In this work, the time-dependent conjugation process between a thiolated molecule (with anti-parkinsonian properties) and gold nanoparticles has been monitored and studied by the combined use of fast acquisition Ultra Violet–Visible (UV–Vis) spectra and the ability of Multivariate Curve Resolution-Alternating Least Squares (MCR-ALS) technique. From the highly informative kinetic profiles obtained it was possible to extract quantitative and qualitative information of the conjugation process which includes i) time-dependent concentration profiles and pure spectra of species involved on conjugation process, ii) estimation of molecule concentration necessary for the completeness of the conjugation reaction, iii) molecule footprint and iv) free energy of molecule adsorption.  相似文献

In this work, the biogeochemistry of marine sediments from the Kiel Bight, coastal SW Baltic Sea, is studied based on the abundance and isotopic composition of organic carbon and different forms of sedimentary sulphur. Active bacterial sulphate reduction, partly under sulphate-limiting conditions, is evident from paired δ(34)S and δ(18)O values of pore water sulphate. The resulting pore water sulphide is partly precipitated as acid-volatile iron sulphide and subsequently forms sedimentary pyrite, partly serves in later diagenetic sulphurisation of organic matter, or remains dissolved in the pore water, all evident from the respective δ(34)S values. Microbial sulphate turnover is associated with an apparent isotopic fractionation between dissolved sulphate and dissolved sulphide (Δ(34)S) that varies between 46 and 66‰.  相似文献

N-l-Leucyldoxorubicin (Leu-Dox) was developed as a prodrug of doxorubicin (Dox) in order to diminish the cardiotoxic side-effect associated with repeated anthracycline treatment. To study the pharmacokinetics of Leu-Dox, Dox and other metabolites a sensitive and selective assay was needed. Leu-Dox and six of its known metabolites were extracted from plasma using an in-line reversed-phase precolumn (40-50 microns C8 particles). The trapped analytes were subsequently flushed to the analytical column (3 microns C18) using 0.5 ml of phosphate buffer (pH 3.5)-acetonitrile (2:1, v/v), which also served as the isocratic mobile phase. Within 12 min, a clean baseline-resolved chromatogram is obtained by fluorescence detection. Recoveries were almost quantitative and highly reproducible, with standard deviations less than or equal to 5.4% and less than or equal to 2.7% at spiked concentrations of 10 and 100 nM. Using 300 microliters of plasma, detection limits ranged from 0.3 to 0.8 nM at a signal-to-noise ratio of 3. The calibration curves were linear from 1 to 300 nM (r2 greater than or equal to 0.999) for each of the seven compounds. The between-day accuracy was in the range 91-99% and 99-105% at 10 and 100 nM, respectively, with standard deviations of 1-4%. Application of the assay to the analysis of plasma from patients after administration of Leu-Dox proved successful.  相似文献

Summary Given two subspaces A0 ⊂ A1 ⊂ W=X ⊕ Y, where X, Y are Banach spaces, we show how to characterize, in terms of generalized boundary conditions, those adjoint pairs A, A* satisfying A0 ⊂ A ⊂ A1, A 1 * ⊂ A∗ ⊂ A 0 * ⊂ W+=Y* ⊕ X*, where X*, Y* are the conjugate spaces of X, Y, respectively. The characterizations of selfadjoint (normal) subspace extensions of symmetric (formally normal) subspaces appear as special cases when Y=X*. These results are then applied to ordinary differential subspaces in W=Lq(ι) ⊕ Lr(ι), 1≦q, r≦∞, where τ is a real interval, and in W=C( ) ⊕ C( ), where is a compact interval. We test the performance of four‐component relativistic density functional theory by calculating the static and frequency‐dependent electric dipole–dipole polarizabilities of all (ground‐state) closed‐shell atoms up to Ra. We consider 12 nonrelativistic functionals, including three asymptotically shape‐corrected functionals, by using two smooth interpolation schemes introduced by the Baerends group: the gradient‐regulated asymptotic connection (GRAC) procedure and the statistical averaging of (model) orbital potentials (SAOP). Basis sets of doubly augmented triple‐zeta quality are used. The results are compared to experimental data or to accurate ab initio results. The reference static electric dipole polarizability of palladium has been obtained by finite‐field calculations using the coupled‐cluster singles, doubles, and perturbative triples method within this work. The best overall performance is obtained using hybrid functionals and their GRAC shape‐corrected versions. The performance of SAOP is among the best for nonhybrid functionals for Group 18 atoms but its precision degrades when considering the full set of atoms. In general, we find that conclusions based on results obtained for the rare‐gas atoms are not necessarily representative of the complete set of atoms. GRAC cannot be used with effective core potentials since the asymptotic correction is switched on in the core region.  相似文献

