The lichen‐derived glycoconjugate gobienine A is structurally more complex than most glycolipids isolated from higher plants by virtue of the all‐cis substituted γ‐lactone substructure embedded into its macrocyclic frame. A concise entry into this very epimerization‐prone and hence challenging structural motif is presented, which relies on an enantioselective cyanohydrin formation, an intramolecular Blaise reaction, a palladium‐catalyzed alkoxycarbonylation, and a diastereoselective hydrogenation of the tetrasubstituted alkene in the resulting butenolide. This strategy, in combination with ring‐closing olefin metathesis for the formation of the macrocyclic perimeter, allowed the proposed structure of gobienine A ( 1 ) to be formed in high overall yield. The recorded spectral data show that the structure originally attributed to gobienine A is incorrect and that it is not the epimerization‐prone ester site on the butanolide ring that is the locus of misassignment; rather, the discrepancy must be more profound. 相似文献

Two small‐molecule–drug conjugates (SMDCs, 6 and 7 ) featuring lysosomally cleavable linkers (namely the Val–Ala and Phe–Lys peptide sequences) were synthesized by conjugation of the αvβ3‐integrin ligand cyclo[DKP–RGD]‐CH2NH2 ( 2 ) to the anticancer drug paclitaxel (PTX). A third cyclo[DKP–RGD]–PTX conjugate with a nonpeptide “uncleavable” linker ( 8 ) was also synthesized to be tested as a negative control. These three SMDCs were able to inhibit biotinylated vitronectin binding to the purified αVβ3‐integrin receptor at nanomolar concentrations and showed good stability at pH 7.4 and pH 5.5. Cleavage of the two peptide linkers was observed in the presence of lysosomal enzymes, whereas conjugate 8 , which possesses a nonpeptide “uncleavable” linker, remained intact under these conditions. The antiproliferative activities of the conjugates were evaluated against two isogenic cell lines expressing the integrin receptor at different levels: the acute lymphoblastic leukemia cell line CCRF‐CEM (αVβ3?) and its subclone CCRF‐CEM αVβ3 (αVβ3+). Fairly effective integrin targeting was displayed by the cyclo[DKP–RGD]–Val–Ala–PTX conjugate ( 6 ), which was found to differentially inhibit proliferation in antigen‐positive CCRF‐CEM αVβ3 versus antigen‐negative isogenic CCRF‐CEM cells. The total lack of activity displayed by the “uncleavable” cyclo[DKP–RGD]–PTX conjugate ( 8 ) clearly demonstrates the importance of the peptide linker for achieving the selective release of the cytotoxic payload. 相似文献

Dendritic copolymers comprising a hydrophobic core and hydrophilic shell with nearly equal numbers of hydroxyl groups in the shell and different densities in the core were prepared by a multi‐step process based on anionic ring‐opening polymerization. The diversity in the core density was obtained by using copolymer stars with poly(tert‐butyl‐glycidylether)‐block‐polyglycidol arms with nearly equal length of hydrophobic blocks and numbers of hydroxyl groups of polyglycidol but different numbers of arms as macroinitiators. The ability of the dendritic copolymers to serve as a nanocontainer for a ruthenium complex Ru(NH3)3Cl3 with anticancer properties was studied. The possibility of improving the water solubility of this poorly soluble drug by loading it onto dendritic copolymers was investigated. The hydroxyl groups of the dendritic copolymers were used for complexation of the ruthenium compound to the shell. The loading efficiency was analyzed by UV–vis spectroscopy. The nominal Global Aerosol Climatology Project (GACP) record of aerosol optical thickness (AOT) and Ångström exponent (AE) over the oceans is extended by 6 months to cover the period from August 1981 through December 2005. The most recent 4-year segment reveals no significant short-term tendencies in globally and hemispherically averaged AOTs and AEs. This finding is consistent with contemporaneous MODIS and MISR results and the accumulating evidence of a gradual transition from global brightening to global dimming. We also analyze the retrieval implications of allowing the imaginary part of the aerosols refractive index Im(m) to change over the duration of the GACP record. Our sensitivity study shows that increasing Im(m) from 0.003 during the 4-year pre-Pinatubo period up to 0.007 during the most recent 4-year segment of GACP data eliminates the previously identified long-term decreasing AOT trend. Should this long-term trend in Im(m) be real then it would cause the global absorption AOT over the oceans to more than double and the global single-scattering albedo to decrease from ~0.95 to ~0.88. Such changes could make tropospheric aerosols significant contributors to the recent surface temperature increase. 相似文献
