elution gradient of acidified water (0.1% acetic acid) and methanol (0.1% acetic acid) at a flow rate of 5 mL min−1. Retention time and relative area standard deviations were below 1% for all isoflavones. The method developed was successfully applied to several soy food samples and spiked samples. Total isoflavone concentration in sampled soy foods ranged from 34.28 mg L−1 to 4.29 mg g−1.  相似文献

Stir bar sorptive extraction was evaluated for analysing volatiles in vinegar. The procedure developed shows detection and quantitation limits, and linear ranges adequate for analysing this type of compounds. The accuracy obtained was close to 100%, with repeatability values lower than 13%. The extraction efficiency is inversely affected by the acetic acid content. Although the absolute areas decrease, the compound area/internal standard area ratio remains constant, so for quantitative analysis, the acetic acid concentration does not affect the analytical data. The method was compared with a previous SPME method. Similar performance characteristics were obtained for both methodologies, with lower detection and quantitation limits and better repeatability reproducibility values for SBSE. Both analytical methods were used to analyse a variety of vinegars. The results obtained from both methods were in agreement.  相似文献

In this study, the classical semiempirical relationships between the electrophoretic mobility and the charge-to-mass ratio (me vs. q/Mα) were used to model the migration behaviour of peptides and glycopeptides originated from the digestion of recombinant human erythropoietin (rhEPO), a biologically and therapeutically relevant glycoprotein. The Stoke’s law (α = 1/3), the classical polymer model (α = 1/2) and the Offord’s surface law (α = 2/3) were evaluated to predict migration of peptides and glycopeptides, with and without sialic acids (SiA), in rhEPO digested with trypsin and trypsin–neuraminidase. The Stoke’s law resulted in better correlations for the set of peptides used to evaluate the models, while glycopeptides fitted better with the classical polymer model. Once predicted migration times with both models, it was easy to simulate their separation electropherogram. Results were later validated predicting migration and simulating separation of a different set of rhEPO glycopeptides and also human transferrin (Tf) peptides and glycopeptides. The excellent agreement between the experimental and the simulated electropherograms with rhEPO and Tf digests confirmed the potential applicability of this simple strategy to predict, in general, the peptide–glycopeptide electrophoretic map of any digested glycoprotein.  相似文献

Salvia officinalis L. oils were isolated from the plant's commercial dried aerial parts, by hydrodistillation, with different distillation times. The essential oils were analysed by gas chromatography and gas chromatography-mass spectrometry. The antioxidant ability was measured using a free radical scavenging activity assay using 2,2-diphenyl-1-picrylhydrazyl (DPPH), a thiobarbituric acid reactive substances (TBARS) assay, a deoxyribose assay for the scavenging of hydroxyl radical, an assay for site-specific actions and a 5-lipoxygenase assay. Antibacterial activity was determined by the agar diffusion method. 1,8-Cineole, α-pinene and camphor were the dominant components of all the essential oils. The different hydrodistillation times did not affect the oil yield nor the relative amount of the oil components. The time of hydrodistillation influenced the antioxidant activity. With the DPPH method, the oils isolated for 2 and 3 h were stronger free radical scavengers, while with the TBARS method, the highest antioxidant values were obtained in the oils isolated for 30 min, 2 and 3 h. Hydroxyl radical scavenging and lipoxygenase activity assays showed the best results with oils isolated for 1 and 3 h. With the deoxyribose method, sage oils at concentrations <1000 mg L(-1) showed better activity than mannitol. The essential oil of S. officinalis showed very weak antimicrobial activity.  相似文献

The use and abuse of illegal drugs affects all modern societies, and therefore the assessment of drug exposure is an important task that needs to be accomplished. For this reason, the reliable determination of these drugs and their metabolites in biological specimens is an issue of utmost relevance for both clinical and forensic toxicology laboratories in their fields of expertise, including in utero drug exposure, driving under the influence of drugs and drug use in workplace scenarios. Most of the confirmatory analyses for abused drugs in biological samples are performed by gas chromatographic–mass spectrometric methods, but use of the more recent and sensitive liquid chromatography–(tandem) mass spectrometry technology is increasing dramatically. This article reviews recently published articles that describe procedures for the detection of opiates in the most commonly used human biological matrices, blood and urine, and also in unconventional ones, e.g. oral fluid, hair, and meconium. Special attention will be paid to sample preparation and chromatographic analysis.  相似文献

