The construction of DNA‐encoded chemical libraries (DECLs) crucially relies on the availability of chemical reactions, which are DNA‐compatible and which exhibit high conversion rates for a large number of diverse substrates. In this work, we present our optimization and validation procedures for three copper and palladium‐catalyzed reactions (Suzuki cross‐coupling, Sonogashira cross‐coupling, and copper(I)‐catalyzed alkyne‐azide cycloaddition (CuAAC)), which have been successfully used by our group for the construction of large encoded libraries.  相似文献

The results of an experimental study of the He I 447.1 nm line and its forbidden component at high electron number density are presented and compared with profiles calculated using computer simulation method. Michelson interferometer at 632.8 nm was used to measure plasma electron number density in the range (1–7) × 1023 m− 3 while electron temperatures for the same experimental conditions in the range of 25 000 K to 35 000 K were determined using several spectroscopic techniques. The agreement of experimental overall line shape with computer simulation results is within 10% of what is well within theoretical and experimental uncertainty. This favorable comparison enabled the development of a simple approximate formula for the evaluation of electron number density from the measurement of wavelength separation between peaks of allowed and forbidden lines. This technique of plasma diagnostics is not sensitive to the presence of self-absorption of strong He I allowed line. The derivation of approximate formula with estimated accuracy of 15% was followed by detailed comparison with other experimental and theoretical data.  相似文献

The conventional four-step sequential extraction method and the EDTA and acetic acid single extraction procedures were applied to sewage sludge and sediment samples. The results obtained with these samples for Cu, Cr, Ni, Pb and Zn using the Tessier method were compared with those supplied by the two single extraction procedures employed. In addition, the Tessier method was also applied to a reference material, CRM 483, and these results were also compared with the certified EDTA and acetic acid values for this sample. As a result, good agreement was found between the metal contents released in the first three fractions of the Tessier method and those leached by the simpler single extraction procedures for the most of the elements studied. Subsequently, the conventional EDTA and acetic acid extraction methods were accelerated by means of microwave energy, in order to reduce the operating time. The extraction efficiency of the first three fractions of the Tessier method was compared with that obtained using the optimised microwave single extraction procedures and only in sewage sludge and CRM 483 samples were satisfactory results found for all the elements studied, except Cr and Pb. This means that the microwave single extraction procedures optimised in this work could be employed as screening methods to evaluate rapidly the easiest mobilizable heavy metals in these samples, although more samples should be analysed to determine their general applicability. The application of the accelerated single extraction procedures to a reference material, CRM 483, provided satisfactory results for all the elements studied, except for Cr in both methods and for Pb in the acetic acid extracts.  相似文献

Aniline/silica sol-gel material was obtained. The aniline was immobilized on the silica surface using chloropropyltrimethoxysilane as bridge reagent. The base activator NaH was used to produce a fast SN2 reaction between the base and the alkylorganosilane. The resulting modified silica was characterized by elemental analysis and infrared spectroscopy using an oven cell. The organic coverage on the surface was proportional to the organic precursor concentration. The aniline/silica materials are thermally stable up to 300°C, in high vacuum.  相似文献

We have measured the adsorption of methane and ethane to high pressure on SBA-2, a structured mesoporous silica composed of spheres connected by narrow channels. The experimental data were analyzed by carrying out Monte Carlo simulations of adsorption in pore structure models of different complexity and then adjusting the parameters of the models to match the Monte Carlo results to the experimental data. We found that a model based on single-sized spherical cavities was inadequate and that it is necessary to explicitly account for the interconnecting channels. Further, we found that despite the basic regularity of the SBA-2 structure, it is necessary to allow for a distribution of the sizes of both the cavities and the channels. These size distributions were obtained by fitting the parameters of the model to the experimental adsorption data, revealing detailed structural information not previously known for this material. The channels were found to be 5-15 A in diameter, while the cavities were 40-50 A in diameter. There is some evidence that the distribution of channel sizes leads to a percolation effect whereby the pore structure is not equally accessible to all adsorptives.  相似文献

For cleanup of animal fat before GC analysis of organochlorine pesticides and polychlorinated biphenyls, freezing-lipid filtration with solid-phase extraction and matrix solid-phase dispersion were evaluated to replace the official Brazilian methodology that uses preparative alumina column chromatography. General drawbacks associated with this last technique, such as the use of large amounts of solvent, laborious and time-consuming procedure could be avoided by using these alternative approaches. Experiments were carried out to study the performance by using different combinations of sorbents and elution solvents. Efficiency of alternative extraction methods in terms of fat removal and recovery capability was monitored by gravimetry, TLC, and GC with electron capture detection. Freezing-lipid filtration with solid-phase extraction afforded better clean up efficiency with recoveries in a range of 54.5 to 103.6% with the relative standard deviation of less than 10% for all compounds under investigation.  相似文献

Abstract

Synthesis of stereoisomeric α-methylene-γ-lactones in furanose-and furanuronoamide derivatives was easily accomplished by Reformatsky Reaction with ethyl bromcmethylacrylic ester and zinc. Pyridinium chlorochromate/3Å molecular sieve powder showed to be an excellent reagent for the oxidation of secondary hydroxyl groups of a furanose system and of α-hydroxy amides.  相似文献

The goal of this study is to explore the application of epigenetic markers in the identification of biofluids that are commonly found at the crime scene. A series of genetic loci were examined in order to define epigenetic markers that display differential methylation patterns between blood, saliva, semen, and epithelial tissue. Among the different loci tested, we have identified a panel of markers, C20orf117, ZC3H12D, BCAS4, and FGF7, that can be used in the determination of these four tissue types. Since methylation modifications occur at cytosine bases that are immediately followed by guanine bases (CpG sites), methylation levels were measured at CpG sites spanning each marker. Up to 11 samples of each tissue type were collected and subjected to bisulfite modification to convert unmethylated CpG-associated cytosine bases to thymine bases. The bisulfite modified DNA was then amplified via nested PCR using a primer set of which one primer was biotin labeled. Biotinylated PCR products were in turn analyzed and the methylation level at each CpG site was quantitated by pyrosequencing. The percent methylation values at each CpG site were determined and averaged for each tissue type. The results indicated significant methylation differences between the tissue types. The methylation patterns at the ZC3H12D and FGF7 loci differentiated sperm from blood, saliva, and epithelial cells. The C20orf117 locus differentiated blood from sperm, saliva, and epithelial cells and saliva was differentiated from blood, sperm, and epithelial cells at a fourth locus, BCAS4. The results of this study demonstrate the applicability of epigenetic markers as a novel tool for the determination of biofluids using bisulfite modification and pyrosequencing.  相似文献
