The asymmetric synthesis of fluorinated isoindolinones has been achieved by a palladium‐catalyzed aminocarbonylation reaction of the corresponding α‐fluoroalkyl o‐iodobenzylamines. A base‐mediated anti β‐hydride elimination process was suggested to explain the partial erosion of the optical purity observed in some cases. This mechanistic rationale enabled the minimization of this partial racemization by fine‐tuning the pKa of the base.  相似文献

The metastable phase diagram of the two-component system heptyloxycyanobiphenyl (7OCB)+nonyloxycyanobiphenyl (9OCB) was determined by means of modulated differential scanning calorimetry (MDSC) and optical microscopy measurements. It was experimentally established that the 7OCB+9OCB two-component system exhibits a monotropic re-entrant nematic behavior. A complete quantitative thermodynamic analysis, through Oonk's equal G analysis, was performed, allowing for the calculation of the monotropic re-entrant behavior and the prediction of two tricritical points, one of them experimentally accessible for the SmAd-to-N transition and the other non-experimentally accessible for the RN-to-SmAd transition. From specific-heat measurements, latent heats were obtained for those mixtures displaying a first-order SmAd-to-N transition. Additionally, for some mixtures, the specific-heat critical exponents (alpha), through the second-order SmAd-to-N transition, were obtained. Both batches of data enable us to access to the experimental tricritical temperature for the SmAd-to-N transition.  相似文献

A rapid and sensitive HPLC enantioselective method with fluorescence detection was developed to determine (-)-(R) and (+)-(S) enantiomers of the metabolites of citalopram, demethyl- and didemethyl-citalopram in plasma and brain tissue. This assay involves pre-column chiral derivatization with (-)-(R)-1-(1-naphthyl)ethyl isocyanate followed by separation on a normal-phase silica column. The developed liquid-liquid extraction procedure permits quantitative determination of analytes with recoveries ranged between 81 and 88% with intra- and inter-day relative standard deviations less than 10.5%. Linearity was obtained over the concentration range 5-1000 ng/mL and 100-10,000 ng/g for spiked drug-free plasma and brain tissue, respectively, with detection limits lower than 2.1 ng/mL and 42.8 ng/g.  相似文献

A sensitive and specific method for determining fenitrothion and its main metabolites, 3-methyl-4-nitrophenol and fenitrooxon, in poplar leaves using deuterated isotopes as the internal standard is described. The analytes and the labeled isotopes were extracted from leaves by solid-phase microextraction and subsequently analysed by gas chromatography coupled with mass spectrometry. The method had a chromatographic run time of 17.0 min and good linearity over the range 0.01-10 mg kg(-1). The detection limits ranged between 2.5 and 0.6 microg kg(-1). The isotopic dilution technique allowed improving significantly the repetitivity even using different fibers with the same coating (RSD<5.1%). The method was applied successfully to study the persistence of fenitrothion in forestal matrices in a poplar forest after cannon spray application of the insecticide.  相似文献

Substitution of the pillaring ligand in the homochiral open-framework [Ni(2)(L-asp)(2)(bipy)] by extended bipy-type ligands leads to a family of layer-structured, homochiral metal-organic frameworks. The 1D channel topology can be modified by the nature of the organic linker, with shape, cross-section and the chemical functionality tuneable. In addition, the volume of these channels can be increased by up to 36 % compared to the parent [Ni(2)(L-asp)(2)(bipy)]. The linker 1,4-dipyridylbenzene (3rbp) gives access to a new layered homochiral framework [Ni(2)(L-asp)(2)(3rbp)] with channels of a different shape. In specific cases, non-porous analogues with the linker also present as a guest can be activated to give porous materials after sublimation. Their CO(2) uptake shows an increase of up to 30 % with respect to the parent [Ni(2)(L-asp)(2)(bipy)] framework.  相似文献

Mechanical interactions between cells and their microenvironment play an important role in determining cell fate, which is particularly relevant in metastasis, a process where cells invade tissue matrices with different mechanical properties. In vitro, type I collagen hydrogels have been commonly used for modeling the microenvironment due to its ubiquity in the human body. In this work, the combined influence of the stiffness of these hydrogels and their ultrastructure on the migration patterns of HCT-116 and HT-29 spheroids are analyzed. For this, six different types of pure type I collagen hydrogels by changing the collagen concentration and the gelation temperature are prepared. The stiffness of each sample is measured and its ultrastructure is characterized. Cell migration studies are then performed by seeding the spheroids in three different spatial conditions. It is shown that changes in the aforementioned parameters lead to differences in the mechanical stiffness of the matrices as well as the ultrastructure. These differences, in turn, lead to distinct cell migration patterns of HCT-116 and HT-29 spheroids in either of the spatial conditions tested. Based on these results, it is concluded that the stiffness and the ultrastructural organization of the matrix can actively modulate cell migration behavior in colorectal cancer spheroids.  相似文献