Transient absorption spectroscopy on the μs-s time scale is used to monitor the yield and decay dynamics of photogenerated holes in nanocrystalline hematite photoanodes. In the absence of a positive applied bias, these holes are observed to undergo rapid electron-hole recombination. The application of a positive bias results in the generation of long-lived (3 ± 1 s lifetime) photoholes.  相似文献

Herein we describe the design and the assembly of temperature sensitive polysulfone (PS)/polyacrylonitrile (PAN) blend membranes using supercritical fluid technology. Blended membranes were prepared using the CO2‐assisted phase inversion method, and their pores were coated with two thermoresponsive hydrogels‐poly(N‐isopropylacrylamide) (PNIPAAm) and poly(N,N′‐diethylacrylamide) (PDEAAm). Permeation experiments of bovine serum albumin (BSA) and lysozyme (LYS) solutions were used to evaluate the performance and temperature‐responsive behavior of coated membranes. While membranes coated with PNIPAAm presented similar protein permeation profiles at temperatures below and above its lower critical solution temperature, PDEAAm coating imparted a temperature‐responsive behavior to PS/PAN (90:10) membranes and selective permeation of proteins with different sizes. The solid-fluid coexistence properties of the n - 6 Lennard-Jones system, n from 7 to 12, are reported. The procedure relies on determining Helmholtz free energy curves as a function of volume for each phase independently, from several NVT simulations, and then connecting it to points of known absolute free energy. For n = 12 this requires connecting the simulated points to states of very low densities on the liquid phase, and to a harmonic crystal for the solid phase, which involves many extra simulations for each temperature. For the reference points of the remaining systems, however, the free energy at a given density and temperature can be calculated relative to the n = 12 system. The method presented here involves a generalization of the multiple histogram method to combine simulations performed with different potentials, provided they visit overlapping regions of the phase space, and allows for a precise calculation of relative free energies. The densities, free energies, average potential energies, pressure, and chemical potential at coexistence are presented for up to T? = 5.0 and new estimations of the triple points are given for the n - 6 Lennard-Jones system.  相似文献

Lectins, proteins that recognize carbohydrates, have been immobilized on inert supports and used in the screening or purification of glycoproteins. Anacardium occidentale bark infusion has been used as a hypoglycemic agent in Brazil. The toxicity of natural products may be evaluated determining their capability to alter the biodistribution of technetium-99M (99mTc). This work reports the isolation and characterization of a lectin from A. occidentale bark (AnocBL), its evaluation as an affinity support for glycoprotein isolation and lectin effect on the uptake of 99mTc by rat adipocytes. AnocBL was isolated from 80?% ammonium sulphate supernatant by affinity chromatography on fetuin?Cagarose. SDS?CPAGE showed a single protein band of 47?kDa. The monossacharide l-arabinose and the glycoproteins fetuin, asialofetuin, ovomucoid, casein, thyroglobulin, peroxidase, fetal bovine serum and IgG inhibited the activity. The lectin activity was stable until 70?°C and at a pH range of 3.0?C7.5. AnocBL?CSepharose column bound fetuin indicating that the lectin matrix may be used to obtain glycoconjugates of biotechnological interest. In vitro assay revealed that glucose and insulin increase 99mTc uptake by rat adipocytes. AnocBL decreases 99mTc uptake, and this effect was not detected in the presence of glucose. Fetuin inhibited AnocBL effect in all insulin concentrations.  相似文献

Phosphate washing waste (PWW) is one of the wastes generated by the phosphate mine with a very high amount. This waste was investigated in this work to study the effect of the calcination of the PWW at four different temperatures 600 °C, 700 °C, 800 °C and 900 °C on its mineralogical and chemical composition. The samples were investigated by X-ray powder diffraction, Fourier transform infrared, differential scanning calorimetry and thermogravimetric analysis, solid-state magic angle spinning nuclear magnetic resonance of 29Si, 27Al and 31P and scanning electron microscope. The results show that the PWW presents a complex system and it suffers a significant change on its mineralogical and chemical composition after calcination. It reveals the presence of carbonate, natural zeolite, fluorapatite, quartz and clay. After calcination, the waste shows the disappearance of some of these phases and the appearance of others and some other phases remain steady.
