This work utilizes on-column ligand synthesis and affinity capillary electrophoresis (ACE) to determine binding constants (Kb) of 9-flourenylmethyloxy carbonyl (Fmoc)-amino acid derivatives to the glycopeptide antibiotics ristocetin (Rist) and teicoplanin (Teic). In this technique, two separate plugs of sample are injected on to the capillary column and electrophoresed. The initial sample plug contains a d-Ala-d-Ala terminus peptide and either one or two non-interacting standard(s). The second plug contains a Fmoc-amino acid-N-hydroxysuccinimide (NHS) ester. The electrophoresis is then carried out with an increasing concentration of Rist or Teic in the running buffer. Upon electrophoresis the initial d-Ala-d-Ala peptide reacts with the Fmoc-amino acid yielding a new Fmoc-amino acid-d-Ala-d-Ala peptide derivative. Continued electrophoresis results in the binding of Rist or Teic to the Fmoc-amino acid-d-Ala-d-Ala peptide derivatives. Analysis of the change in the relative migration time ratio (RMTR) or electrophoretic mobility () of the Fmoc-amino acid-d-Ala-d-Ala peptide derivatives relative to the non-interacting standards, as a function of the concentration of Rist and Teic, yields a value for
Kb. These findings demonstrate the advantage of coupling on-column ligand synthesis to ACE for estimating binding parameters between antibiotics and ligands.Abbreviations Rist

O4 has been studied. The galvanostatic discharge curves show that the best performance is obtained for y = 0.25, where a tetragonal to cubic structural transformation occurs. The thermodynamics and kinetics of the process of insertion
of the lithium into the tetragonal spinel Li
x
ZnNi0.25Mn1.75O4 (x = 0.05–1.3) have been studied. The molar thermodynamic quantities, such as enthalpy, entropy and free energy determined by
EMF-T measurements, varied with the lithium concentration in the oxide structure, and a major variation was observed around
x = 0.8. The chemical diffusion coefficient of lithium in these spinels was also determined. Structural analysis, degree of
oxidation and magnetic susceptibility measurements were carried out for the lithiated oxides in order to obtain the cationic
distribution as a function of x. It has been possible to demonstrate that, upon lithium insertion, Mn4+ ions on B sites are reduced to Mn3+ and then to Mn2+. A cooperative Jahn-Teller effect is present in these spinel manganese-nickel oxides.

Different chemical modifiers for use with electrothermal atomic absorption spectrometry (ET AAS) were investigated in relation
to determining the selenium in human urine samples. The samples were diluted in a solution containing 1% v/v HNO3 and 0.02% m/v cetyltrimethylammonium chloride (CTAC). Studying the modifiers showed that the use of either Ru or Ir as the
permanent modifier gave low sensitivity to Se and the peak shape was very noisy, while Zr or Rh gave no peak at all. The same
occurred when Zr was used in solution. For mixtures of permanent modifiers, Ir plus Rh or Zr plus Rh gave very low sensitivity,
Zr plus Rh with co-injection of Ir in solution was also not efficient, Zr plus Rh in solution gave good sensitivity, but the
best results were obtained with a mixture of Zr and Rh as the permanent modifier and co-injection of Rh in solution. Using
this last modifier, the following dilutions with the HNO3 and CTAC were studied: 1:1, 1:2, 1:3 and 1:4. The best dilution was 1:1, which promoted good sensitivity and a more defined
peak shape and made it possible to correct for the background using a deuterium arc lamp. Under these conditions, a characteristic
mass of 26±0.2 pg was obtained for Se in aqueous solution. Six certified urine samples were analyzed using matrix matching
calibration and the measured concentrations were in agreement with the certified values, according to a t-test at the 95%
confidence level. Recovery tests were carried out and the recoveries were in the range 100–103%, with relative standard deviation
better than 9%. The limit of detection (LOD, 3 sd, n=10) was 3.0 μg L−1 in the sample. The treated graphite tube could be used for at least 600 atomization cycles without significant alteration

The standard (p degrees = 0.1 MPa) molar enthalpies of formation of 2-, 3-, and 4-chloroaniline were derived from the standard molar energies of combustion, in oxygen, at T = 298.15 K, measured by rotating bomb combustion calorimetry. The Calvet high-temperature vacuum sublimation technique was used to measure the enthalpies of vaporization or sublimation of the three isomers. These two thermodynamic parameters yielded the standard molar enthalpies of formation of the three isomers of chloroaniline, in the gaseous phase, at T = 298.15 K, as 53.4 +/- 3.1 kJ.mol(-1) for 2-chloroaniline, 53.0 +/- 2.8 kJ.mol(-1) for 3-chloroaniline, and 59.7 +/- 2.3 kJ.mol(-1) for 4-chloroaniline. These values, which correct previously published data, were used to test the computational methodologies used. Therewith, gas-phase acidities, proton affinities, electron donor capacities, and N-H bond dissociation enthalpies were calculated and found to compare well with available experimental data for these parameters. 相似文献

A flow injection (FI) spectrophotometric method was proposed for the determination of chloride ion in natural waters. The determination of chloride was carried out by reaction with Hg(SCN)2 immobilized in an epoxy resin bead in a solid-phase reactor (SPR) and the thiocyanate ions released were determined spectrophotometrically at 480 nm after complexing reaction with Fe(III). The analytical curve for chloride was linear in the concentration range from 5.6 × 10−5 to 2.2 × 10−4 mol l−1 with a detection limit of 1.4 × 10−5 mol l−1. The relative standard deviation (R.S.D.) was 2.2% for a solution containing 2.2 × 10−4 mol l−1 (n = 10). The simple manifold allows a routine analytical frequency of 100 determinations per hour. The main advantage of the developed method is the 400% reduction of the Hg waste solution generated when compared to conventional methods for chloride determination based on the same spectrophotometric reaction. 相似文献