In order to calculate the residual stresses in a flat sheet of glass during annealing on the basis of its thermal history, the viscosity and coefficient of thermal expansion must be determined throughout the transformation range. The structural modifications of the material are taken into account by using fictive temperature concept. Three constitutive assumptions for the rate of the fictive temperature are discussed. A very good agreement is obtained between measured and calculated residual stresses.  相似文献

Oxidative stress originates from an elevated intracellular level of free oxygen radicals that cause lipid peroxidation, protein denaturation, DNA hydroxylation, and apoptosis, ultimately impairing cell viability. Antioxidants scavenge free radicals and reduce oxidative stress, which further helps to prevent cellular damage. Medicinal plants, fruits, and spices are the primary sources of antioxidants from time immemorial. In contrast to plants, microorganisms can be used as a source of antioxidants with the advantage of fast growth under controlled conditions. Further, microbe-based antioxidants are nontoxic, noncarcinogenic, and biodegradable as compared to synthetic antioxidants. The present review aims to summarize the current state of the research on the antioxidant activity of microorganisms including actinomycetes, bacteria, fungi, protozoa, microalgae, and yeast, which produce a variety of antioxidant compounds, i.e., carotenoids, polyphenols, vitamins, and sterol, etc. Special emphasis is given to the mechanisms and signaling pathways followed by antioxidants to scavenge Reactive Oxygen Species (ROS), especially for those antioxidant compounds that have been scarcely investigated so far.  相似文献

Four-component relativistic time-dependent density functional theory (TD-DFT) is used to study charge-transfer (CT) excitation energies of the uranyl molecule as well as the uranyl tetrachloride complex. Adiabatic excitation energies and vibrational frequencies of the excited states are calculated for the lower energy range of the spectrum. The results for TD-DFT with the CAM-B3LYP exchange-correlation functional for the [UO(2)Cl(4)](2-) system are in good agreement with the experimentally observed spectrum of this species and agree also rather well with other theoretical data. Use of the global hybrid B3LYP gives qualitatively correct results, while use of the BLYP functional yields results that are qualitatively wrong due to the too low CT states calculated with this functional. The applicability of the overlap diagnostic of Peach et al. (J. Chem. Phys.2008, 128, 044118) to identify such CT excitations is investigated for a wide range of vertical transitions using results obtained with three different approximate exchange-correlation functionals: BLYP, B3LYP, and CAM-B3LYP.  相似文献

The final disposal of residues generated at sewage treatment plants (STPs) has become a major problem for cities, due to the increase in the amount of treated sewage. One of the alternatives for the residue, labeled "sewage sludge," is its reuse in agriculture and in degraded soil. However, not all pathogens and metals present in it are eliminated during treatment. Diplopods have been used as bioindicators in ecotoxicological tests as they are constantly in close contact with the soil. Owing to this fact, the purpose of this study was to expose specimens of the diplopod Rhinocricus padbergi to substrate containing sewage sludge collected at STPs to analyze morphological alterations in their parietal and perivisceral fat body, where substances are stored. The exposures were held for 7, 15, or 90 days at different concentrations of sewage sludge (control, 1%, 10%, and 50%). The parietal fat body showed no alterations in any of the three exposure periods or concentrations. Alterations in the perivisceral fat body were observed for all exposure periods. According to the results, we suggest that the sludge used has toxic agents responsible for changing the animal's perivisceral fat body.  相似文献