Ab initio accurate all-electron relativistic molecular orbital Dirac–Fock self-consistent field calculations are reported for the linear symmetric XeF2 molecule at various internuclear distances with our recently developed relativistic universal Gaussian basis set. The nonrelativistic limit Hartree–Fock calculations were also performed for XeF2 at various internuclear distances. The relativistic correction to the electronic energy of XeF2 was calculated as ~ ?215 hartrees (?5850 eV) by using the Dirac–Fock method. The dominant magnetic part of the Breit interaction correction to the nonrelativistic interelectron Coulomb repulsion was included in our calculations by both the Dirac–Fock–Breit self-consistent field and perturbation methods. The calculated Breit correction is ~6.5 hartrees (177 eV) for XeF2. The relativistic Dirac–Fock as well as the nonrelativistic HF wave functions predict XeF2 to be unbound, due to neglect of electron correlation effects. These effects were incorporated for XeF2 by using various ab initio post Hartree–Fock methods. The calculated dissociation energy obtained using the MP 2(full) method with our extensive basis set of 313 primitive Gaussians that included d and f polarization functions on Xe and F is 2.77 eV, whereas the experimental dissociation energy is 2.78 eV. The calculated correlation energy is ~ ?2 hartrees (?54 eV) at the predicted internuclear distance of 1.986 Å, which is in excellent agreement with the experimental Xe—F distance of 1.979 Å in XeF2. In summary, electron correlation effects must be included in accurate ab initio calculations since it has been shown here that their inclusion is crucial for obtaining theoretical dissociation energy (De) close to experimental value for XeF2. Furthermore, relativistic effects have been shown to make an extremely significant contribution to the total energy and orbital binding energies of XeF2. © 1995 John Wiley & Sons, Inc. 相似文献

The present paper proposes the application of multiwall carbon nanotubes (MWCNTs) as a solid sorbent for lead preconcentration using a flow system coupled to flame atomic absorption spectrometry. The method comprises the preconcentration of Pb (II) ions at a buffered solution (pH 4.7) onto 30 mg of MWCNTs previously oxidized with concentrated HNO3. The elution step is carried out with 1.0 mol L−1 HNO3. The effect of the experimental parameters, including sample pH, sampling flow rate, buffer and eluent concentrations were investigated by means of a 24 full factorial design, while for the final optimization a Doehlert design was employed. Under the best experimental conditions the preconcentration system provided detection and quantification limits of 2.6 and 8.6 μg L−1, respectively. A wide linear range varying from 8.6 up to 775 μg L−1 (r > 0.999) and the respective precision (relative standard deviation) of 7.7 and 1.4% for the 15 and 200 μg L−1 levels were obtained. The characteristics obtained for the performance of the flow preconcentration system were a preconcentration factor of 44.2, preconcentration efficiency of 11 min−1, consumptive index of 0.45 mL and sampling frequency estimated as 14 h−1. Preconcentration studies of Pb (II) ions in the presence of the majority foreign ions tested did not show interference, attesting the good performance of MWCNTs. The accuracy of the method was assessed from analysis of water samples (tap, mineral, physiological serum and synthetic seawater) and common medicinal herbs submitted to the acid decomposition (garlic and Ginkgo Biloba). The satisfactory recovery values obtained without using analyte addition method confirms the feasibility of this method for Pb (II) ions determination in different type of samples. 相似文献

Sesquiterpene lactones (SL) have been reported with various biological effects. Among the described SL skeletons, hirsutinolide and glaucolide have not been extensively studied by mass spectrometry (MS), especially how to distinguish them in organic matrices. Thus, this paper reports (1) a strategy of their differentiation based on MS behavior during the ionization and (2) a proposal of the fragmentation pattern for both SL‐subtypes. ESI(+)‐HRMS data of four isolated SL (hirsutinolides 1 and 3 ; glaucolides 2 and 4 ) were recorded by direct and UPLC water‐sample combined injections. These analyses revealed that hirsutinolides and glaucolides formed [M+Na]+ ion during the operation of the direct MS injection, and ([M+Na]+ and [M+H‐H2O]+) and [M+H]+ ions were respectively observed for hirsutinolides and glaucolides during the operation of combined UPLC water and sample MS injection. Computational simulations showed that the complex hirsutinolide ( 1 )‐Na+ formed with a lower preparation energy compared with the complex glaucolide ( 2 )‐Na+. However, despite their different behavior during the ionization process, ESI(+)‐HRMS/MS analyses of 1 ‐ 4 gave similar fragmentation patterns at m/z 277, 259, 241, and 231 that can be used as diagnostic ions for both skeletons. Moreover, the differentiation strategy based on the nature of the complex SL‐adducts and their MS/MS fragmentation pattern were successfully applied for the chemical characterization of the extract from Vernonanthura tweedieana using UPLC‐ESI‐HRMS/MS. Among the characterized metabolites, SL with hirsutinolide and glaucolide skeletons showed the aforementioned diagnostic fragments and an ionization behavior that was similar to those observed during the water‐sample combined injection. 相似文献
